Background and study aims: Esophageal burns due to ingestion of corrosive substances are frequently seen in both children and adults. However, there is no standard method of treatment to prevent associated mortality and morbidity. Therefore, this study aimed to evaluate the effects of known antioxidants, namely N-acetyl cysteine and ethyl pyruvate, on esophageal damage due to sodium hydroxide-induced corrosive burns. Materials and methods: Thirty-five female rats were randomly assigned to five equal groups. Group 1 was the sham group, while Group 2 was the control group. Group 3 received N-acetyl cysteine, Group 4 received ethyl pyruvate, and Group 5 received both N-acetyl cysteine and ethyl pyruvate. Rats in the burn groups were gavage-fed with 0.2mL of 25% NaOH. All esophagi were extracted on day 4 for histopathological evaluation. Results: Total histopathological damage scores were evaluated at the end of the study. Groups 3 and 5 were significantly different from the control group in terms of total histopathological scores (p = 0.001), while no significant difference was seen with Group 4. Stenosis index results in groups 3 and 5 were similar to those seen with total histopathological scores (p = 0.004). Conclusion: N-acetyl cysteine, alone or in combination with ethyl pyruvate, may be useful in the treatment of esophageal damage associated with corrosive substances and in achieving histopathological improvement in an experimental setting. Purpose: To investigate the healing effect of resveratrol on ischemia, inflammation and oxidative injury in brain and heart tissue of rats in an experimentally induced carbonmonoxide (CO) intoxication model. Methods: Twenty-eight mature female Spraque Dawley rats were randomized into four groups of seven animals each. Groups I and II inhaled room air for 60 minutes. Groups III and IV inhaled a high concentration of a 5000 ppm CO gas mixture at 4 L/min until loss of consciousness. Blood CO levels were measured from 1 mL blood specimens collected from the tail vein. Groups I and III received 1 mL of normal saline while groups II and IV received 25 mg/kg resveratrol intraperitoneally. At the end of 48 hour, the rats were sacrified and later blood, brain and heart tissue samples were taken for histopathological and biochemical evaluation. Neuron damage scores, percentages of degenerate neurons and apoptosis rates in the cortex and hypothalamus were calculated in brain tissues. Myocyte degeneration, vascular congestion and myocyte apoptosis rates were calculated in heart tissues. Total antioxidant (TAS), total oxidant (TOS) and oxidative stress index (OSI) levels were also calculated from the tissue samples. Results: Histopathological evaluation revealed that resveratrol significantly reduced tissue damage in the brain when groups III and IV were compared (p<0.05). Resveratrol also reduced cardiac myocyte degeneration and myocyte apoptosis rate in heart tissue (p=0.004 and p=0.002). No positive effect was determined at analysis of antioxidant parameters. Conclusion: The results of this study investigated that resveratrol administration in experimental setting may be beneficial in healing injury secondary to CO intoxication in brain and heart tissue.

İncele - MakaleYalnız künye2020Scopus SJRQ2Effect of Turkish Propolis on miRNA Expression, Cell Cycle, and Apoptosis in Human Breast Cancer (MCF-7) Cells
Misir, Sema; Aliyazicioglu, Yüksel; Demir, Selim; Turan, Ibrahim; Hepokur, Ceylan
Enriched in flavonoid compounds, phenol acids, and terpene derivatives, propolis has been shown to regulate apoptosis signaling pathways and alter the expression of microRNAs (miRNAs). In the present study, it has been aimed to examine the effects of Turkish propolis on miRNA levels of breast cancer (MCF-7) cells, and its relationship with cell proliferation and apoptosis. Cytotoxic activity of ethanolic propolis extract (EEP) was evaluated using MTT assay. Mechanisms involved in the cytotoxic action of Turkish propolis in MCF-7 cells were investigated with regard to apoptosis and cell cycle using flow cytometry and western blot. Mitochondrial membrane potential (MMP) were evaluated by spectrofluorometric method. miRNA levels were detected by qRT-PCR method. EEP exhibited selective toxicity against MCF-7 cells compared to normal fibroblast cells. EEP increased the cell cycle arrest at the G1 phase. EEP elevated the apoptotic cell death through increasing pro-apoptotic protein levels (p21, Bax, p53, p53-Ser46, and p53-Ser15), decreasing MMP and altering the expression levels of specific tumor suppressors (miR-34, miR-15a, and miR-16-5p) and oncogenic (miR-21) miRNAs. These data support that Turkish propolis may be evaluated as a potential natural agent for new anticancer drugs in future.

Zhang, Jin; Ozturk, Samet; Singh, Rakesh K.; Kong, Fanbin
This study aimed to evaluate the effect of cellulose nanofiber (CNF)-based coating containing chitosan (CHI) and trans-cinnamaldehyde (TC) on microbial growth of foodborne pathogens and spoilage microorganisms on cantaloupe rind and fresh-cut pulp. Coated/uncoated cantaloupe rind and pulp samples were inoculated with Salmonella enterica, Escherichia coli O157:H7 and Listeria monocytogenes and subsequently subjected to storage for 20 d at 4 °C or 12 d at 22 °C. Results showed that coating with only CNF (0.5%, dry-base) was effective in the elimination of S. enterica and E. coli O157:H7. Adding CHI (1%, w/w) and TC (1%, w/w) into CNF-based coating enhanced film formation on the surface and increased the antimicrobial activity on rind sample, but the reduction effect was less significant on pulp samples. The CNF (0.5%, dry-base) +CHI (1%, w/w) +TC (1%, w/w) coating resulted in 2.23, 2.43 and 3.22 log10 CFU/cm3 reduction of S. enterica, E. coli O157:H7 and L. monocytogenes, respectively, on pulp samples after 20 d at 4 °C. The formulation also significantly inhibited the growth of natural spoilage microorganisms, and no growth was observed up to 18 d at 4 °C. Results indicated that CNF-based coating significantly improved microbial safety of cantaloupe rind and fresh-cut pulp during storage by forming a physical barrier reducing activities of oxygen-dependent microorganisms.

- MakaleAçık Erişim2020Scopus SJRQ3The impact of dietary tarragon (Artemisia dracunculus) on serum apelin, brain-derived neurotrophic factor, cardiac troponin concentrations and histopathology of liver tissue in laying hens housed at different stocking densities
Bayraktar, Bulent; Tekce, Emre; Kaya, Hacer; Karaalp, Musa; Turunc, Eylul
Due to its association with several other stress factors (poultry house gases, inadequate ventilation, heat, cold and poor hygiene), the high stocking density is a major stress factor that adversely affects the health and performance of poultry and the quality of the poultry products. Therefore, this experimental study was aimed at analysing the impact of different doses of dietary tarragon (Artemisia dracunculus) on the serum apelin, plasma brain-derived neurotrophic factor (p-BDNF), and cardiac troponin I (cTnI) concentrations, and the correlation between these indicators in laying hens housed at different stocking densities. The aim of this study is to investigate the effects of adding tarragon in different ratios to laying hen rations in the 2ndovulation period on the cTnI, apelin, and BDNF hormone concentrations and the liver histopathology. The experiment was carried out over a period of eight weeks, with 192 Lohman Brown commercial hybrids at 50 weeks of age. Eight groups (four replicates each), composed of laying hens of equal body weight, which were housed at stocking densities of 580 cm2/hen and 810 cm2/hen and received 0, 1, 5 and 10 mmol/kg of tarragon (Artemisia dracunculus) in the feed, were established. At the end of the trial, 96 of the housed egg-laying hens (3 birds in each subgroup, a total of 12 birds in each group) were randomly selected and blood samples were taken from the vena subcutanea ulnaris. The samples collected were analysed for the apelin, p-BDNF, and cTnI contents. The analysis results demonstrated that tarragon supplementation had no effect on the serum apelin, p-BDNF and cTnI concentrations (P > 0.05). The Sub-Groups ST1, ST1.2, and ST6 presented with severe hyperaemia of the sinusoidal, portal and acinar blood vessels, whilst the hyperaemia of these blood vessels was moderate in Sub-Group ST12. Apelin, BDNF, and cTnI can act as protective factors against negative consequences of stress (e.g., stocking density or heat stress).

- MakaleAçık Erişim2020Scopus SJRQ2The Costs of Disability in Turkey
İpek, Egemen
In this study, the additional economic cost of disability for households is calculated using a standard of living approach (SoL) for Turkey. Firstly, the Standard of Living (SoL) index is determined using principal components analysis through the Household Budget Survey data for the year 2014. The cost of disability is estimated using ordered logit method by dividing the index into three income groups such as low, middle and high. As a result of the analysis, it is estimated that households with disabilities need additional financial support of at least 9.1% of household income in order to reach the same standard of living as those without disabled individuals. In addition, two different levels of disability, which are working life disability and daily life disability, are considered in estimating the cost of disability. While the cost caused by individuals’ disabilities in working life is 14.6% of their household's income, the ratio is 9.1% for the individuals’ disabilities in daily life.

Kotiloğlu, Deniz; Acet, Tuba; Özcan, Kadriye
In Anatolia, the tubers of Dactylorhiza romana subsp. georgica orchid are consumed as food and frequently used in folk medicine to alleviate the symptoms of some disorders. However, these properties have not been scientifically studied until the present day. In this research, we aimed to identify phenolic profile of the orchid tubers and determine their several features and functions. Among these, there are the antioxidant and antimicrobial effects and the inhibitory effect against diabetes related enzymes (α-amylase and α-glucosidase). In this sense, total phenolic content (Folin–Cicalteu assay), total antioxidant capacity (Phosphomolybdate method), antioxidant (ABTS and DPPH assays) and enzyme inhibition effects of tuber extracts obtained with different solvents (chloroform, ethyl acetate, methanol, ethanol, or hexane) were determined by using spectrophotometric methods, and the phenolic profile was also analysed by HPLC. In addition, antimicrobial activity of the extracts against 3 g (+) and 3 g (−) bacteria and 2 forms of yeasts was determined by disc diffusion and micro dilution methods. Based on our findings, the total phenolic content and antioxidant properties of ethyl acetate extract were considerably higher than the others. The most abundant among the screened phenolic standards was found to be benzoic acid. The highest α-amylase and α-glucosidase enzyme inhibition was measured in methanol and chloroform extracts, respectively. The highest antibacterial (B. cereus) and anticandidal (C. tropicalis and C. albicans) activities were detected in the chloroform and ethyl acetate extracts, respectively. In short, it was clearly seen that the D. romana subsp. georgica tuber had a great potential for being used in food and drug industries as a source of bioactive compounds.

Objective: The COVID-19 pandemic is a new and highly detrimental event that has influenced the world over. Besides the obvious health risks, COVID-19 has also affected people psychologically. The high risk of contamination can increase the perceived COVID-19 threat and death anxiety in individuals. The aim of the present study was to investigate the mediating role of self-compassion in the relationship between perceived COVID-19 threat and death anxiety. Additionally, the study also investigated the role of gender, profession and income level in the perceived COVID-19 threat and death anxiety. Method: The research sample was composed of 562 individuals from the community. The data was gathered by using personal information form and the Perceived COVID-19 Threat Form, Self-Compassion Scale and Turkish Death Anxiety Scale. Results: The relationship between perceived COVID-19 threat and death anxiety was partially mediated by self-compassion. Moreover, people in the high income group had a lower level of perceived COVID-19 threat and death anxiety score compared to the low-income group. Perceived COVID-19 threat levels were lower among health care professionals compared to unemployed individuals. Female participants had a higher level of perceived COVID-19 threat and death anxiety score compared to male participants. Discussion: Self-compassion may be an essential variable to help cope with problems related to perceived COVID-19 threat and death anxiety. Improving our understanding of the psychological impacts of COVID-19 is necessary and essential.
