- MakaleAçık Erişim2021Investigation the Different Extraction Temperature Effect of Artvin Savsat Region Propolis PBS Extracts on Antioxidant Properties and Eryhrocyte Hemolysis Inhibition
Canbolat, Deniz; Turan, Ibrahim; Küpeli, Yunus Emre; Kilinç, Sedanur; Piliç, Sevim
Propolis, which has been used in traditional medicine for hundreds of years all over the world, is a resinous natural product with a complex content obtained by honey bees by mixing the buds of plants and pollen grains with enzymes they secrete. Propolis may have different contents due to the gathering region, plant source, bee variety and climate of the place where it is collected. Although there are many studies related to different regions, so far the propolis of the Savsat region in Artvin have not been studied yet. The aim of this study is to determine the antioxidant properties (determination of iron reducing power, 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, total phenolic and total flavonoid content) of propolis extracts of Artvin Savsat region prepared with phosphate buffer salt solution (PBS) prepared at different temperatures (45 and 60 degrees C) and to determine the erythrocyte hemolysis inhibition properties of the extracts. Total phenolic (11.91 +/- 0.5-24.14 +/- 1.38 mg gallic acid equivalent to g propolis) and total flavonoid (1.79 +/- 0.03-2.13 +/- 0.07 mg quercetin equivalent to g propolis) content of PBS extracts, iron power determination (12.80 +/- 0.36-27.15 +/- 0.12 mg ascorbic acid equivalent to g propolis), 2,2-diphenyl-1-picrilhydrazyl (DPPH) radical scavenging activity (%22.86 +/- 0.5-31.24 +/- 1.33) and erythrocyte hemolysis inhibition (%55.58 +/- 0.57-96.37 +/- 0.02) were determined using spectrophotometric methods. As a result of the experiments, it was determined that the extract with PBS obtained at a temperature of 60 degrees C was higher than the extract obtained at 45 degrees C, in terms of antioxidant properties and erythrocyte hemolysis inhibition ratio. Further studies are required to identify molecules that inhibit erythrocyte hemolysis in extracts and have antioxidant properties.

In this study, phenolic composition, and in vitro biological activities of ethyl acetate (EAE) and methanol (ME) extracts obtained from the aerial parts of endemic Tanacetum erzincanense were investigated. Total phenolic and flavonoid content of the extracts were determined by Folin-Ciocalteu and aluminum chloride colorimetric methods, respectively. Antioxidant capacity of the extracts was evaluated over radical scavenging (DPPH and ABTS) and metal ion reducing power (FRAP and CUPRAC) tests. Individual phenolic compounds in ME was analyzed by high-performance liquid chromatography coupled to electrospray ionization quadrupole time-of-flight mass spectrometry (LC-ESI-QTOF/MS). Cell inhibitory potential of the extracts was tested against colorectal adenocarcinoma (HT-29), breast adenocarcinoma (MCF-7), and hepatocarcinoma (HepG2) cells by 2,3-bis(2-methoxy-4-nitro-5-sulfophenyl)-2H-tetrazolium-5-carboxanilide (XTT) assay. The results showed that ME contains higher TPC (64.4 mg GAE/g) and TFC (62.2 mg QE/g) than those of EAE (41.5 mg GAE/g and 40.0 mg QE/g). LC-ESI-QTOF/MS analysis revealed that ME is rich in phenolic compounds, namely, chlorogenic acid, apigenin, quercetin, luteolin, and diosmetin. Antioxidant assay results indicated that ME possess stronger activity than EAE and a power that competes with synthetic antioxidants. XTT assay results demonstrated that although both extracts displayed a considerable cytotoxicity against the tested cancer cell lines in a time and dose-dependent manner, ME expressed its selective inhibitory action towards MCF-7 cells with an IC50 value of 20.4 μg/mL for 72 h. These results may serve as a basis for further in vivo studies to examine the potential applications of T. erzincanense in food and pharmaceutical industries.

In this study, changes in ascorbic acid (AA), phenolics, anthocyanins (ACNs) and antioxidant activity (AOA) were monitored during storage at − 25, 4, 10 and 20 °C to observe the stability of bioactive compound in strawberry juice concentrates (SJCs). Half-life periods (t1/2) for ACN and AA degradations were 7.9 and 50 days at 20 °C, respectively. On the contrary, much higher t1/2 values were found for the reduction in AOA and phenolics, i.e. 251 and 430 days at 20 °C, respectively. AOA and phenolics were more stable during storage than AA and ACNs. p-Coumaric acid was the major phenolic identified in SJC. p-coumaric acid (33.8%), catechin (23.1%), vanilic acid (18.6%) and epicatechin (16.1%), which constituted 91.6% of phenolics in SJC, followed by chlorogenic acid (4.9%) and caffeic acid (3.2%). Chlorogenic acid was the least stable phenolic in SJC stored at 20 °C. Strong correlations were found between AOA and ACNs (r = 0.99), phenolics (r = 0.94) and AA (r = 0.92) in SJC stored at 20 °C. Significant ACN and AA degradations occurred at storage temperature as low as 4 °C. Therefore, storage at − 25 ℃ was suggested for SJC to maximize the stabilities of especially ACNs and AA in SJC.

The cytotoxic effect of the extract on five cancers (human breast, colon, lung, prostate, and liver carcinoma) and human fibroblast cells was determined using the MTT assay. TPC and reducing power values of extract were found 173.2 +/- 3.97 mg gallic acid equivalents, and 629.5 +/- 3.49 mg trolox equivalents per g sample, respectively. R. luteum leaf extract exhibited selective cytotoxicity especially against colon (1.9 fold) and liver (2.2 fold) cancer cells compared to normal fibroblast cells. This is the first study to reveal the cytotoxic effect of R. luteum leaf extract. Further studies are now needed to identify the cytotoxic molecules in the extract and their mechanisms.

Objective: Propolis is an important bee product, rich in polyphenolic compounds. It has antitumoral, antioxidant, antimutagenic, and other useful activities. Biological activities of propolis are generally attributed to its substance of polyphenolic compounds. The aim of this study was to investigate the in vitro antioxidant properties and reduction amount of intracellular reactive oxygen species (ROS) in human normal foreskin fibroblast cells by Turkish propolis ethanolic extract (EEP). Method: Total phenolic and flavonoid contents, reducing power, radical scavenging capacity, and iron chelating activity of EEP were determined using spectrophotometric methods. Intracellular ROS levels were determined by spectrofluorometric analysis with CM-H2 DCFDA. Results: Our results indicated that antioxidant activity was correlated with the higher total phenolic and flavonoid contents of EEP. The generated ROS levels were significantly decreased by EEP compared to only t-BHP group. Conclusion: Ethanolic propolis extract may be evaluated as a potency antioxidant resource and/or a novel natural agent in food and drug industries.
