Phenolic compounds are versatile bioactive molecules, particularly in food applications. In this study, the antioxidant capacity of Shogaol, a member of the phenolic class, was evaluated using several methods. Additionally, its inhibitory effects on human carbonic anhydrase I and II (hCA I and II), acetylcholinesterase (AChE), butyrylcholinesterase (BChE), α-glycosidase, and α-amylase enzymes, as well as its antiproliferative effects on A-549 lung cancer, HT-29 colon cancer, MCF-7 and MDA-MB-453 breast cancer cell lines, and the 3T3-L1 mouse fibroblast normal cell line, were investigated. To determine the antioxidant capacity of Shogaol, DPPH and ABTS radical scavenging assays, as well as Cu²⁺, Fe³⁺, and FRAP reduction methods, were employed. Its inhibitory effects on hCA I and II, AChE, BChE, α-glycosidase, and α-amylase enzymes were examined. Furthermore, cell counting was performed using the TBE method, and cell viability was measured with the XTT assay to assess the antiproliferative effects of Shogaol on A-549, HT-29, MCF-7, MDA-MB-453, and 3T3-L1 cell lines. Shogaol demonstrated higher antioxidant capacity than BHA, BHT, and Trolox in the FRAP assay (60 µg/mL), higher than BHT in the DPPH scavenging assay, and higher than Trolox in the ABTS, Cu²⁺, and Fe³⁺ reduction assays. Shogaol also exhibited significant inhibitory effects against hCA I, hCA II, AChE, BChE, and α-glycosidase, with IC₅₀ values of 75.91, 47.04, 37.44, 24.11, and 35.72 nM, respectively. Moreover, Shogaol showed a more potent antiproliferative effect than the standard chemotherapeutic drug Carboplatin in A-549 lung cancer and HT-29 colon cancer cell lines, and greater efficacy than Docetaxel in the MCF-7 breast cancer cell line.

Propolis is a sticky substance produced by bees because of the reaction of beeswax, pollen, and bee enzymes. Particularly, their biological activity and chemical content attract attention. Thus, in this study, the total amount of phenolic and flavonoid substances, Fe3+-Fe2+, Cu2+ (cupric ions reducing activity [CUPRAC]), and Fe3+-TPTZ (ferric ions reducing antioxidant power [FRAP]) reducing, and DPPH• and ABTS•+ scavenging assays in vitro antioxidant properties of propolis samples obtained from four different provinces of Türkiye were determined. In addition, the chemical content of propolis samples was quantitatively determined by LC–MS/MS, and the antiglaucoma property was revealed by hCAII enzyme inhibition. Propolis samples from Ordu presented the highest amounts of total phenolic and flavonoid content (492.3 ± 5.8 and 96.1 ± 2.1, respectively) and also highest antioxidant activity (DPPH• and ABTS•+ IC50 [μg/mL]: 8.884 ± 0.84 and 4.589 ± 0.80, respectively; Fe+3, CUPRAC, and FRAP: 1.051 ± 0.012, 1.021 ± 0.008, and 0.957 ± 0.007 μg/mL, respectively). hCAII enzyme inhibition was highest in Muş propolis (IC50 [μg/mL]: 8.6) as determined. By LC–MS/MS, 53 different components were screened and 35 bioactive components were determined. According to the results, propolis was found to be a raw material because it contains high concentrations of acacetin, chrysin, caffeic acid, and quinic acid (123.824, 24.759, 47.779, and 16.32 mg analyte/g extract, respectively).

Çakmakçı, Songül; Gülçin, İlhami; Gündoğdu, Engin; Ertem Öztekin, Hatice; Taslimi, Parham

Oxidation is one of the most important factors limiting shelf life and is a major deterioration process affecting both the sensory and nutritional quality of food. The high oxidation stability of lipids, which can be improved by the addition of antioxidants, is important for health protection, food quality, and economic reasons. In recent years, research on plant-derived antioxidants for use in human health and food has steadily increased. The aim of this study was to compare the antioxidant effects of green tea powder (GTP) in butter with those of commercial antioxidants (BHA, BHT, α-tocopherol, and Trolox). In addition, the effects on colour, sensory, gross physicochemical properties, and β-carotene content were investigated in butter. After the separation of butter into five pieces, the first part was chosen as the control sample without GTP; the second part has 100 mg/kg of BHT added to it; and the third, fourth, and fifth parts had 1, 2, and 3% of GTP added in the samples. They were stored at 4 ± 1 °C. Analysis was performed at intervals of 15 days. According to the iron reduction, CUPRAC and FRAP methods were performed, and parallel results were observed. Using the radical elimination methods (ABTS, DPPH•, and DMPD•+), IC50 values were calculated for the samples. According to the IC50 values, the GTP-containing samples were good antioxidants. The total phenolic andf β-carotene contents increased as the GTP addition increased. The addition of GTP had an antioxidant capacity equal to or higher than that of the BHT-added sample. For the production of a sensory-pleasing, greenish-coloured, new functional butter, the 1% GTP addition showed the most positive results.

Yapıcı, İsmail; Altay, Ahmet; Öztürk Sarıkaya, Beyza; Korkmaz, Mustafa; Atila, Alptuğ; Gülçin, İlhami; Köksal, Ekrem

In this study, phenolic composition, and in vitro biological activities of ethyl acetate (EAE) and methanol (ME) extracts obtained from the aerial parts of endemic Tanacetum erzincanense were investigated. Total phenolic and flavonoid content of the extracts were determined by Folin-Ciocalteu and aluminum chloride colorimetric methods, respectively. Antioxidant capacity of the extracts was evaluated over radical scavenging (DPPH and ABTS) and metal ion reducing power (FRAP and CUPRAC) tests. Individual phenolic compounds in ME was analyzed by high-performance liquid chromatography coupled to electrospray ionization quadrupole time-of-flight mass spectrometry (LC-ESI-QTOF/MS). Cell inhibitory potential of the extracts was tested against colorectal adenocarcinoma (HT-29), breast adenocarcinoma (MCF-7), and hepatocarcinoma (HepG2) cells by 2,3-bis(2-methoxy-4-nitro-5-sulfophenyl)-2H-tetrazolium-5-carboxanilide (XTT) assay. The results showed that ME contains higher TPC (64.4 mg GAE/g) and TFC (62.2 mg QE/g) than those of EAE (41.5 mg GAE/g and 40.0 mg QE/g). LC-ESI-QTOF/MS analysis revealed that ME is rich in phenolic compounds, namely, chlorogenic acid, apigenin, quercetin, luteolin, and diosmetin. Antioxidant assay results indicated that ME possess stronger activity than EAE and a power that competes with synthetic antioxidants. XTT assay results demonstrated that although both extracts displayed a considerable cytotoxicity against the tested cancer cell lines in a time and dose-dependent manner, ME expressed its selective inhibitory action towards MCF-7 cells with an IC50 value of 20.4 μg/mL for 72 h. These results may serve as a basis for further in vivo studies to examine the potential applications of T. erzincanense in food and pharmaceutical industries.

Altay, Ahmet; Koktepe, Tubanur; Durmaz, Lokman; Topal, Fevzi; Gülçin, İlhami; Köksal, Ekrem

Cress (Lepidium sativum) is an annual herb from Brassicaceae family, and in some regions. It is known as peppergrass, garden cress, garden pepper-cress, pepperwort, or poor man’s pepper. Cress is an important medicinal plant in some countries including Turkey. This plant has major scientific and therapeutic significance. Peroxidase (POD, E.C.1.11.1.7) is an oxidor-eductase enzyme produced by a number of organisms. In this study, POD enzyme was purified from cress by ammonium sulphate precipitation, gel filtration, and CM-Sephadex ion-exchange chromatographies. K m and V max values were calculated from the Lineweaver-Burk graph for H 2 O 2 and guaiacol substrates and the substrate specificity of POD was investigated. The results showed that substrate specificity of H 2 O 2 is better than substrate specificity of guaiacol for this enzyme. The inhibition effects of three cations (Al 3+ , Cu 2+ , and Cr 3+ ) and one organic compound of cetyl trimethylammonium bromide were performed and their inhibition types were determined. Finally, optimum pH, optimum temperature, optimum ionic strength, and stable pH were determined.
