Bereketoglu, Ceyhun; Arga, Kazim Yalcin; Eraslan, Serpil; Mertoglu, Bulent
Bioaccumulative environmental estrogen, nonylphenol (NP; 4-nonylphenol), is widely used as a nonionic surfactant and can affect human health. Since genomes of Saccharomyces cerevisiae and higher eukaryotes share many structural and functional similarities, we investigated subcellular effects of NP on S. cerevisiae BY4742 cells by analyzing genome-wide transcriptional profiles. We examined effects of low (1 mg/l; <15% cell number reduction) and high (5 mg/l; >65% cell number reduction) inhibitory concentration exposures for 120 or 180 min. After 120 and 180 min of 1 mg/l NP exposure, 187 (63 downregulated, 124 upregulated) and 103 genes (56 downregulated, 47 upregulated), respectively, were differentially expressed. Similarly, 678 (168 repressed, 510 induced) and 688 genes (215 repressed, 473 induced) were differentially expressed in cells exposed to 5 mg/l NP for 120 and 180 min, respectively. Only 15 downregulated and 63 upregulated genes were common between low and high NP inhibitory concentration exposure for 120 min, whereas 16 downregulated and 31 upregulated genes were common after the 180-min exposure. Several processes/pathways were prominently affected by either low or high inhibitory concentration exposure, while certain processes were affected by both inhibitory concentrations, including ion transport, response to chemicals, transmembrane transport, cellular amino acids, and carbohydrate metabolism. While minimal expression changes were observed with low inhibitory concentration exposure, 5 mg/l NP treatment induced substantial expression changes in genes involved in oxidative phosphorylation, cell wall biogenesis, ribosomal biogenesis, and RNA processing, and encoding heat shock proteins and ubiquitin-conjugating enzymes. Collectively, these results provide considerable information on effects of NP at the molecular level.

Introduction: Nutmeg is the dried seed of an evergreen aromatic tree from the plant family Myristicaceae. It is widely used in traditional medicine. Although misuse is rare, it needs to be understood that this plant, widely used as a spice in various parts of the world, can be misused and can result in emergency presentation due to intoxication. Case Report: We describe a case of intoxication in a 22-year-old male student after he mixed nutmeg with fruit juice and drank it. Neurological symptoms and anticholinergic signs were present during presentation. Conclusion: Nutmeg intoxication must be one of the possibilities considered at differential diagnosis in patients presenting to the emergency department with findings of anticholinergic intoxication.

Gokcen, Taner; Al, Melike; Topal, Meryem; Gulcin, Ilhami; Ozturk, Turan; Goren, Ahmet C.
Carbonic anhydrase inhibitor; sulphonamides; p-hydroxybenzoic acid; 3,4,5-trihydroxybenzoic acid (gallic acid).Carbonic anhydrase inhibitors are both in clinical use as antiglaucoma, diuretics, antiepileptics and management of altitude sickness, and under investigation as anticancer, anticonvulsant and antiobesity agents. Sulphonamides have been known for decades as carbonic anhydrase inhibitors and are in clinical use. Sulphonamide derivatives of p-hydroxybenzoic acid and 3,4,5-trihydroxybenzoic acid (gallic acid) were synthesized and their inhibition values over hCA I and hCA II isozymes, purified from human erythrocyte cells by Sepharose-4B-L-tyrosine-sulphanilamide, were determined. Compounds synthesized showed efficient carbonic anhydrase inhibition activity at low nM levels.

Mn(II), Co(II), Ni(II), Cu(II), and Zn(II) transition metal complexes of 2-hydroxy-5-[(E)-(4-phenyl) diazenyl] benzaldehyde oxime and 2-hydroxy-5-[(E)-(4-nitrophenyl) diazenyl] benzaldehyde oxime ligands were synthesized and characterized through NMR, IR, ESI mass, and UV analysis. DNA binding abilities of the complexes were revealed using a UV-Vis spectrophotometer with the absorption titration and competitive binding techniques. Hydrolytic and oxidative DNA cleavage activities under different conditions were also proved. Topoisomerase I inhibition efficiencies and in vitro free radical scavenging activities of all complexes were examined. Finally, the selective cytotoxic potentials of all complexes were evaluated in human colon cancer, normal colon, and fibroblast cell lines using the water-soluble tetrazolium salt (WST-1) assay. The complexes had the ability to intercalate into stacked base pairs of DNA and topoisomerase I activity was reasonably inhibited in their presence in 0.4 mM concentrations. The abilities for scavenging of DPPH and hydroxyl radicals were found to be higher than those of known standard antioxidants (ascorbic acid, butylated hydroxyanisole, and mannitol). The results obtained from the cytotoxicity experiments are especially promising for further research, which must be carried out for the evaluation of the studied complexes as anticancer drugs.

ObjectivesWe aimed to test the left atrial (LA) mechanics and contraction synchrony by 2D strain imaging, in patients with Wolff-Parkinson-White (WPW) syndrome, before and after radiofrequency catheter ablation (RFCA). MethodsStudy population consisted of 25 patients with WPW scheduled for RFCA and 30 healthy controls. The peak LA strain at the end of the ventricular systole (LAs strain) and the LA strain with LA contraction (LAa Strain) were obtained. To assess LA dyssynchrony, septal versus lateral wall time-to-peak strain measurements were measured. ResultsThere was no difference between the patients with WPW and control subjects with regard to peak LAs and LAa strain. Patients with WPW demonstrated higher global time-to-peak LAs and LAa strain values compared with the control group. Peak LAs strain and LAa strain values, measured before and after the RF ablation of the accessory pathway, were comparable (34.33.92 vs. 34.6 +/- 3.2, P=0.816, 14.7 +/- 2.8 vs. 15.3 +/- 2.3, P=0.052, respectively). Global time-to-peak LAs and LAa strain measurements were significantly shorter after the RFCA compared with the values obtained before the RFCA. However, septo-lateral times to peak LA strain differences were found to be comparable in both WPW versus control and pre- versus postablation groups. ConclusionLA mechanical function assessed by 2D strain imaging was comparable between patients with WPW and control subjects. Patients with WPW had more prominent LA dyssynchrony during atrial pump phase as compared with the controls, a condition which could not improve after successful elimination of the accessory pathway by RFCA.

Mansiz Kaplan, Basak; Yagci, Ilker; Akdeniz Leblebicier, Merve
Objectives: To detect the causes of shoulder pain with shoulder ultrasonography in patients with carpal tunnel syndrome (CTS) and compare the findings of the patients with CTS to healthy controls. Patients and Methods: Patients with CTS and healthy controls were evaluated clinically, electrophysiologically. and ultrasonographically by three masked researchers. Electrophysiological assessment consisted median and ulnar nerve conduction studies (NCS). In ultrasonographic assessment, cross-sectional areas of the median nerve were measured at hamatum hook, psiform bone, radio-ulnar joint levels. Shoulder joint, axial and longitudinal biceps tendon, subscapularis tendon, supraspinatus tendon, infraspinatus tendon, rotator cuff, acromioclavicular joint were evaluated by a standardized protocol. Results: The study was completed with 30 patients with CTS and 30 healthy subjects. The presence of shoulder pain in the last week was 53.3% in the CTS group and 30% in the control group but there was no statistically significant difference between the groups (p=0.16; p>0.05). Subdeltoid bursitis was significantly more common in CTS group when compared to the control group (p=0.03) in ultrasonographic assessment, and there was no significant difference between the two groups for other parameters. Conclusion: Shoulder pain and subdeltoid bursitis were common in patients with CTS however, shoulder pain was not directly related to shoulder problems alone. Central sensitization may play a role for shoulder pain rather than shoulder problems.

Background. Foreign bodies in the gastrointestinal tract are important morbid and mortal clinical conditions. Particularly, emergency treatment is required for cutting and drilling bodies. The majority of ingested foreign bodies (80-90%) leave gastrointestinal tract without creating problems. In 10-20% of cases, intervention is absolutely required. Less than 1% of cases need surgery. In this paper, we present a schizophrenia patient who swallowed multiple lighters. Case. A 21-year-old male schizophrenic patient who uses psychotic drugs presented to the emergency department with the complaints of abdominal pain, severe vomiting, and inability to swallow for a week. His physical examination revealed epigastric tenderness. A plain radiograph of the abdomen revealed multiple tiny metallic densities. Gastroscopy was performed. The lighters were not allowing the passage, and some of them had penetrated the gastric mucosa, and bezoars were observed. One lighter was extracted with the help of the polypectomy snare. Other lighters as a bezoar were removed by surgery. Conclusion. Excessive vomiting of swallowed foreign bodies in the etiology of psychotic patients should be kept in mind. Endoscopic therapy can be performed in the early stages in these patients, but in the late stage surgery is inevitable.

- MakaleYalnız künye2016Increased expressions of ADAMTS-13 and apoptosis contribute to neuropathology during Toxoplasma gondii encephalitis in mice
Dincel, Gungor Cagdas; Atmaca, Hasan Tarik
Toxoplasma gondii (T. gondii) is a protozoan parasite with the potential of causing severe encephalitis among immunocompromised humans and animals. Our previous study showed that T. gondii induces high nitric oxide (NO) production, high glial activation (GFAP) and neurofilament expressions, leading to severe neurodegeneration in toxoplasma encephalitis (TE) in the central nervous system (CNS). The aim of this experimental study was to investigate ADAMTS-13 expression and apoptosis in CNS and to identify whether they have any correlation with toxoplasmosis neuropathology and neurodegeneration. Mice were infected with ME49 strain T. gondii and the levels of ADAMTS-13, caspase 3, caspase 8, caspase 9, TNFR1 and Bcl-xL expressions were examined in brain tissues by immunohistochemistry, during the development and establishment of chronic infections at 10, 30 and 60days post-infection. Results of the study revealed that the levels of ADAMTS-13 (P < 0.005), caspase 3 (P < 0.05), caspase 8 (P < 0.05), caspase 9 (P < 0.005) and TNFR1 (P < 0.05) expressions in the brain markedly increased while Bcl-xL expression decreased (P < 0.005). The most prominent finding from our study was that 10, 30 and 60days post-infection ADAMTS-13 increased significantly and this may play an important role in the regulation and protection of the blood-brain barrier integrity and CNS microenvironment in TE. These results also suggest that T. gondii-mediated apoptosis might play a pivotal role and a different type of role in the mechanism of neurodegeneration and neuropathology in the process of TE. Furthermore, expression of ADAMTS-13 might give an idea of the progress and is critical for diagnosis of this disease. To the best of the authors' knowledge, this is the first report on ADAMTS-13 expression in the CNS of T. gondii-infected mice.

Aim of Study: Propolis is a resinous bee product, rich of polyphenolic compounds and flavonoids. It is known that in different geographic zones its chemical composition varies due to the different plant sources. Many biological properties including antimicrobial, antioxidative, anti-inflammatory, antitumoral, antigenotoxic, antimutagenic, cytostatic activities have been ascribed to propolis. These biological effects are predominantly attributed to its content of polyphenols. In this study, we aimed to evaluate the radioprotective effect of ethanolic extract of Turkish propolis. (EETP) against γ-ray-induced DNA damage on fibroblast cells using comet assay for the first time. Materials and Methods: Fibroblast cells were pretreated 15 and 30 min with concentrations of 100, 200 and 300 μg/mL EETP then they were exposed to 3 Gy γ-rays. Amifostine (synthetic aminothiol compound) was used as a positive control. Results: The results showed a significant decrease in γ-ray-induced DNA damage on cells treated with EETP and amifostine when compared to only irradiated cells. (P < 001). Conclusion: It was concluded that EETP prevent γ-ray-induced DNA damage in fibroblast cells and might have radioprotective activity.

A series of hydroxy and phenolic compounds have been assayed for the inhibition of two physiologically relevant carbonic anhydrase (CA, EC 4.2.1.1) isozymes, the cytosolic human isozymes I and II. The investigated molecules showed inhibition constants in the range of 1.07-4003 and 0.09-31.5 μM at the hCA I and hCA II enzymes, respectively. In order to investigate the binding mechanisms of these inhibitors, in silico studies were also applied. Molecular docking scores of the studied compounds are compared using three different scoring algorithms, namely Glide/SP, Glide/XP and Glide/IFD. In addition, different ADME (absorption, distribution, metabolism and excretion) analysis was performed. All the examined compounds were found within the acceptable range of pharmacokinetic profiles.
- MakaleYalnız künye2016Influence of CrN Coating on Electrochemical Behavior of Plasma Nitrided Pure Titanium in Bio-simulated Environment
Çelik, Ilhan
Titanium and its alloys are widely used as materials for bio-medical applications, such as implants. However, ions of the alloy can release to the body region and spread into the blood circulation. In this study, plasma nitriding and CrN coating techniques are used in order to overcome the problem of ion release. The objective of this study was to investigate the effects of plasma nitrided pure titanium on the structural properties and corrosion behaviors before and after CrN coating in Ringer's solution at 37 °C. The structural properties were investigated using Scanning Electron Microscopy (SEM) and X-ray diffraction (XRD). A diffusion layer and a compound layer composed of δ-TiN and ε-Ti2N phases were observed on the surface of nitrided pure titanium. Corrosion tests were made for the determination of electrochemical properties with the help of Potentiostat/Galvanostat device. The results show that corrosion behaviors of untreated and treated samples have similar characteristic.

Purpose: To investigate the apoptosis-inducing capacity of dimethyl sulfoxide (DMSO) extracts of bee pollen and propolis in HL-60 Myeloid Cancer Cell Lines. Methods: DMSO extracts of pollen and propolis were incubated separately with HL-60 cells, and caspase-3 activity evaluated. In order to determine the cell cycle characteristics of HL-60 cells with and without extracts of pollen and propolis, the cells were analysed using flow cytometry. Results: The DMSO extract of propolis (0.5 mg/mL) increased apoptosis from undetectable levels to 60.1%, while maintaining cell viability. The DMSO extract of pollen (2 mg/ml) increased apoptosis from undetectable levels to 52.2% while decreasing cell viability by 62%. Caspase-3 activity in HL-60 cells incubated with DMSO extracts of pollen and propolis were 3.6-to 12-fold higher than in controls. Conclusion: Turkish pollen and propolis individually increase apoptosis and the activity of caspase-3 in HL-60 cells. This finding indicates that bee products may have beneficial effects in the treatment of cancer.

N-substituted maleimides were synthesized from maleic anhydride and primary amines. 1,4-Dibromo-dibenzo[e,h]bicyclo-[2,2,2]octane-2,3-dicarboximide derivatives (4a-f) were prepared by the [4+2] cycloaddition reaction of dibromoanthracenes with the N-substituted maleimide derivatives. The carbonic anhydrase (CA, EC 4.2.1.1) inhibitory effects of the new derivatives were assayed against the human (h) isozymes hCA I, II, IX, and XII. All tested bicyclo dicarboximide derivatives exhibited excellent inhibitory effects in the nanomolar range, with Ki values in the range of 117.73-232.87 nM against hCA I and of 69.74-111.51 nM against hCA II, whereas they were low micromolar inhibitors against hCA IX and XII. A series of 9,10-dibromo-N-aryl-9,10-dihydro-9,10-[3,4]epipyrroloanthracene-12,14-diones (4a-f) were synthesized from N-substituted maleimide derivatives and 9,10-dibromoanthracene. Compounds 4a-f were assayed against human carbonic anhydrases (hCA) IX and XII, which are the two tumor-associated isozymes, and hCA I and II, which represent the most common off-targets for the development of selective anticancer CA inhibitors.
- MakaleAçık Erişim2016Effect of piracetam and nimodipine on full-thickness skin burns in rabbits
Sari, Elif; Dincel, Gungor C.
The potential of several drugs for full-thickness skin burns has been investigated, but the treatment of such burns remains a challenge in plastic surgery. The present study was designed to determine the effect of systemic and topical administration of piracetam and nimodipine on full-thickness skin burn wound healing. A total of 36 New Zealand male rabbits were divided into six groups. Full-thickness skin burns were produced in all the groups, except the control group. Piracetam was administered systemically (piracetam-IV) and topically (piracetam-C) for 14 days, and nimodipine was administered systemically (nimodipine-IV) and topically (nimodipine-C) over the burn wounds for 14 days. The sham group underwent burn injury but was not administered any drug. After 21 days, gross examination and histopathological analysis were performed and the results were compared statistically. Nimodipine-C and nimodipine-IV had no effect on burn wound healing. However, both piracetam-IV and piracetam-C significantly enhanced the healing of the full-thickness skin burn wounds, although the latter was more effective, useful and practical in burn wound healing. The histopathological features of the wounds in the piracetam-C group were closer to those of the control group than those of the other groups. Piracetam-C rather than piracetam-IV may promote full-thickness burn wound healing in rabbits.
- MakaleYalnız künye2016Positive correlation between insulin resistance and iron overload-induced oxidative stress in patients with fanconi anemia (FA)- and non-FA-related bone marrow failure: The results of a multicenter study

This study investigated the relationship between DNA, protein, and lipid oxidations and insulin resistance in patients with Fanconi anemia (FA)- and non-FA-related bone marrow failure. Sixteen patients with FA, 7 non-FA-related aplastic anemia, and 10 controls were included in the study. Fasting blood glucose, simultaneous insulin, hepcidin, ferritin, 8-hydroxy deoxyguanosine (8-OHdG), protein carbonyls, malondialdehyde (MDA), and homeostatic model assessment-insulin resistance (HOMA-IR) were investigated in the patients and controls. Diepoxybutane test-positive (DEB+) patients were diagnosed with FA, whereas DEB-patients were diagnosed as non-FA. 8-OHdG levels in both FA and non-FA patients were significantly higher than those in the controls (P =.001 and P =.005, respectively). Serum ferritin levels were also higher in FA and non-FA patients than in the controls (P =.0001 and P =.005, respectively). Insulin resistance (IR) was significantly higher in FA patients than in non-FA patients and controls (P =.005 and P =.015, respectively). Significant differences were observed between 8-OHdG, ferritin, and MDA levels in patients with or without IR (P =.009, P =.001, and P =.013, respectively). Moderate and strong relations of 44% and 85% were determined between IR and ferritin levels in patients with FA or non-FA (P =.08 and P =.014, respectively). FA and non-FA patients exhibited a tendency to IR. IR was related to ferritin levels, and ferritin levels were also correlated with oxidative stress. These findings suggest that the increased rate of IR in patients with FA and non-FA may derive from increased oxidative stress, which may in turn be due to elevated serum ferritin levels.

Cynarin is a derivative of hydroxycinnamic acid and it has biologically active functional groups constituent of some plants and food. We elucidated the antioxidant activity of cynarin by using different in vitro condition bioanalytical antioxidant assays like DMPD•+, ABTS•+, DPPH• and H2O2 scavenging effects, the total antioxidant influence, reducing capabilities, Fe2+ chelating and anticholinergic activities. Cynarin demonstrated 87.72% inhibition of linoleic acid lipid peroxidation at 30 g/mL concentration. Conversely, some standard antioxidants like trolox, tocopherol, butylated hydroxytoluene (BHT), and butylated hydroxyanisole (BHA) exhibited inhibitions of 90.32, 75.26, 97.61, 87.30%, and opponent peroxidation of linoleic acid emulsion at the identical concentration, seriatim. Also, cynarin exhibited effective DMPD•+, ABTS•+, DPPH•, and H2O2 scavenging effects, reducing capabilities and Fe2+ chelating effects. On the contrary, IC50 and Ki parameters of cynarin for acetylcholinesterase enzyme inhibition were determined as 243.67 nM (r2: 0.9444) and 39.34 ± 13.88 nM, respectively. This study clearly showed that cynarin had marked antioxidant, anticholinergic, reducing ability, radical-scavenging, and metal-binding activities.

- MakaleAçık Erişim2016Increased expressions of eNOS and iNOS correlate with apoptosis of diabetic nephropathy in streptozotocin-induced type 1 diabetic rats; [Streptozotosin kaynaklı tip 1 diyabetik sıçanlarda meydana gelen nefropatilerde artan eNOS ve iNOS sunumlarının apoptozisle ilişkilendirilmesi]
Dinçel, Güngör Çağdaş; Yildirim, Serkan
The present study was designed to evaluate the effects of high level of nitric oxide (NO) production and oxidative stress (OS) on nephropathology and to identify whether NO and OS have any correlation with apoptosis seen in diabetic kidney, elucidating the underlying mechanism(s) involved in the development of nephropathology in streptozotocin (STZ)-induced diabetic rats. Expression levels of caspase 3, caspase 9, endothelial nitric oxide synthase (eNOS), inducible nitric oxide synthase (iNOS), 8-hydroxy-2′-deoxyguanosine (8-OHdG), for the detection of oxidative damage to DNA, were examined in diabetic kidney tissues. Results of the study revealed that the levels of 8-OHdG (P<0.005), eNOS (P<0.005), iNOS (P<0.005), caspase 3 (P<0.005) and caspase 9 (P<0.005) were remarkably higher in diabetic kidney tissues than in controls. In addition, STZ-treated animals showed significant loss of body weight and renal enlargement. It was suggested that apoptosis, OS and increased NO levels are involved in the pathogenesis of diabetic nephropathy. The results also strongly suggested that STZ-induced apoptosis through activation of the intrinsic pathway and that might be most likely related to increased NO levels. Moreover, high NO production was not only mediated by eNOS but also by iNOS. Increased NO production may contribute to hyperfiltration and microalbuminuria in early diabetic nephropathy. Furthermore, expression of 8-OHdG might give an idea of the progress and may be essential as it has a diagnostic significance for this disease. In conclusion, we believe that eNOS and iNOS overexpressions induce diabetic nephropathy by mediating apoptosis in STZ-induced rats.
- MakaleYalnız künye2016A semiautomatic segmentation approach to biometric measurement of the talus bone of sedentary women and ballerinas using CT images
Reis, Hatice Catal; Bayram, Bulent; Seker, Dursun Zafer
Background: Ballet produces much stress on bones in the feet of ballerinas. Monitoring and detecting talus bone deformation is important for their quality of health and profession. Objectives: To determine differences in the talus bone between ballerinas and sedentary women. Methods: We evaluated biometric differences in the talus bone of 5 ballerinas referenced to 5 similar sedentary women recruited into the present study. We acquired 20 multidetector computed tomographic images including right and left feet. Semiautomatic region-based image processing using 3D-Doctor (Able Software Corp) was used to create three-dimensional (3D) virtual models of the talus bones. Biometric measurements were made on the 3D models and statistical analysis conducted. Results: The mean talus bone length of ballerinas was 3.37 cm (SD 0.12; range 3.11 to 3.52). The talus bone length of sedentary women was 3.29 cm (SD 0.16; range 3.04 to 3.65), and tended to be shorter than that of the ballerinas (P = 0.08, t test). However, the test was insufficiently powered. The mean volume of the talus bones from the ballerinas was 24.8 cm3 (SD 0.83) and smaller than that from the sedentary women, 26.9 cm3 (SD 1.25) at P < 0.001 (t = 4.38, 18 degrees of freedom. Difference 2.1. Two-tailed 95% confidence interval for difference of means: 1.08 to 3.08). We found less variation in the feet of ballerinas than sedentary women. Conclusions: Volumetric measurements show that feet of ballerinas are smaller and retain similar shape and size than the irregular feet of sedentary women.
