- BildiriAçık Erişim2016Scopus SJRQ4Assessment of sessile oak (quercus petraea L.) leaf as bioindicator for exploration geochemistry
Vural, A.
The element contents of sessile oak (Quercus petraea L.) leaves which grow on hydrothermally altered area with high element contents were studied at Gümüshane and its near vicinity. The aim of this study is to determine threshold element contents of Q. petraea L. leaves growing on the area. For this purpose, oak leave samples collected were analyzed by inductively coupled plasma-mass spectrometry for concentrations of some elements which were the group of most common trace elements in exploration geochemistry studies. The assessment of the element contents of the oak leaf is based on the descriptive statistics and computations of threshold values of leaves depending on statistical parameters and average element contents of the plants growing on lands in normal background values for the elements. According to Q-Q (normal quantile-quantile) plots, the Kolmogorov-Smirnov tests etc., all the elements, do not pass normality tests. Because of that, threshold values of the elements in leaves of oak were computed by median+median absolute deviation (MAD) and median+2MAD used in raw values for the elements. The data revealed that concentration levels of the examined elements in the leaves in the study area were determined as above and/or exceeded the upper limit of the normal range than in leaf of oak (Q. petraea L.) which grows on lands with normal elements background. Therefore, it is concluded that leaves of Q. petraea L. can be used as bioindicator in exploration geochemistry, especially for base metal ore deposits.

Kaya, Ayberk; Alemdağ, Selçuk; Dağ, Serhat; Gürocak, Zülfü
The objective of this study was to examine the effects on the stability of slope debris of planned slope excavations around the vicinity of Gumushane Imam-Hatip High School in NE Turkey. To assess the geotechnical properties of slope debris along the determined cross-sections, four exploration boreholes with a total length of 100 m were drilled and undisturbed soil samples were collected. Slope debris is composed of two soil zones and its depth varies between 15 m and 21 m. It was determined from the experimental results that the soil class of the upper zone is CL (low plasticity clay) and CH (high plasticity clay) whereas the soil class of the lower zone is SC (clayey sand) and GC (clayey gravel). Peak cohesions of the upper and lower zones were 26.3 kPa and 10.2 kPa, and their peak internal friction angles were 16° and 24°, respectively. Limit equilibrium (LE) analyses for the slopes were evaluated using Slide v5.0 software for the pre-excavation, post-excavation, and supported cases. The results of LE analyses showed the existence of a landslide along the SW direction; the landslide would extend to cover the whole region if the excavation were to be performed without applying engineering measures. Finally, the precautions to be taken were determined and stability analyses for the supported case were evaluated. These analyses showed that the active lateral forces applied by the proposed retaining walls to the cut slope should be 8325 and 3103 kN, respectively.

Dinçel, Güngör Çağdaş; Yildirim, Serkan
The present study was designed to evaluate the effects of high level of nitric oxide (NO) production and oxidative stress (OS) on nephropathology and to identify whether NO and OS have any correlation with apoptosis seen in diabetic kidney, elucidating the underlying mechanism(s) involved in the development of nephropathology in streptozotocin (STZ)-induced diabetic rats. Expression levels of caspase 3, caspase 9, endothelial nitric oxide synthase (eNOS), inducible nitric oxide synthase (iNOS), 8-hydroxy-2′-deoxyguanosine (8-OHdG), for the detection of oxidative damage to DNA, were examined in diabetic kidney tissues. Results of the study revealed that the levels of 8-OHdG (P<0.005), eNOS (P<0.005), iNOS (P<0.005), caspase 3 (P<0.005) and caspase 9 (P<0.005) were remarkably higher in diabetic kidney tissues than in controls. In addition, STZ-treated animals showed significant loss of body weight and renal enlargement. It was suggested that apoptosis, OS and increased NO levels are involved in the pathogenesis of diabetic nephropathy. The results also strongly suggested that STZ-induced apoptosis through activation of the intrinsic pathway and that might be most likely related to increased NO levels. Moreover, high NO production was not only mediated by eNOS but also by iNOS. Increased NO production may contribute to hyperfiltration and microalbuminuria in early diabetic nephropathy. Furthermore, expression of 8-OHdG might give an idea of the progress and may be essential as it has a diagnostic significance for this disease. In conclusion, we believe that eNOS and iNOS overexpressions induce diabetic nephropathy by mediating apoptosis in STZ-induced rats.

Tekce, E.; Gül, M.
This study compared the effects on performance parameters, intestinal microflora and histology of adding Origanum syriacum (OS) essential oil and an antibiotic (Avilamycin) to the rations of broilers fed under heat stress conditions. The 42-day study used 500 day-old male broilers (Ross-308) divided into 10 groups of 50 animals each. Then, each of these groups was further divided into 5 groups of 10 each. Groups were either kept at 22°C (stress-free) or at 36°C (heat stressed). Birds were fed with 5 experimental diets: control (basic feed (BF) without feed additive), antibiotic (BF + 100 mg/kg Avilamycin), OS 100 (BF + 100 mg/kg OS), OS 300 (BF + 300 mg/kg OS, OS 600 (BF + 600 mg/kg OS). At the end of the study period, some performance parameters, including feed intake, feed-conversion ratio, body weight, average daily weight gain, ileum crypt depth, villus height and microflora, decreased in groups under heat stress without additives (P < 0.01). However, comparison with the control group and the antibiotic group showed that adding OS essential oil provided a benefit close to that of the antibiotic, yet still inadequate, even at the 600 mg/kg dose.

Şeker, I.T.; Ertop, M.H.; Hayta, M.
In this study the valorisation of a gilaburu (Viburnum opulus) fruit pomace, a waste product remaining after juice extraction, was investigated. The pomace was incorporated into cake formulation at the level of 0, 5, 10 and 15%, based on wheat flour weight. Results indicated that the batter viscosity increased and pH decreased by the increasing of the concentration of pomace level in cake formulation. Depending on incorporation level, reduction on the volume and height of the cake samples were observed. The hardness values increased whereas the chewiness and gumminess values of the cake samples decreased with an increasing of pomace incorporation level. Gilaburu pomace addition caused significant (P<0.05) changes on L*, a*, and b* values of crumb, whereas no significant differences have been determined on the L* and b* values of crust of the samples. In sensory analyses, 5% gilaburu pomace added cake sample had the highest overall acceptability score. The total phenolic content and radical-scavenging activity values of cake samples increased proportionally with the level of pomace incorporation. According to the study, evaluation of gilaburu pomace waste as ingredient in developing alternative functional and/or value-added products by the food industry could be suggested.

- MakaleAçık Erişim2016Scopus SJRQ3Role of oxidative stress in the pathophysiology of Toxoplasma gondii infection
Dincel, Gungor Cagdas; Atmaca, Hasan Tarik
Oxidative stress (OS) plays an essential role in the pathogenesis of common neurodegenerative diseases. We have previously shown that Toxoplasma gondii (T. gondii) induces high nitric oxide (NO) production, glial activation, and apoptosis that altogether cause severe neuropathology in toxoplasma encephalitis (TE). The objective of this study was to investigate the cytotoxic effect of OS and to identify a correlation between the causes of T. gondii induced neuropathology. Expression levels of glutathione reductase (GR), Cu/Zn superoxide dismutase (SOD1), neuron specific enolase (NSE), and 8-hydroxy-2'-deoxyguanosine (8-OHdG) were investigated. Results of the study revealed that the levels of GR (P <0.005) and NSE (P <0.001) expression in the brain tissue markedly increased while SOD1 activity decreased (P <0.001) in the infected group compared to the non-infected group. In addition, intense staining for 8-OHdG (P <0.05) was observed both in the nucleus and the cytoplasm of neurons and glial cells that underwent OS. These results were reasonable to suggest that T. gondii-mediated OS might play a pivotal role and a different type of role in the mechanism of neurodegeneration/neuropathology in the process of TE. The results also clearly indicated that increased levels of NO and apoptosis might contribute to OS-related pathogenesis of TE. As a result, OS and expression of NSE might give an idea of the disease progress and may have a critical diagnostic significance for patients with T. gondii infection.

Aygul, Imdat; Yaylaci Karahalil, Fatma; Supuran, Claudiu T.
Polyphenols are important secondary products of plants with the potential to inhibit carbonic anhydrases. The aim of this study was to investigate the inhibition effects of various phenolic standards, honey, propolis, and pollen species on human carbonic anhydrase I and II. The inhibition values (IC50) of the phenolics (gallic acid, protocatechuic acid, quercetin, catechin, tannic acid, and chrysin) ranged from 0.009 to 0.32 μg/mL, tannic acid emerging as the best inhibitor. The inhibition values of three different types of honey, heather, rhododendron, and chestnut ranged between 2.32 and 25.10 μg/mL, the chestnut honeys exhibiting the best inhibition. The ethanolic extracts of pollen and propolis exhibited good inhibitory properties, with IC50 values between 0.486 and 3.320 μg/mL. In order to evaluate the phenolic composition of bee products, phenolic profiles and total phenolic contents (TFC) were also measured. The inhibition ranking among the natural products studied was phenolic standards > propolis > pollen > honeys, and inhibition was related to TFC.

Ünker, F.; Çuvalci, O.
This paper deals with the investigation of optimum values of the stiffness and damping which connect two gyroscopic systems formed by two rotors mounted in gimbal assuming negligible masses for the spring, damper, and gimbal support. These coupled gyroscopes use two gyroscopic flywheels, spinning in opposing directions to have reverse precessions to eliminate the forces due to the torque existing in the torsional spring and the damper between gyroscopes. The system is mounted on a vertical cantilever with the purpose of studying the horizontal and vertical vibrations. The equation of motion of the compound system (gyro-beam system) is introduced and solved to find the response measured on the primary system. This is fundamental to design, in some way, the dynamic absorber or neutralizer. On the other hand, the effect of the angular velocities of the gyroscopes are studied, and it is shown that the angular velocity (spin velocity) of a gyroscope has a significant effect on the behavior of the dynamic motion. Correctness of the analytical results is verified by numerical simulations. The comparison with the results from the derivation of the corresponding frequency equations shows that the optimized stiffness and damping values are very accurate.

Risk factors in the workplace vary according to the sector and scale of the business. Small and medium-sized enterprises, especially those within the scope of the wood-products manufacturing industry, are considered to be risky, and have a relatively high accident rate. Here, we focus on the timber and furniture industries, two subsectors of the wood-products industry. A total of 47 enterprises was visited and asked to participate in a formal structured questionnaire. The findings show that the usage of personal protective equipment (PPE) was low, that lighting was often inadequate, and that these enterprises typically lacked routine organising and cleaning practices. The subsectors and the occupational health and safety practices were found to be independent, and a statistically significant relationship could only be established between the subsectors in terms of the use of dust masks and goggles. Despite the unfavourable working conditions, only one business out of 47 reported having an employee with symptoms of occupational diseases.

Mineralogical and chemical properties, as well as the pozzolanic activities, of the volcanic rocks in the northeast of the Black Sea were examined. Physical and mechanical tests were conducted on samples collected from the study area and the feasibility of using volcanic rocks as pozzolanic admixture in the concrete industry was investigated. Mortar samples containing admixture in the ratios of 0%, 10%, 20%, 30%, 40%, and 50% were prepared by keeping fineness of pozzolanic admixtures fixed. By using these mortar samples, prismatic mortar samples of 4×4×16 cm were prepared and the samples were cured in water. Their 7-, 28-, and 90-day tensile strengths, compressive strengths, and ultrasonic pulse velocity were determined. As a result, it was determined that volcanic rocks can be used to produce blended cement by considering their specific gravity, pozzolanic activities, and compressive strengths. It was noted that increase in trass rate significantly reduced tensile and compressive strength of cement in the early stage as expected.

Demand for organic milk is partially driven by consumer perceptions that it is more nutritious. However, there is still considerable uncertainty over whether the use of organic production standards affects milk quality. Here we report results of meta-analyses based on 170 published studies comparing the nutrient content of organic and conventional bovine milk. There were no significant differences in total SFA and MUFA concentrations between organic and conventional milk. However, concentrations of total PUFA and n-3 PUFA were significantly higher in organic milk, by an estimated 7 (95% CI -1, 15)% and 56 (95% CI 38, 74) %, respectively. Concentrations of alpha-linolenic acid (ALA), very long-chain n-3 fatty acids (EPA + DPA + DHA) and conjugated linoleic acid were also significantly higher in organic milk, by an 69 (95% CI 53, 84) %, 57 (95% CI 27, 87)% and 41 ( 95% CI 14, 68) %, respectively. As there were no significant differences in total n-6 PUFA and linoleic acid (LA) concentrations, the n-6:n-3 and LA: ALA ratios were lower in organic milk, by an estimated 71 (95% CI -122, -20)% and 93 (95% CI -116, -70) %. It is concluded that organic bovine milk has a more desirable fatty acid composition than conventional milk. Meta-analyses also showed that organic milk has significantly higher alpha-tocopherol and Fe, but lower I and Se concentrations. Redundancy analysis of data from a large cross-European milk quality survey indicates that the higher grazing/conserved forage intakes in organic systems were the main reason for milk composition differences.

Demand for organic meat is partially driven by consumer perceptions that organic foods are more nutritious than non-organic foods. However, there have been no systematic reviews comparing specifically the nutrient content of organic and conventionally produced meat. In this study, we report results of a meta-analysis based on sixty-seven published studies comparing the composition of organic and non-organic meat products. For many nutritionally relevant compounds (e.g. minerals, antioxidants and most individual fatty acids (FA)), the evidence base was too weak for meaningful meta-analyses. However, significant differences in FA profiles were detected when data from all livestock species were pooled. Concentrations of SFA and MUFA were similar or slightly lower, respectively, in organic compared with conventional meat. Larger differences were detected for total PUFA and n-3 PUFA, which were an estimated 23 (95% CI 11, 35)% and 47 (95% CI 10, 84)% higher in organic meat, respectively. However, for these and many other composition parameters, for which meta-analyses found significant differences, heterogeneity was high, and this could be explained by differences between animal species/meat types. Evidence from controlled experimental studies indicates that the high grazing/forage-based diets prescribed under organic farming standards may be the main reason for differences in FA profiles. Further studies are required to enable meta-analyses for a wider range of parameters (e.g. antioxidant, vitamin and mineral concentrations) and to improve both precision and consistency of results for FA profiles for all species. Potential impacts of composition differences on human health are discussed.

Duong, Angela; Che, Yi; Ceylan, Deniz; Pinguelo, Arsene; Andreazza, Ana C.; Young, L. Trevor; Berk, Michael
Phenomenologically, bipolar disorder (BD) is characterized by biphasic increases and decreases in energy. As this is a state-related phenomenon, identifying regulators responsible for this phasic dysregulation has the potential to uncover key elements in the pathophysiology of BD. Given the evidence suggesting mitochondrial complex I dysfunction in BD, we aimed to identify the main regulators of complex I in BD by reviewing the literature and using the published microarray data to examine their gene expression profiles. We also validated protein expression levels of the main complex I regulators by immunohistochemistry. Upon reviewing the literature, we found PARK-7, STAT-3, SIRT-3 and IMP-2 play an important role in regulating complex I activity. Published microarray studies however revealed no significant direction of regulation of STAT-3, SIRT-3, and IMP-2, but a trend towards downregulation of PARK 7 was observed in BD. Immunocontent of DJ-1 (PARK-7-encoded protein) were not elevated in post mortem prefrontal cortex from patients with BD. We also found a trend towards upregulation of DJ-1 expression with age. Our results suggest that DJ-1 is not significantly altered in BD subjects, however further studies are needed to examine DJ-1 expression levels in a cohort of older patients with BD. Gocer, Hulya; Topal, Fevzi; Topal, Meryem; Küçük, Murat; Teke, Dilek; Gülçin, Ilhami; Alwasel, Saleh H.; Supuran, Claudiu T.
Taxifolin, also known as dihydroquercetin, is a flavonoid commonly found in plants. Carbonic anhydrase (CA, EC 4.2.1.1) plays an important role in many critical physiological events including carbon dioxide (CO2)/bicarbonate (HCO3-) respiration and pH regulation. There are 16 known CA isoforms in humans, of which human hCA isoenzymes I and II (hCA I and II) are ubiquitous cytosolic isoforms. In this study, the inhibition properties of taxifolin against the slow cytosolic isoenzyme hCA I, and the ubiquitous and dominant rapid cytosolic isoenzyme hCA II were studied. Taxifolin, as a naturally bioactive flavonoid, has a K-i of 29.2nM against hCA I, and 24.2nM against hCA II. For acetylcholinesterase enzyme (AChE) inhibition, K-i parameter of taxifolin was determined to be 16.7nM. These results clearly show that taxifolin inhibited both CA isoenzymes and AChE at the nM levels.

PURPOSE We aimed to demonstrate the success and reliability of a novel puncture, aspiration, injection, and reaspiration (PAIR) technique in liver hydatid cysts. METHODS Percutaneous treatment with ultrasonographic guidance was performed in 493 hepatic hydatid cysts in 374 patients. Patients were treated with a new PAIR technique by single puncture method using a 6F trocar catheter. The results of this novel technique were evaluated with regards to efficacy and safety of the procedure and complication rates. RESULTS Out of 493 cysts, 317 were Gharbi type I (WHO CE 1) and 176 were Gharbi type II (WHO CE 3A). Of all cysts, 13 were referred to surgery because of cystobiliary fistulization. Recurrence was observed in 11 cysts one month later. Therefore, the success rate of the PAIR technique was 97.7% (469/480). Minor complications (fever, urticaria-like reactions, biliary fistula) were seen in 44 treated patients (12%, 44/374); the only major complication was reversible anaphylactic shock which was observed in two patients (0.5%, 2/374). CONCLUSION This novel modified PAIR technique may be superior to catheterization by Seldinger technique due to its efficiency, easier application, lower severe complication rate, and lower cost. Further comparative studies are required to confirm our observations.

- MakaleAçık Erişim2016Scopus SJRQ1Complex subvolcanic magma plumbing system of an alkali basaltic maar-diatreme volcano (Elie Ness, Fife, Scotland)
Gernon, T. M.; Upton, B. G. J.; Ugra, R.; Yücel, C.; Taylor, R. N.; Elliott, H.
Alkali basaltic diatremes such as Elie Ness (Fife, Scotland) expose a range of volcanic lithofacies that points to a complex, multi -stage emplacement history. Here, basanites contain phenocrysts including pyrope garnet and sub-calcic augites from depths of similar to 60 km. Volcanic rocks from all units, pyroclastic and hypabyssal, are characterised by rare earth element (REE) patterns that show continuous enrichment from heavy REE (HREE) to light REE (LREE), and high Zr/Y that are consistent with retention of garnet in the mantle source during melting of peridotite in a garnet lherzolite fades. Erupted garnets are euhedral and unresorbed, signifying rapid ascent through the lithosphere. The magmas also transported abundant pyroxenitic clasts, cognate with the basanite host, from shallower depths (similar to 35-40 km). These clasts exhibit wide variation in texture, mode and mineralogy, consistent with growth from a range of compositionally diverse melts. Further, clinopyroxene phenocrysts from both the hypabyssal and pyroclastic units exhibit a very wide compositional range, indicative of polybaric fractionation and magma mixing. This is attributed to stalling of earlier magmas in the lower crust principally from similar to 22 to 28 km as indicated by pyroxene thermobarometry. Many clinopyroxenes display chemical zoning profiles, occasionally with mantles and rims of higher magnesium number (Mg#) suggesting the magmas were mobilised by juvenile basanite magma. The tuffs also contain alkali feldspar megacrysts together with Feclinopyroxene, zircon and related salic xenoliths, of the 'anorthoclasite suite' - inferred to have crystallised at upper mantle to lower crustal depths from salic magma in advance of the mafic host magmas. Despite evidence for entrainment of heterogeneous crystal mushes, the rapidly ascending melts experienced negligible crustal contamination. The complex association of phenocrysts, megacrysts and autoliths at Elie Ness indicates thorough mixing in a dynamic system immediately prior to explosive diatreme-forming eruptions.
