Impact of Phytobiotic Supplementation Administered in Drinking Water on Meat Quality in Heat-Stressed Broilers

    Kaya, Hacer; Kamiloğlu, Aybike

This study evaluated the effects of a phytobiotic additive, hawthorn fruit extract (HFE), administered via drinking water on meat quality of broilers subjected to heat stress (HS). A total of 300 one-day-old broiler chicks were distributed into six experimental groups according to a 2×3 factorial design after a one-week acclimation period, with five replicates of 10 chicks each. Chicks had ad libitum access to feed and water for the duration of the 42-day trial and were exposed to a 24-hour lighting program. HS decreased the b* color value of breast meat while increasing L* and a* values and pH of thigh meat. Cooking loss (CL) of both breast and thigh meat was reduced, whereas water-holding capacity (WHC) remained unaffected. HS did not significantly alter thiobarbituric acid-reactive substances (TBARS), total saturated fatty acids (ΣSFA), or total unsaturated fatty acids (ΣUFA), but decreased monounsaturated fatty acids (ΣMUFA) and increased emulsion stability (ES) and polyunsaturated fatty acids (ΣPUFA). Adding HFE to the drinking water did not influence breast meat pH or color intensity, but WHC improved in breast meat with 0.2 mL/L HFE and in thigh meat at both HFE concentrations. The 0.4 mL/L HFE dose increased ΣSFA and decreased ΣUFA and ΣPUFA levels, while ΣMUFA decreased at both HFE concentrations. Overall, HFE supplementation mitigated several adverse effects of HS, notably enhancing ES and reducing redness of thigh meat. These findings suggest that HFE can be an effective phytobiotic strategy to improve meat quality in broilers under thermal stress conditions.

Serum and hepatic lipid fractions and total protein status of laying hens supplemented with tarragon (Artemisia dracunculus L.) under different stocking densities

Kaya, Hacer; Karaalp, Musa; Kaynar, Özgür; Elmastaş, Mahfuz

High stocking density can induce metabolic disturbances in laying hens, particularly in lipid metabolism. This study evaluated the effects of dietary tarragon (Artemisia dracunculus L.) supplementation on serum and hepatic total protein and lipid parameters in laying hens reared under different stocking densities. A total of 192 50-week-old Lohmann Brown laying hens were assigned to a 2 × 4 factorial arrangement with two stocking densities, 5 or 7 hens per cage, and four dietary tarragon treatments providing 0, 1, 5, or 10 mmol/kg feed of tarragon-derived additional antioxidant activity for 8 weeks. Serum total protein concentration was not affected by stocking density but increased with tarragon supplementation, rising from 7.5 to 8.3 g/dL. Liver total protein concentration remained unchanged. SDS-PAGE analysis of pooled treatment-group samples identified 24 serum and 39 hepatic protein bands. High stocking density altered the serum lipid profile by increasing cholesteryl ester, free fatty acid, and acylglycerol proportions, as well as total cholesterol and triacylglycerol concentrations, while decreasing triacylglycerol and phospholipid proportions. Tarragon supplementation increased serum cholesteryl ester and phospholipid proportions and reduced serum triacylglycerol and free fatty acid proportions, together with lower total cholesterol and triacylglycerol concentrations. In the liver, high stocking density increased cholesteryl ester and free cholesterol proportions and triacylglycerol concentration, whereas it decreased triacylglycerol and phospholipid proportions and total cholesterol concentration. Tarragon supplementation increased hepatic cholesteryl ester, free cholesterol, and phospholipid proportions and reduced hepatic triacylglycerol proportion and concentration. Significant stocking density × tarragon-derived additional antioxidant activity interactions were observed for serum free fatty acid and hepatic free fatty acid and free cholesterol. Overall, stocking density had a stronger effect on lipid metabolism than on total protein status.

Influence of Hawthorn (Crataegus tanacetifolia) Fruit Vinegar Supplementation on Meat Quality and Gut MicrobiotainHeat-StressedBroilers; [Alıç (Crataegus tanacetifolia) Meyve Sirkesi Takviyesinin Isı Stresi Altındaki Etlik Piliçlerde Et Kalitesi ve Bağırsak Mikrobiyotası Üzerindeki Etkisi]

    Kaya, Hacer; Kamiloğlu, Aybike

This study investigated the influence of the consumption of hawthorn (Crataegus tanacetifolia) fruit vinegar (HFV), a fermented product, on meat quality and gut microbiota in broiler chickens under cyclic heat stress (HS). A total of 300 one-day-old male Ross 308 chicks were divided into six groups with five replicates of ten birds, following a 2×3 factorial design with two temperatures (24 and 35°C) and three HFV supplementing levels (0, 2, and 4 mL/L) after one-week acclimation. Broilers in the thermoneutral groups were kept at 24°C, while HS groups experienced 35°C for 8 hours daily from day 21. Feed and water were provided ad libitum under a 24-hour lighting program. HS increased breast pH, reduced b* color value, raised ΣSFA and ΣMUFA levels, and lowered ΣUFA and ΣPUFA ratios. Supplementing 4 mL/L HFV increased breast L* color value and ΣSFA level while reducing ΣUFA ratio. The 2 mL/L HFV dose decreased ΣMUFA and increased ΣPUFA levels. Overall, HFV supplementation improved meat quality and beneficially altered fatty acid composition in heat-stressed broiler breasts. These results suggest that HFV supplementation may be a practical natural additive for broilers under cyclic heat stress and provide new evidence regarding its combined effects on meat quality and intestinal microbiota. Hayvan yemlerine yeşil sentez metalik nanopartikül ilavelerinin etkileri

    Kaya, Hacer

    Kaya, Hacer; Mazlum, Halit

Response of tibia bone morphology, biomechanics and minerals to Organic Hawthorn (Crataegus tanacetifolia) Fruit Vinegar application in broiler chickens exposed to cyclic heat stress

    Kaya, Hacer

This study aimed to determine the effects of adding different doses of organic hawthorn fruit vinegar (HFV) to the drinking water of male broilers (Ross 308) reared under cyclic heat stress (CHS) on tibia bone morphology, biomechanics, and minerals. Broilers were distributed into six groups based on a 2 × 3 factorial design, testing two ambient temperatures (24 and 35°C CHS for 8 h/day starting from day 21) and three HFV levels (0, 2, and 4 ml/L). Cyclic heat stress had no effect on the morphological and biomechanical properties of the tibia bone, except for a decrease in cortex thickness (P < 0.05). However, CHS significantly decreased the bone Ca, P, Mg, Cu, Zn, Mn, and K levels, while increasing the Fe levels (P < 0.01). The addition of 2 and 4 ml/L HFV had no effect on the morphological and biomechanical properties of the tibia bone (P > 0.05), except for an increase in bone diameter with 2 ml/L HFV (P < 0.05). HFV additions were not sufficient to increase the Ca level decreased by CHS, and the tibia bone Ca level of birds under CHS decreased further with HFV additions (P < 0.01). Conversely, 2 ml/L HFV increased Mg levels, and both 2 and 4 ml/L HFV increased Cu, Fe, Zn, and Mn levels, whereas P levels decreased (P < 0.01). K level decreased with 2 ml/L HFV and increased with 4 ml/L HFV supplementation. In conclusion, HFV supplementation has positive effects on bone mineralization in broilers exposed to HS and can be used up to 4 ml/L to mitigate the negative effects of heat stress.

The Effect of Vitamin C and E Supplementation into Drinking Water on Carcass Characteristics, Meat Quality and Intestinal Microflora During Pre-Slaughter Feed Withdrawal in Broiler Chickens

    Kaya, Hacer

This study investigates the effects of adding vitamin C and E to the drinking water on carcass characteristics, meat quality and intestinal microflora populations in broiler chickens during the 10-h pre-slaughter feed withdrawal (FW) period. As study materials, forty male broilers at the age of 42 days were used. The broilers were randomly divided into four groups: Control (non-vitamin, NV), vitamin C (1000 mg/L, VC), vitamin E (500 mg/L, VE) and vitamin combination (1000 mg/L VC+500 mg/L VE, VCE). In the study, vitamin additions didn’t affect carcass characteristics, visceral weights and the pH values of the digestive system (P>0.05). The addition of VC and VE increased the weight of the Bursa of Fabricius, and the addition of VE increased the weight of thymus (P<0.05). Additions of vitamin decreased tendency of carcass contamination (P<0.01) and increased pH45min and pH24h of thigh meat and pH24h of breast meat (P<0.05, P<0.01, P<0.01, respectively). While a* color intensity of breast and thigh meat increased with all vitamin supplements, L* and b* values of thigh meat decreased (P<0.01). Vitamin supplements, especially VE, reduced the drip loss of breast and thigh meat (P<0.05) and the pathogenic microorganism populations of intestinal contents (P<0.01). As a result, it is thought that the addition of 500 mg/L vitamin E to the drinking water of broiler chickens exposed to the pre-slaughter fasting period will be beneficial to improve meat quality and reduce intestinal pathogenic microorganism load. However, more extensive experimental studies are needed.

The Effects of Hawthorn (Crataegus oxyacantha) Fruit Extract on Performance, Carcass and Some Serum Parameters of Broilers Reared Under Heat-Stress Conditions[2]

    Kaya, Hacer

Background: The present experiment was conducted to investigate the effect of heat stress (HS) and hawthorn (Crataegus oxyacantha) fruit extract (HFE) on performance, carcass characteristics and serum parameters in broiler chickens. Methods: Experimental treatments included two different ambient temperatures (24°C: thermoneutral conditions, or 35°C: HS conditions) and three different HFE (0, 0.2 and 0.4 ml/L) in a 2×3 factorial arrangement. A total of 300 1-day-old broiler chicks were randomly allotted 1 of 6 treatments with five replicates. A total of 300 1-day old broiler chicks were randomly allocated to 6 groups with five replications of 10 birds each. HFE supplementation had no effect on growth performance, carcass characteristics and visceral weight in stressed groups (P>0.05). It was also determined that it has a lowering effect on serum malondialdehyde (MDA), free fatty acid (FFA), triacylglyceride (TAG), total lipid (TL) and low density lipoproteins (LDL) (P<0.05), but it has no effect on other parameters (P>0.05). Result: As a result, it is thought that the addition of HFE to the drinking water of broiler chickens exposed to HS has no effect on the performance characteristics, HFE has a hypolipidemic effect and would be beneficial in order to strengthen the immune system.

Progranulin (PGRN) is a multifunctional protein that is critical in the retention of embryos in the endometrium of the uterus and the protection of cervical tissue during fetal growth. In case of PGRN release and dysfunction, cervical tissue may remodel (regeneration) and lead to the risk of abortion. Serum PGRN level in sheep and its variation according to different physiological periods and its relationship with some biochemical parameters are unknown. In this study, the relationship between early lactation, early and late pregnancy serum PGRN levels and some biochemical parameters in Akkaraman sheep was investigated for the first time. In the literature search, no similar studies were found on the relationship between early lactation, early and late pregnancy serum PGRN levels and some biochemical parameters in Akkaraman sheep. The research was carried out on 90 Akkaraman sheep reared in the same herd under the same care and feeding conditions. In the study, Akkaraman ewe were divided into 3 groups as early lactation, early pregnancy and late pregnancy. 1st group is early lactation period ewe (first 60 days of lactation), (n = 30), 2nd group is in the first period of pregnancy (on the 0-90th day of the first pregnancy (n = 30)), 3rd group is in the late pregnancy of pregnancy (100-150 days of pregnancy (n = 30)) was created. Blood samples from vena jugularis were taken before morning feeding from the animals that formed the study groups. PGRN levels in serum were determined by ELISA method. In addition, glucose, calcium (Ca), magnesium (Mg), phosphorus (P), total cholesterol (TC), triglyceride (TG), low density lipoprotein (LDL), high density lipoprotein (HDL), very low density lipoprotein (VLDL) levels were analyzed in biochemistry autoanalyzer. As a result of the study, lactation and early pregnancy and late pregnancy mean serum PGRN levels were determined as 1.93, 1.78, 1.33 ng mL-1, respectively. The serum PGRN, glucose, Ca, Mg, P, TC, TG, LDL, HDL and VLDL levels of sheep with late pregnancy, early lactation and early pregnancy periods, which constitute the study groups, were found to be statistically significant (P<0.05). Our results have shown that due to the physiological role of PRGN in keeping the embryo in the endometrium of the uterine and in preserving the cervical tissue during fetal growth, there may be a useful parameter in the pursuit of the pregnancy process.

-In this study, we aimed to determine the effect of Moringa oleifera essential oil (MOEO) supplementation to rations of Japanese quail (Coturnix coturnix japonica) grown under heat stress (HS) on some adipokine (visfatin, adiponectin, and chemerin), intestinal (citrulline), 3,5,3 '-triiodothyronine (T3), thyroxine (T4), and thyroid-stimulation hormone (TSH) levels in the serum. For this purpose, 320 day-old male quail chicks were used. The study was conducted over 42 days, including a 7-d adaptation period and a 35-d experimental period. Eight trial groups were established, each including 10 quail: CT (control temperature, 25 celcius), MOEO 200, MOEO 400, MOEO 600, HSC (HS control), SMOEO 200, SMOEO 400, and SMOEO 600. Based on the results, a dose-dependent decrease was observed on days 21 and 42 in serum adiponectin and T3 in the stress and stress-free groups compared with the control group due to MOEO supplementation. The lowest decrease was observed in the MOEO 600 mg/kg dose group. In addition, an increase in stressed groups was observed when examining serum citrulline levels, while chemerin levels did not have a statistically significant effect. While the decrease in serum visfatin, T4, and TSH levels on the 21st day did not have a statistically significant effect, a significant effect was determined on the 42nd day. The addition of MOEO at 600 mg/kg to the ration may be beneficial in preventing intestinal damage and inflammation that may occur due to HS.

Bayraktar, Bulent; Tekce, Emre; Kaya, Hacer; Gürbüz, Ahmet Burak; Dirican, Ebubekir; Korkmaz, Serol; Atalay, Banu; Ülker, Ufuk

The aim of this study was to investigate the effect of Lactobacillus reuteri E81 (LRE) probiotic supplementation on heat stress responses in chukar partridges (Alectoris chukar). The birds were divided into two groups, one of which was exposed to heat stress (HS). Within each group, four subgroups, each including 64 birds, were created for the three treatment doses (200, 400 or 600 mg/kg) of LRE and the control. The experiment was started with day-old birds, kept at a temperature of 25 degrees C or 37 degrees C. After a 7-day adjustment period, the LRE supplementation lasted for 35 days. The levels of different adipokines, including visfatin (VF), adiponectin (ADP), chemerin (CHEM), as well as the concentration of plasma citrulline (CIT) and the levels of thyroid hormones (T3 and T4) and thyroid-stimulating hormone (TSH) in the blood were measured at 21 and 42 days of age. A significant correlation (P < 0.01) was found between LRE supplementation and the decrease in serum VF, ADP, CIT, T3 and T4 levels in partridges exposed to HS. On the other hand, no significant relationship was found between LRE supplementation and the serum CHEM and TSH levels (P > 0.05). We concluded that the addition of 600 mg/kg LRE is beneficial in preventing intestinal damage and inflammation provoked by HS.

The Effects of Boron (Orthoboric Acid) Supplementation into Diets on Serum Lipid and Protein Profiles of Hens During Late Laying Period

    Kaya, Hacer; Macit, Muhlis

This study was carried out to determine the effects of boron addition into the diets on serum parameters of hens. A total of 288 Lohman laying commercial hybrid chickens of 62 weeks were fed for 12 weeks with four different rations containing basal feed 0 (B0), 50 (B50), 75 (B75) and 150 (B150) mg / kg B. Each group was replicated in 18 cages, four hens per cage. During the study, 16 hours lighting program was applied, feed and water were given as ad-libitum. The effect of boron supplementation was not significant on the rates of diacylglycerol and hydrocarbon+cholesterol esters (P>0.05) but the rates of polarlipid, triacylglycerol (P<0.05), and serum total cholesterol (P<0.01) were found significant. Lipid peroxidation values of groups fed with diets including 75 and 150 mg/kg B were determined lower than others. Addition of ration B affected different molecular weight proteins at different levels. In conclusion, it has been determined that the addition of B at different levels into the diets of laying hens during late laying period significantly influenced the serum lipid peroxidation, serum lipid and serum protein profile, but further researches are needed to clarify the changes related to the effects of B addition into diets on mentioned parameters of laying hens.

The impact of dietary tarragon (Artemisia dracunculus) on serum apelin, brain-derived neurotrophic factor, cardiac troponin concentrations and histopathology of liver tissue in laying hens housed at different stocking densities

Bayraktar, Bulent; Tekce, Emre; Kaya, Hacer; Karaalp, Musa; Turunc, Eylul

Due to its association with several other stress factors (poultry house gases, inadequate ventilation, heat, cold and poor hygiene), the high stocking density is a major stress factor that adversely affects the health and performance of poultry and the quality of the poultry products. Therefore, this experimental study was aimed at analysing the impact of different doses of dietary tarragon (Artemisia dracunculus) on the serum apelin, plasma brain-derived neurotrophic factor (p-BDNF), and cardiac troponin I (cTnI) concentrations, and the correlation between these indicators in laying hens housed at different stocking densities. The aim of this study is to investigate the effects of adding tarragon in different ratios to laying hen rations in the 2ndovulation period on the cTnI, apelin, and BDNF hormone concentrations and the liver histopathology. The experiment was carried out over a period of eight weeks, with 192 Lohman Brown commercial hybrids at 50 weeks of age. Eight groups (four replicates each), composed of laying hens of equal body weight, which were housed at stocking densities of 580 cm2/hen and 810 cm2/hen and received 0, 1, 5 and 10 mmol/kg of tarragon (Artemisia dracunculus) in the feed, were established. At the end of the trial, 96 of the housed egg-laying hens (3 birds in each subgroup, a total of 12 birds in each group) were randomly selected and blood samples were taken from the vena subcutanea ulnaris. The samples collected were analysed for the apelin, p-BDNF, and cTnI contents. The analysis results demonstrated that tarragon supplementation had no effect on the serum apelin, p-BDNF and cTnI concentrations (P > 0.05). The Sub-Groups ST1, ST1.2, and ST6 presented with severe hyperaemia of the sinusoidal, portal and acinar blood vessels, whilst the hyperaemia of these blood vessels was moderate in Sub-Group ST12. Apelin, BDNF, and cTnI can act as protective factors against negative consequences of stress (e.g., stocking density or heat stress).

Response of Japanese quails (Coturnix coturnix japonica) to dietary inclusion of Moringa oleifera essential oil under heat stress condition

Tekce, Emre; Bayraktar, Bülent; Aksakal, Vecihi; Dertli, Enes; Kamiloğlu, Aybike; Çinar Topcu, Kübra; Takma, Çiğdem; Gül, Mehmet; Kaya, Hacer

The present study was aimed at investigating the effects of the dietary use of different doses of Moringa oleifera essential oil (MOEO) on performance parameters, internal organ weights and meat quality in Japanese quails (Coturnix coturnix japonica) exposed to Heat Stress (HS). For this purpose, 320 male day-old quail chicks were used. Seven days of preliminary (physical exercise) and a 35-day fattening period were applied. The animals were divided into 8 groups, each including 40 animals according to temparature (C: 25 °C and SC: 37 °C) and dietary doses (C, 200, 400 and 600). Each group was divided into 4 subgroups, each comprising 10 animals. The consequences of this study showed that, in the stress-free (HSF) groups, while all of the different doses of dietary MOEO positively affected growth performance, feed intake and feed efficiency, in the HS groups, similar positive effects were observed with the use of 200 ppm of dietary MOEO. While no effect was detected on internal organ weights on Days 21 and 42 in the HSF and HS groups, a dose-dependent increase was determined in the intestinal pH value on day 42 in the HS group (p <.05). Additionally, in the analysis of meat quality on the 21st and 42 days compared to the controls, no effect was observed on the meat colour parameters value and TBARS level in the HS and HSF groups. Based on the consequences of this study, it is considered that, MOEO could be used as an alternative product in poultry, at a dose of 200 ppm, to reduce the detrimental effects of HS.HIGHLIGHTS Temperature stress, performance parameters and serious problems in the health of poultry. Moringa olifera helps regulate performance parameters in poultry exposed to heat stress. Since the addition of Moringa oleifera to the diets does not affect the organ weight and meat quality in 21 and 42 days, it does not cause any negative effects for the consumer.

The Effects of Boron (Orthoboric Acid) Supplementation Into Diets of Hens During Late Laying Period on Performance and Some Serum Parameters

    Kaya, Hacer; Macit, Muhlis

This study was carried out to determine the effects of boron (orthoboric acid) addition to the diets of hens in the late laying period on performance and some serum parameters. Two hundred eighty-eight Lohman commercial laying hens which are 62 weeks old were fed with 0, 50, 75, and 150 mg/kg of B for 12 weeks. The research was carried out in 18 replicates, and four laying hens were used for each replicate. During the research, 16-hour lighting was applied and feed and water were given as ad-libitum. Supplementation of different level of B did not alter the live weight change and average egg weight from performance characteristics, but egg production increased and also feed efficiency and daily feed intake reduced (P<0.01). Serum glucose, cholesterol, triglycerides, creatinine, total and direct bilirubin values were not affected by additional B. While addition of B into the diets of laying hens did not affect the outcomes of serum alkaline phosphatase (ALP), aspartate amino transaminase (AST) and gamma glutamyl transaminase (GGT) enzyme activities (P>0.05), but alanin amino transaminase (ALT) was affected by Boron (P<0.05). It is concluded that in order to have a positive effect on performance of laying hens in the late laying period, 50 mg/kg B supplementation is adequate and may be suggested.
