Acet, Tuba; Özcan, Kadriye; Sefalı, Abdurrahman
Psephellus appendicigerus, is a plant endemic to Turkey, belonging to the Asteraceae family, which is traditionally used in folk medicine in Turkey. The aim of this study is to reveal for the first time the biological activities of this plant and evaluate its potential use as a natural product. For this reason, different extracts (ethanol, methanol, ethyl acetate, and hexane) of the plant’s aerial parts were used in determining the antioxidant (2,20-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid and 1,1-diphenyl-2-picrylhydrazyl assays), antimicrobial (disc diffusion and microdilution methods), and enzyme inhibition activity (calorimetric methods) related to diabetes (α-amylase and α-glucosidase). Moreover, the phenolic constituents were analysed by high performance liquid chromatography. Methanol extract of the plant exhibited significant biological activity compared to other extracts. This extract had the highest amount of chlorogenic acid as phenolic compound. In addition, all extracts showed inhibition against at least one test microorganism. According to the results, P. appendicigerus has the potential to be used as a natural antioxidant, antimicrobial and enzyme inhibitor.

Kaplan, Mahmut; Yüksel, Ferhat; Karaman, Kevser
In this study, deep fried gluten free chips were manufactured using sorghum flour at different concentrations (5, 20, 35 and 50 g/100 g) and in vitro glycemic index, total antioxidant capacity and some physicochemical characteristics (dry matter, ash, oil, protein and color) and overall acceptability of the samples were investigated. Protein, oil and total dietary fiber levels of the samples ranged between 5.39–8.28, 23.58–29.10 and 5.59–6.60 g/100 g, respectively and increase in sorghum flour level resulted in an increase in these studied quality parameters of the chips samples. Ferric reducing antioxidant activity and DPPH radical scavenging performance were affected positively by the addition of sorghum flour. Total starch hydrolysis index and estimated glycemic index values were affected by the sorghum flour addition negatively due to the increased total dietary fiber content of the chips samples. All chips samples enriched with sorghum flour showed similar overall acceptability and increased sorghum concentration decreased the preference scores slightly. All these results could be considered for the manufacturing of the gluten free deep fried sorghum based chips samples.

Valerian (Valeriana dioscoridis Sm.) is a perennial herb of the Caprifoliaceae family. The genus Valeriana L. is represented by 14 species (15 taxa) in Turkey. This plant contains flavone glycosides, iridoids and lignans. Among these components, the medically important active ingredient is valerianic acid. Essential oils from valerian roots and rhizomes are used for the treatment of various diseases, including insomnia, mental illness, anxiety, menstrual cramps and physical stress conditions. In this study, evaluations were made of the root and rhizome of valerian plants grown in a natural environment and in greenhouse condition. The macro and micro nutrient contents of the powdered plant samples, and the antioxidant and antimicrobial activity values of the extracts were reported. Except for some macro and micro elements, it was determined that antioxidant and antimicrobial activities of plants cultivated and grown in nature were not different. The major components of both the natural and cultivated forms were determined to be 9-Borabicyclo [3.3.1] nonane, 9-[3-(dimethylamino) propyl]- (17.55% and 22.65%, respectively). The heavy metales such as Fe (415.21±47.8 mg/kg), Cu (50.9±0.2 mg/kg) and Mn (274.6±9.5 mg/kg), were obtained above limit values in grown plants of natural environment conditions.

Kotiloğlu, Deniz; Acet, Tuba; Özcan, Kadriye
In Anatolia, the tubers of Dactylorhiza romana subsp. georgica orchid are consumed as food and frequently used in folk medicine to alleviate the symptoms of some disorders. However, these properties have not been scientifically studied until the present day. In this research, we aimed to identify phenolic profile of the orchid tubers and determine their several features and functions. Among these, there are the antioxidant and antimicrobial effects and the inhibitory effect against diabetes related enzymes (α-amylase and α-glucosidase). In this sense, total phenolic content (Folin–Cicalteu assay), total antioxidant capacity (Phosphomolybdate method), antioxidant (ABTS and DPPH assays) and enzyme inhibition effects of tuber extracts obtained with different solvents (chloroform, ethyl acetate, methanol, ethanol, or hexane) were determined by using spectrophotometric methods, and the phenolic profile was also analysed by HPLC. In addition, antimicrobial activity of the extracts against 3 g (+) and 3 g (−) bacteria and 2 forms of yeasts was determined by disc diffusion and micro dilution methods. Based on our findings, the total phenolic content and antioxidant properties of ethyl acetate extract were considerably higher than the others. The most abundant among the screened phenolic standards was found to be benzoic acid. The highest α-amylase and α-glucosidase enzyme inhibition was measured in methanol and chloroform extracts, respectively. The highest antibacterial (B. cereus) and anticandidal (C. tropicalis and C. albicans) activities were detected in the chloroform and ethyl acetate extracts, respectively. In short, it was clearly seen that the D. romana subsp. georgica tuber had a great potential for being used in food and drug industries as a source of bioactive compounds.

Ilyasoglu, Huri; Nadzieja, Marcin; Guo, Zheng
The aim of this study was to develop a novel dual-functional stabilizer for food-grade Pickering emulsions by grafting caffeic acid onto chitosan via a free radical mediated method. The grafting of caffeic acid onto chitosan was confirmed by UV-vis absorption spectra, FTIR spectra and H-1 NMR spectra and DSC thermogram. The antioxidant properties of the caffeic acid grafted chitosan (CA-g-Ch) were measured by three in vitro assays. Caffeic acid-grafted chitosan displayed much better Fe2+ chelating property than both chitosan and caffeic acid, and retained comparable DPPH radical scavenging activity as caffeic acid. Moreover, the formation of the CA-g-Ch based emulsion was monitored by the turbidity, zeta potential, and particle size measurements. The CA-g-Ch polymers were found to be pH-responsive. The oil (linoleic acid) in water emulsions with the CA-g-Ch (as emulsifier/Pickering stabilizer) was prepared to examine its potential to stabilize Pickering emulsions, as well as its additive antioxidant properties. The results (zeta potential, mean droplet size, creaming index and TBARS values etc) demonstrated that the CA-g-Ch could act as a dual-functional stabilizer for Pickering emulsions. The emulsions made with the CA-g-Ch were found to be stable at a broad pH range of 3-6. Our findings proved that CA-g-Ch as novel dual-functional polymeric surfactant, may have great potential to find applications in food and pharmaceutical industries.

Tiryaki, Deniz; Aydın, İhsan; Atıcı, Ökkeş
We have isolated psychrotolerant bacteria from the leaf apoplast of cold-adapted wild plants and aimed to investigate their effect on the cold resistance of bean (Phaseolus vulgaris L.). Based on the findings of 16S rRNA gene sequence analysis, 20 isolates belonging to 5 bacteria species (Pseudomonas fragi, P. chloropaphis, P. fluorescens, P. proteolytica and Brevibacterium frigoritolerans) were identified in the leaf apoplastic fluid of Draba nemorosa, Galanthus gracilis, Colchicum speciousum, Scilla siberica, Erodium cicutarium, respectively. We have determined that 6 of the 20 isolates have exhibited ACC (1-aminocyclopropane-1-carboxylate) deaminase activity and secreted different extracellular proteins under cold condition (+4 °C) compared to normal growth condition (28 °C). The six isolates were then inoculated independently of each other to the leaves of 10-day-old bean seedlings growing under normal conditions (25/22 °C, 16/8 h photoperiod), and the inoculated and uninoculated (control) seedlings were transferred to cold (9/5 °C, 16/8 h photoperiod) for 3 days. The bacterial inoculations have decreased freezing injury, ice nucleating activity and lipid peroxidation content in parallel with the decrease of reactive oxygen species level such as O2.- and H2O2 in the inoculated seedlings compared to the control. In addition, the inoculations of the isolates have stimulated the activity of apoplastic antioxidant enzymes including superoxide dismutase, catalase, peroxidase, and glutathione reductase. The results show that the inoculations improve the cold resistance of bean seedlings and the psychrotolerant bacterial isolates can be evaluated within the group of plant growth promoting bacteria (PGPB) which can increase tolerance of cold-sensitive crops.

Akar, Zeynep; Burnaz, Nesibe Arslan
Cupric reducing antioxidant capacity (CUPRAC) method is one of the most widely used antioxidant methods. However, its need for a spectrophotometer, as in many other methods, is an important disadvantage. Cupric reducing antioxidant capacity was determined based on color analysis with Image J program in computer with the new method developed in this study and thus, use of spectrophotometer was eliminated. The new colorimetric CUPRAC assay was applied to various standard antioxidants and plant extracts. The reaction mixtures were dropped on TLC plate. The colors formed on TLC were transferred to the computer by using scanner images, and CTEAC value in μmol/L of each sample was determined with the image J program. The CTEAC values were compared with the TEAC values measured with the spectrometric method. A linear relationship was found between both (C)TEAC values of antioxidant standards (R2 = 0.991) and plant extracts (R2 = 0.994). Thus, the new method developed for the CUPRAC antioxidant assay can be easily performed in any environment without spectrophotometer device, and it is a low-cost method.

Akar, Zeynep; Küçük, Murat; Doğan, Hacer
2,2-Diphenyl-1-picrylhydrazyl (DPPH•) radical scavenging, the most commonly used antioxidant method with more than seventeen thousand articles cited, is very practical; however, as with most assays, it has the major disadvantage of dependence on a spectrophotometer. To overcome this drawback, the colorimetric determination of the antioxidant activity using a scanner and freely available Image J software was developed. In this new method, the mixtures of solutions of DPPH• and standard antioxidants or extracts of common medicinal herbs were dropped onto TLC plates, after an incubation period. The spot images were evaluated with Image J software to determine CSC50 values, the sample concentrations providing 50% colour reduction, which were very similar with the SC50 values obtained with spectrophotometric method. The advantages of the new method are the use of lower amounts of reagents and solvents, no need for costly spectrophotometers, and thus significantly lowered costs, and convenient implementation in any environment and situation.

Key compound 2-(4-amino-5-oxo-3-(thiophene-2-ylmethyl)-4,5-dihydro-1,2,4- tiazole-1-yl) acetohydrazide (3) was synthesized by reacting hydrazine hydrate with ethyl-2-(4-amino-5-oxo-3-(thiophene-2-ylmethyl)-4,5-dihydro-1,2,4-tiazole- 1yl)acetate (2), obtained in basic media from 4-amino-5-(thiophene-2-ylmethyl)- 2H-1,2,4-triazole-3(4H)-one (1). Compound 3 was converted to thiosemicarbazide derivatives (4a-d) and Schiff base derivatives 6a-e and 7a-e. The treatment of compound 4 with NaOH gave 4-amino-2-((4-(4-aryl)-5-mercapto-4H-1,2,4-triazole-3- yl)methyl)-5-(thiophene-2-ylmethyl)-2H-1,2,4-triazole-3(4H)-ones (5a-d). All newly compounds, well characterized by elemental analyses, IR, 1H NMR, 13C NMR and mass spectral studies were tested for their antioxidant and antimicrobial activities. Thiosemicarbazide derivatives (4a-d) were highly active in two antioxidant tests with 69.0-88.2% DPPH scavenging and 503-1257 μM TEAC values, while the others showed lower or no activity. The results of the two antioxidant tests correlated well. Moreover, Thiosemicarbazide derivatives (4a-d) also showed antibacterial activity against Staphylococcus aureus, Bacillus cereus, and Mycobacterium smegmatis. Thiosemicarbazide group deserves attention in the synthesis of bioactive compounds.

The aim of this study was to investigate the effect of black cumin (Nigella sativa L.) essential oil on butter stability. For this aim, 0.05, 0.1 and 0.2 wt-% essential oil was added to the butter. Antioxidant activity of the essential oil was compared to that of synthetic antioxidant BHT (100 ppm). All samples were stored at 4±1°C for 90 days. Thiobarbituric acid and peroxide values of all samples containing essential oil were decreased depending on concentrations. The amount of 0.2% of essential oil had showed strong antioxidant activity, which was almost equal to that of BHT. The addition of essential oil reduced total aerobic mesophilic bacteria, lactic acid bacteria and coliform bacteria count during storage period but did not show remarkable antifungal activity. Samples containing essential oil were preferred by the panellists compared to Control sample. Present results indicate that black cumin essential oil may be considered as a new source of natural antioxidant.

Testicular torsion and detorsion are important clinical problems for infertile man and oxidative stress may have a role in this clinical situation. The aim of this study was to investigate the protective role of erdosteine, an antioxidant, on unilateral testicular reperfusion injury in rats. The rats were divided into four groups including seven rats in each group: control, torsion, torsion/detorsion and torsion/detorsion+erdosteine. Rats, except the sham operation group, were subjected to left unilateral torsion (720 degrees rotation in the clockwise direction) without including the epididymis. The experiments were finished after sham operation time for control, 120 min torsion for torsion group and 120 min torsion and 240 min detorsion for torsion/detorsion groups. Bilateral orchiectomy was performed for all groups of rats. The ipsilateral and controlateral testis were divided into two pieces to analyse biochemical parameters and to investigate the light microscopic view. Malondialdehyde level of ipsilateral testis was increased in torsion and torsion/detorsion groups in comparison with the other groups (p < 0.05). Erdosteine treatment ameliorated lipid peroxidation after torsion/detorsion in ipsilateral testis (p < 0.05). Also, xanthine oxidase activity of ipsilateral testis was increased in torsion/detorsion group in comparison with the others (p < 0.05). Nitric oxide (NO) level of ipsilateral testis was higher in all experimental groups than sham operated control group (p < 0.05). Also, NO level of torsion group was increased in comparison with detorsion groups (p < 0.05). Erdosteine treatment caused increased glutathione peroxidase activity in comparison with torsion and torsion/detorsion groups and catalase activity in comparison with the other groups in ipsilateral testis (p < 0.05). Superoxide dismutase activity of ipsilateral testis was higher in torsion/detorsion and torsion/detorsion+erdosteine groups than control and torsion groups (p < 0.05). The biochemical parameters were not affected in controlateral testis in all groups. Torsion, torsion/detorsion and torsion/detorsion+erdosteine groups showed ipsilateral testicular damage in the histological examination, but the specimens from torsion/detorsion had a significantly greater histological injury than those from the other groups (p < 0.05). Control rats showed normal seminiferous tubule morphology. Rats in torsion group had slight-to-moderate disruption of the seminiferous epithelium. Rats in torsion/detorsion group displayed moderate-to-severe disruption of the seminiferous epithelium. In all animals from torsion/detorsion+erdosteine group, the testicular tissues were affected with slight-to-moderate degenerative changes of the seminiferous epithelium. Administration of erdosteine resulted in a significantly reduced histological damage associated with torsion of the spermatic cord compared with torsion/detorsion. In all groups, the contralateral testes were histologically normal. In conclusion, the results clearly displayed that erdosteine treatment may have a protective role on testicular torsion/detorsion injury.
