- MakaleAçık Erişim2025Assessment of antioxidant, antimicrobial, antibiofilm, carbonic anhydrase and α-glucosidase inhibitory activities of Alchemilla vulgaris L. extracts
Düzgün, Azer Özad; Akar, Zeynep; Kardil, Ugur
This study was to investigate the biological properties (antioxidant, enzyme inhibitory and antimicrobial activity) of water extract (WE), 50:50% water:ethanol extract (WEE) and ethanol extract (EE) of aerial parts of Alchemilla vulgaris L. which was traditionally used to alleviate and treat many diseases. The WEE extract exhibited the highest total phenolic content (TPC), with a value of 612 µg/mL GAE, while the EE extract demonstrated the highest total flavonoid content (TFC), with a value of 0.086 µg/mL GAE. Also, the data demonstrated that the WEE extract exhibited the highest radical scavenging capacities and antioxidant activities, exhibiting 0.0056, 0.0028 mg/mL SC50 and 267, 0.081 μM TEAC, as determined by the DPPH, ABTS, FRAP and CUPRAC assays, respectively. The WEE extract was found to be an effective bovine carbonic anhydrase (BCA) and α-glucosidase inhibitor with an IC50 values of 0.0628 and 0.0535 mg/mL, respectively. The EE extract showed activity against both Gram-negative and Gram-positive bacteria used in the study. EE extract has the highest activity against Yersinia pseudotuberculosis and the lowest activity against Streptococcus pyogenes. Furthermore, the extracts significantly reduced the biofilm ability of Acinetobacter baumannii isolate, which has strong biofilm ability. EE and WE reduced the biofilm formation capacity of the strain to a weak level, while WEE extract reduced it to a moderate level. The results suggest that EE has a significant effect in combating antibiotic resistance, while WEE has high antioxidant activity and good inhibitory effect against BCA and α-glycosidase enzymes. In addition, the results revealed that solvent extracts with different solvent composition and polarity may have different effects on diverse bioactivity tests.

The growing need for eco-friendly nanomaterials has directed attention toward plant-based green synthesis as a sustainable approach for biomedical applications. This study reports the biosynthesis of silver (YAgNPs) and copper nanoparticles (YCuNPs) using Morus nigra L. leaf extract and evaluates their multifunctional properties. The nanoparticles were produced under alkaline aqueous conditions, and their crystalline nature and nanoscale morphology were confirmed by standard structural and elemental analyses. Both YAgNPs and YCuNPs exhibited enhanced antioxidant capacities compared to the crude extract. YAgNPs showed the highest ferric reducing antioxidant power (250.33 mg FeSO₄/g), while YCuNPs demonstrated superior DPPH radical scavenging activity (114.12 mg AAE/g). In antidiabetic assays, YAgNPs strongly inhibited α-amylase (IC₅₀ = 112.02 μg/mL) and α-glucosidase (IC₅₀ = 38.27 μg/mL), approaching the potency of acarbose. Antimicrobial analysis revealed distinct activity profiles: YAgNPs were highly effective in broth cultures with low MIC and MBC values (0.09–0.18 mg/mL against B. subtilis and E. coli), whereas YCuNPs achieved larger inhibition zones in diffusion assays. Checkerboard testing confirmed synergistic interactions between nanoparticles and antibiotics, with FICI values ≤0.5 against E. faecium, E. coli, and P. aeruginosa, highlighting their ability to enhance antibiotic efficacy. YAgNPs exhibited notable cytotoxic effects against HeLa and HCT116 cancer cell lines, with IC₅₀ values of 9.84 and 6.83 μg/mL, respectively. In contrast, YCuNPs displayed weaker anticancer activity. Overall, these findings demonstrate that M. nigra derived nanoparticles combine antioxidant, antidiabetic, antimicrobial, and anticancer activities with synergistic antibiotic-enhancing effects, underscoring their potential as sustainable, multifunctional therapeutic agents.

Cengiz, Sevil
Professional search and rescue (SAR) dogs, which are members of urban search and rescue teams with the help of their higher sense organs with rigorous training, are very helpful in finding buried or missing persons in disasters. In these environments where chaos and disorder prevail, it becomes inevitable for them to face very dangerous and risky situations. As a result of a disaster caused by chemical, biological, radiological and nuclear (CBRN) accidents, dangers such as chemical leaks, pathogenic microorganisms, toxins, vectors, and radioactive and nuclear materials in the environment are among the forgotten or neglected facts that SAR dogs affect as well as affect humanity. Working with little or no personal protective equipment during search and rescue interventions makes them completely vulnerable to these hazards. In the limited resources in the literature on CBRN risks and dangers that search and rescue dogs working in disaster areas may encounter, SAR dogs are exposed to many chemicals and toxic agents during these studies, as well as biological hazards such as protozoal or coliform bacterial organisms, and radiological hazards such as the risk of ingesting radioactive materials and breathing dust. their arrival has been determined. With this study, it was aimed to emphasize the dangers faced by SAR dogs and to emphasize what has been done and what should be done to reduce the possible risks. As a result, necessary precautions should be taken to prevent and/or reduce these possible exposures of SAR dogs, and further studies should be carried out for appropriate standard procedures for appropriate rehabilitation and decontamination.

Antioxidant defense mechanisms play an important role in the prevention and treatment of oxidative stress diseases in humans. In the present study, the flower and leaf parts of Linum mucronatum subsp. armenum were extracted in five different solvents. The antioxidant activities of the extracts were determine using six antioxidant activity determination assays (Iron (III) reducing/antioxidant power (FRAP), DPPH radical scavenging activity, copper (II) reducing antioxidant activity (CUPRAC), ABTS radical scavenging capacity, total flavonoid content and total phenolic content). While, the methanol extract showed the highest activity for the flower part, ethanol extracts of leaf part showed the highest antioxidant activity in the DPPH, FRAP and CUPRAC tests. The highest activity values in both flower and leaf parts was measured in acetone extract with SC50=0.287 mg/mL and (SC50=0.163 mg/mL in ABTS test, respectively. Lowest activity values of solvent extracts were measured in hexane extracts in all tests. Phenolic compounds of the plant were identified using LC-MS/MS. These phenolics are kaempferol, vanillin, protecatechuic acid, caffeic acid, p-coumaric acid, p-OH benzoic acid, salicylic acid, quercetin and rutin. The leaf and flower parts have α-glucosidase enzyme inhibitor effect. It was determined that the leaf part of the plant (IC50=4.533 mg/mL) have higher enzyme inhibition than in the flower (IC50=6.096 mg/mL). As a result, it was determined that the plant showed the biological activity. The results will contribute to the studies on the biological activity of the other plant.

Sahin, Ceyda Uzun; Kulakaç, Nurşen
The present study was conducted to determine of the factors affecting the COVID-19 knowledge levels and the status of being diagnosed with COVID-19 in healthcare employees struggling with the pandemic. This study, which was planned in cross-sectional and descriptive-correlational design. The healthcare employees working in hospitals in 6 city centers in the Black Sea Region, which has the highest number of COVID-19 cases in Turkey, were included in the study (n=385). It was determined in the present study that 33.85±9.11 (min.20, max.60), 57.9% of the participants were female, 43.4% were nurses, and 47% were infected with coronavirus. It was found that the factors that affected the coronavirus knowledge scores of health employees significantly were being diagnosed with COVID-19, presence of chronic diseases, receiving psychological support, female gender, and fighting COVID-19 in the front line. The risk factors for being infected with coronavirus disease were found to be the lack of personal protective equipment, unit worked at, and N95 mask replacement time (p<0.05). As a result of this study, ıt is recommended to provide evidence-based trainings to prevent transmission of future outbreaks and to support coping strengths since infection in healthcare employees is high.
- MakaleAçık Erişim2020Analysis of The Dynamics of The Classical Epidemic Model with Beta Distributed Random Components
Merdan, Mehmet; Bekiryazıcı, Zafer; Kesemen, Tülay
In this study, the classical epidemic model of Kermack and McKendrick is analyzed with beta distributed random components. A random analysis is done for the deterministic epidemic model by transforming the parameters and initial values of the system to random variables with beta distribution. The approximations for the expectations of the model variables are compared with the deterministic results to comment on the randomness of the cases with random parameters and random initial values. Results for some numerical characteristics of these two cases are also given to investigate the accuracy of the approximations for the expected values.
