Investigation of the enzyme-inhibitory, antibacterial, and anticancer properties of metal phthalocyanines

Topal, Meryem; Topal, Fevzi; Yilmaz, Fırat; Bulut Atalay, Esra; Koçyiğit, Ümit Muhammet; Güzel, Emre

In this study, the enzyme inhibitory, antibacterial, and anticancer activities of a phthalonitrile derivative and its metal phthalocyanines (Pcs; indium, zinc, copper, cobalt, and manganese; 1–6) were investigated. The cobalt and manganese Pcs were synthesized for the first time in this study. The inhibitory activities of the symmetric Pcs, which were expected to interact with acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) via their 4-(isopropylbenzyl)oxy substituents through host–guest interactions, were investigated. More selective inhibition of AChE than BChE was observed for these compounds. For AChE, CoPc was identified as the most potent inhibitor (IC50 = 14.81 nM), whereas for BChE, InPc was identified as the most potent inhibitor (IC50 = 56.35 nM). The Ki values indicated that most compounds exhibited competitive inhibition; copper phthalocyanine (CuPc) showed particularly strong inhibition against AChE (Ki = 3.08 nM ± 1.12), whereas the lowest Ki value against BChE was observed for MnPc (Ki = 25.98 ± 1.97 nM). Most compounds exhibited competitive inhibition; however, CuPc showed competitive inhibition toward AChE but a noncompetitive inhibition pattern toward BChE. Dual inhibition of AChE and BChE by these compounds may be promising for addressing cholinergic deficits associated with Alzheimer’s disease. In addition, the acceptability of compounds 1–6 with respect to pharmacological drug-likeness criteria was assessed based on predicted absorption, distribution, metabolism, excretion (ADME) outcomes. In antibacterial tests, varying levels of inhibition were observed against selected bacterial strains. In anticancer assays, all compounds exhibited high cytotoxicity against the MCF-7 breast cancer cell line. Higher antiproliferative activity was observed for CuPc than for the other compounds. Morphological changes were induced in cancer cells by CuPc and MnPc. Overall, these compounds may have potential as enzyme inhibitors and as antibacterial and anticancer agents.

Arctic governance through discourse: Arctic and Arctic council observer states

    Limon, Onur; Gürdal Limon, Elif

This article examines how Arctic governance is discursively constituted through national Arctic policy and strategy documents and how these discursive constructions differentiate governance roles between Arctic states and Arctic Council observer states. Rather than treating national strategies as neutral policy instruments, the study conceptualizes them as political texts that articulate authority, responsibility, and legitimacy within an asymmetrical governance system. Methodologically, the article employs qualitative discourse analysis of the most recent official Arctic strategies of eight Arctic states and twelve observer states. Using MAXQDA (version 24), the analysis is organized around six thematic domains: security policy; climate change and environment; international cooperation; resource management; Indigenous peoples and rights; and economic development and sustainability. The findings reveal a high degree of discursive convergence across both actor groups, particularly around climate change, sustainability, environmental protection, and cooperation. However, this convergence does not indicate substantive policy alignment or egalitarian governance. Arctic states frame governance through narratives of sovereignty, stewardship, and regulatory responsibility, while observer states construct legitimacy through scientific contribution, multilateralism, and normative alignment. The article argues that discursive convergence can function as a kind of governance infrastructure that enables participation while tending to stabilize asymmetrical authority.

Evaluation of antioxidant and antiproliferative activities of shogaol and its modulatory effects on key metabolic enzymes

    Yapıcı, İsmail; Gülçin, İlhami

Phenolic compounds are versatile bioactive molecules, particularly in food applications. In this study, the antioxidant capacity of Shogaol, a member of the phenolic class, was evaluated using several methods. Additionally, its inhibitory effects on human carbonic anhydrase I and II (hCA I and II), acetylcholinesterase (AChE), butyrylcholinesterase (BChE), α-glycosidase, and α-amylase enzymes, as well as its antiproliferative effects on A-549 lung cancer, HT-29 colon cancer, MCF-7 and MDA-MB-453 breast cancer cell lines, and the 3T3-L1 mouse fibroblast normal cell line, were investigated. To determine the antioxidant capacity of Shogaol, DPPH and ABTS radical scavenging assays, as well as Cu²⁺, Fe³⁺, and FRAP reduction methods, were employed. Its inhibitory effects on hCA I and II, AChE, BChE, α-glycosidase, and α-amylase enzymes were examined. Furthermore, cell counting was performed using the TBE method, and cell viability was measured with the XTT assay to assess the antiproliferative effects of Shogaol on A-549, HT-29, MCF-7, MDA-MB-453, and 3T3-L1 cell lines. Shogaol demonstrated higher antioxidant capacity than BHA, BHT, and Trolox in the FRAP assay (60 µg/mL), higher than BHT in the DPPH scavenging assay, and higher than Trolox in the ABTS, Cu²⁺, and Fe³⁺ reduction assays. Shogaol also exhibited significant inhibitory effects against hCA I, hCA II, AChE, BChE, and α-glycosidase, with IC₅₀ values of 75.91, 47.04, 37.44, 24.11, and 35.72 nM, respectively. Moreover, Shogaol showed a more potent antiproliferative effect than the standard chemotherapeutic drug Carboplatin in A-549 lung cancer and HT-29 colon cancer cell lines, and greater efficacy than Docetaxel in the MCF-7 breast cancer cell line.

Efficacy and safety of doxorubicin-ifosfamide versus gemcitabine-docetaxel as first-line therapy in advanced nonuterine leiomyosarcoma: A multicenter real-world study

Tunbekici, Salih; Sahin, Gokhan; Yuksel, Haydar Cagatay; Acar, Caner; Akin, Imge; Aslanhan, Hasan; Isci, Mert; Kabaoglu, Ceren; Bal, Hamit; Sakin, Abdullah

Nonuterine leiomyosarcoma (NU-LMS) is a rare and biologically distinct subtype of soft tissue sarcoma (STS) with clinical behavior differing from that of uterine leiomyosarcoma. In advanced or metastatic STS, immunotherapy has demonstrated only modest activity, and chemotherapy remains the cornerstone of treatment. However, there is a persistent unmet need for more effective and better-tolerated options in this population. This multicenter, retrospective study included 150 patients with histologically confirmed metastatic NU-LMS who received first-line doxorubicin-ifosfamide (AI) or gemcitabine-docetaxel (GD) at 26 oncology centers in Turkey between 2010 and 2024. The primary endpoint was overall survival (OS). Survival was analyzed using Kaplan-Meier estimates and Cox proportional hazards models. Of 150 patients, 79 received AI and 71 received GD. Median OS for the entire cohort was 21.0 months. Median OS was 22.0 months with AI and 19.1 months with GD (p = .18), median progression-free survival was 7.3 versus 6.1 months (p = .66), and overall response rate was 25.3% versus 29.6% (p = .58), indicating no statistically significant differences in efficacy between the two regimens. Hematologic toxicities were less frequent with GD. AI and GD demonstrated comparable efficacy as first-line regimens for advanced or metastatic NU-LMS, with fewer hematologic toxicities observed for GD. Tumor location may have prognostic implications. These findings support individualized treatment selection and highlight the need for prospective studies in this rare and understudied sarcoma subtype, while the recent LMS-04 trial underscores the importance of future comparisons with doxorubicin-trabectedin to further refine first-line strategies in LMS.

Preferences for mechanical ventilation modes among intensivists in Türkiye: a nationwide point-prevalence study

Yildirim, Suleyman; Kilinçözgün, Nurhayat; Alpdogan, Ozcan; Uçar, Huseyin; Taskiran, Imren; Ata, Adnan; Özcan, Ozhan; Öncü, Kivanc; Acarbay, Saba Mukaddes; Güner, Temel

Background/aim: Invasive mechanical ventilation (IMV) is a fundamental intervention for patients with respiratory failure in intensive care units (ICUs). This nationwide, multicenter point-prevalence study aimed to describe current mechanical ventilation mode preferences (conventional, adaptive, and biphasic) in Turkish ICUs and to report associated clinical outcomes descriptively, without assessing causal relationships. Materials and methods: A nationwide, multicenter point-prevalence study was conducted on 17 April 2024 and included adult patients (>= 18 years) who had been receiving IMV for more than 24 h. Data on patient demographics, ventilation mode distribution, ventilatory parameters, and descriptive clinical outcomes on day 28 (weaning status, tracheostomy, and mortality) were recorded without comparative outcome analysis. Results: A total of 426 patients were included. Conventional modes were used in 84.5% of patients, adaptive modes in 10.6%, and biphasic modes in 4.9%. Synchronized intermittent mandatory ventilation (SIMV) was the most commonly used conventional mode. The primary indication for IMV was acute respiratory failure (61%), with pneumonia being the leading cause. Among the 350 orotracheally intubated patients, 25.6% were in the weaning phase on the study day. A total of 59 (16.9%) patients were extubated, 150 (42.9%) underwent tracheostomy, and 64 (18.2%) remained intubated on day 28. Overall, 185 (43.4%) patients died during their ICU stay, 152 (35.7%) remained in the ICU, and 89 (20.9%) were successfully discharged from the ICU. Conclusion: Conventional ventilation modes, particularly SIMV, were more commonly used in Turkish intensive care units (ICUs), whereas adaptive modes were less frequently applied. These patterns may reflect factors such as clinician familiarity, institutional practices, and equipment availability rather than definitive preferences. Although the impact of ventilation modes on clinical outcomes was not comparatively evaluated in this study, the choice of ventilation mode may still influence patient outcomes. Therefore, further prospective and comparative studies are warranted to better elucidate this relationship.

Ethical Principles and Frameworks in Dementia Care: A Scoping Review

Serrat, Rodrigo; Lauridsen, Sigurd; Bobrowicz-Campos, Elzbieta; Brandao, Tania; Brites, Rute; Cannella, Valentina; Celdrán, Montserrat; Chacur-Kiss, Karima; Cin, Aynur; Fabà, Josep

Providing ethical care for people living with dementia is essential for safeguarding their rights, dignity, and well-being. However, ethical decision-making in dementia care is complex, often involving tensions between competing moral claims such as autonomy, beneficence, nonmaleficence, justice, and non-discrimination. This scoping review aimed to map and describe the ethical principles, related normative considerations, and ethical frameworks discussed in the scientific literature on dementia care, and to identify how these have been conceptualized. Following the methodological guidance of Arksey and O'Malley (2005), Levac et al. (2010), and Peters et al. (2021, 2022), a comprehensive search across five databases (Web of Science, Scopus, CINAHL, Medline/PubMed, and PsycInfo) identified peer-reviewed papers published in English. In total, 388 papers published between 1981 and 2024 were included. The results revealed a wide range of ethical principles and related normative considerations frequently cited in the literature, although they were typically discussed in an isolated way without reference to other principles or broader ethical frameworks. In contrast, ethical frameworks were mentioned far less often, and when they were, they mostly reflected traditional approaches such as principlism. Four gaps were identified: the plurality of ethical frameworks and limited conceptual integration underpinning dementia care research and practice; insufficient attention to ethnic and socioeconomic diversity among people living with dementia; limited consideration of diverse care settings and geographical regions; and the limited inclusion of people living with dementia themselves in the studies reviewed. These findings underscore the need for conceptually integrative, inclusive, and context-sensitive approaches to dementia care ethics that can better inform both ethical reflection and care practices.

Comparative study of magnetic and electric quadrupole moments in boron (5B) isotopes

Ali, Ahmed H.; Tfiha, Amir D.; Zakaly, Hesham M. H.; Jarallah, Naz T.; Issa, Shams A. M.; Abdulqader, Abdulqader S.; Çelik, Necati; Bhuyan, M.

The magnetic moments and electric quadrupole moments of the B-5 isotopes were calculated using different nuclear interactions and sets of effective charges. Instead of relying on core polarization, the effective charges were adopted through a microscopic theoretical approach. The one-body density matrix (OBDM) elements for these isotopes were calculated using the NuShellX@MSU code. The calculations were carried out within the shell model framework, using both the p- and psd-model spaces (MS). These model spaces were selected for nuclei with proton number Z = 5 and neutron number N > 3. The theoretical results for magnetic and quadrupole moments were compared with available experimental data. Theoretical magnetic moments of the B-8,B-10,B-11,B-12,B-13 isotopes showed good agreement with experiment when using the p-MS with the pwt interaction, and the psd-MS with the psdmwk interaction. Similarly, the theoretical electric quadrupole moments reasonably well with the experimental values when using the p-MS with the pkuo interaction, and the psd-MS with the psdmwk interaction.

Inspiratory muscle training enhances jumping power and shooting performance in elite air pistol athletes

Aydin, Ahmet Serhat; Yilmaz, Coskun; Söyler, Mehmet; Güney, Gamze; Eyüboglu, Ender; Karakulak, Izzet; Ceylan, Tulay; Altug, Tolga; Burac, Daniela Giconda; Sahinler, Yunus

Background Inspiratory muscle training (IMT) has been shown to enhance respiratory efficiency, postural stabilization, and neuromuscular coordination in athletic populations. However, its effects in precision sports such as air pistol shooting remain insufficiently explored. This study investigated the impact of a 4-week IMT program on respiratory muscle strength, explosive lower-limb performance, reaction time, and shooting accuracy in competitive air pistol athletes. Methods Twenty trained male air pistol athletes (age 18-35 years) were randomly assigned to an IMT group (n = 10) or a control group (n = 10). The IMT group performed supervised inspiratory muscle training twice daily (30 breaths/session), six days per week, for four weeks, using a threshold-loading device set initially at 40% of maximal inspiratory pressure (MIP), with progressive overload applied weekly. Both groups maintained their regular shooting training routines. Pre- and post-intervention assessments included countermovement jump (CMJ) and squat jump (SJ) performance (OptoJump), visual and auditory reaction time (Cognitech), and shooting performance evaluated via the SCATT system. A two-way repeated-measures ANOVA (group & times; time) was used for statistical analysis. Results Significant group & times; time interactions were observed for CMJ height (p < 0.001, eta & sup2;p = 0.618), CMJ power (p = 0.007, eta & sup2;p = 0.345), SJ height (p = 0.050, eta & sup2;p = 0.184), and SJ power (p = 0.050, eta & sup2;p = 0.181), indicating meaningful improvements in explosive performance in the IMT group. Shooting accuracy demonstrated a large and statistically significant interaction effect (p < 0.001, eta & sup2;p = 0.532), with substantial improvement observed only in the IMT group. Reaction time variables showed favourable but non-significant trends (p > 0.05). Conclusion A short-term IMT intervention significantly enhanced lower-limb explosive performance and shooting accuracy in competitive air pistol athletes without altering overall training load. These findings suggest that IMT may represent an effective, low-cost adjunct strategy to improve neuromuscular stability and fine-motor precision in precision-sport athletes.

A multi-evidence computational and experimental strategy for Silene nerimaniae: combining phytochemical characterization, bioactivity assessment, and ADEFRank-HCT-116-based metabolite prioritization

Erharman, Ahmet; Yagi, Sakina; Uba, Abdullahi Ibrahim; Nikolic, Filip; Gasic, Uros; Acar, Mikail; Dogan, Nurettin; Koyuncu, Ismail; Yapici, Ismail; Gulcin, Ilhami

This study presents the first comprehensive evaluation of the phytochemical composition, antioxidant properties, enzyme inhibitory effects, and cytotoxic potential of Silene nerimaniae G.E.Gen & ccedil;, Kandemir & & Idot;.Gen & ccedil; (family Caryophyllaceae). Ethyl acetate, acetone, 70% acetone, and aqueous extracts were obtained from the aerial parts of the plant through maceration. Among the tested extracts, the acetone extract contained the highest total phenolic (24.37 mg GAE per g) and flavonoid (24.64 mg RE per g) contents. LC-MS analysis identified 52 bioactive constituents, including sugars, organic acids, phenolic acids, flavonoids, and fatty acids. The aqueous extract demonstrated the strongest radical scavenging activity against DPPH (23.02 mg TE per g) and ABTS (55.76 mg TE per g), as well as the highest Fe3+ reducing power (37.53 mg TE per g). In combination with the acetone extract, it also exhibited notable Cu2+ reducing activity (47.78 mg TE per g). Regarding enzyme inhibition, the ethyl acetate, acetone, and hydroacetone extracts displayed the strongest inhibitory activities against acetylcholinesterase (1.87 mg GALA per g), butyrylcholinesterase (2.92 mg GALA per g), and tyrosinase (60.44 mg KAE per g), respectively. All three extracts also inhibited alpha-amylase activity within the range of 0.73-0.77 mmol ACAE per g (p >= 0.05). In addition, the hydroacetone extract showed considerable inhibition of human carbonic anhydrase isoenzymes I (IC50 = 38.78 mu g mL-1) and II (IC50 = 19.59 mu g mL-1). The ethyl acetate extract exhibited the strongest cytotoxic effect against the colorectal cancer cell line HCT-116, with an IC50 value of 0.639 mu g mL-1 and a selectivity index of 1.7. To contextualize this HCT-116-associated activity at the metabolite level, an ADEFRank-HCT116 (abundance-domain-evidence fusion ranking) workflow was applied by integrating EtOAc abundance, HCT-116-oriented QSAR evidence, applicability-domain confidence, and structure-assignment reliability. This analysis prioritized diosmetin, luteolin, eriodyctiol, acacetin, and isorhamnetin as core candidates for structure-based follow-up. Molecular docking screening predicted favorable docking profiles for the prioritized candidates, while 100 ns molecular dynamics simulations were used to assess the dynamic stability of the representative luteolin-PI3K alpha, diosmetin-AKT1, and isorhamnetin-GSK-3 beta complexes. Taken together, these results indicate that S. nerimaniae may represent a valuable source of active compounds for various functional nutraceutical, cosmetic and pharmaceutical uses.

Comparison of plyometric repeated sprint and plyometric aerobic training on physical performance in youth soccer players

Selmi, Mohamed Amin; Hammami, Raouf; Ceylan, Halil Ibrahim; Duncan, Michael; Sassi, Radhouane Haj; Morgans, Ryland; Bayrakdaroglu, Serdar; Bragazzi, Nicola Luigi

High-intensity interval training (HIIT) and repeated-sprint sets (RSS) are established conditioning methods in soccer, yet their integration with plyometric training among highly trained youth soccer players remains underexplored. This study investigated the effects of an 8-week program combining plyometric training with either RSS (PLYO-RSS) or HIIT (PLYO-HIIT) on physical fitness in young soccer players. Twenty-nine male players (14-15 years) were randomly assigned to PLYO-RSS (n = 14) or PLYO-HIIT (n = 15). Both interventions lasted 20 minutes per session and replaced part of regular soccer training. Physical fitness assessments included countermovement jump (CMJ), 15-s repeated vertical jumps (15''RJ), five-jump test (5JT), 10 m and 30 m sprint, maximal aerobic speed (MAS), VO2max, and repeated sprint performance (total sprint time, best sprint, fatigue index). Physiological markers (lactate, rating of perceived exertion, and HRmax) were also monitored. Significant improvements over time were observed in muscle power, sprint speed, aerobic capacity, and repeated-sprint ability (p <= 0.001; eta & sup2;p = 0.339-0.489). However, no main effects or group & times; time interaction effects were observed, indicating no evidence of differential responses between the two training modes. In conclusion, integrating plyometric training with either RSS or HIIT effectively enhances physical fitness in highly trained youth soccer players, offering flexible options for training prescription.

Chemical profiling, biological evaluation, and QSAR-informed metabolite prioritization of Hypericum ekerii: a first report on antioxidant, enzyme inhibitory, and cytotoxic activities

Zengin, Gokhan; Yagi, Sakina; Cetiz, Mehmet Veysi; Uba, Abdullahi Ibrahim; Nikolic, Filip; Gasic, Uros; Balos, Mehmet Maruf; Erharman, Ahmet; Dogan, Nurettin; Koyuncu, Ismail

Hypericum ekerii Y & uuml;ce and Ayta & ccedil;, first described in 2017, remains poorly characterized in terms of its phytochemical composition and biological properties. This study investigated aerial-part extracts of H. ekerii prepared with ethyl acetate, acetone, 70% acetone-water, and water. The hydroacetone extract showed the highest total phenolic and flavonoid contents. LC/MS analysis enabled the annotation of 99 constituents, including phenolic acids, flavonoids, xanthones, anthraquinones, and acylphloroglucinols. The hydroacetone extract exhibited the strongest radical-scavenging and reducing activities, whereas acetone and ethyl acetate extracts showed the highest metal-chelating capacity. In enzyme assays, hydroacetone was the most active extract against acetylcholinesterase, tyrosinase, and hCA II, while ethyl acetate showed greater inhibition of butyrylcholinesterase and hCA I. The aqueous extract was the most active against alpha-glucosidase, and hydroacetone and ethyl acetate showed comparable alpha-amylase inhibition. Cytotoxicity remained limited overall; the lowest IC50 value was observed for the ethyl acetate extract against HCT-116 cells (95.09 & micro;g mL-1), with weak selectivity (SI = 1.2). A QSAR-informed prioritization framework identified myricetin as the leading cross-layer candidate, p-coumaric acid as a target-specific priority metabolite, and ellagic acid, quercetin, and caffeic acid as antioxidant-priority candidates. Molecular docking supported favorable binding profiles for selected metabolites, while molecular dynamics simulations were used to assess the stability of representative complexes. Overall, H. ekerii appears to be a chemically diverse species with notable antioxidant and enzyme-inhibitory potential, providing a focused basis for future compound-level validation.

Geochemical signature, detrital zircon U-Pb, and mica Ar-Ar age systematics from the Karadere Basement Unit of the İstanbul-Zonguldak Terrane (NW, Türkiye): evidence for an Avalonian-Cadomian active continental margin

Çimen, Okay; Gücer, Mehmet Ali; Göncüoglu, Mehmet Cemal; Akal, Cuneyt; Arslan, Mehmet; Simonetti, Antonio; Karaoglan, Fatih

The & Idot;stanbul-Zonguldak Terrane is located in northwest Anatolia and includes several outcrops with pre-Cambrian basement units. The Karadere Basement Unit represents the easternmost basement of this terrane and is composed of paragneiss, quartzite, and orthogneiss. The paragneiss and quartzite are cut by orthogneisses and disconformably overlain by Lower Ordovician to Middle Devonian cover units. Its pre-Cambrian to Palaeozoic tectonic evolution remains poorly understood, and therefore, this study reports whole-rock trace element signatures, detrital zircon U-Pb, and muscovite Ar-Ar age data for samples of paragneiss, quartzite, amphibolite, and orthogneiss. The whole-rock geochemical data indicate that the protoliths of the paragneiss and quartzite samples investigated here could have been derived from arc-related felsic magmatic units located in an active continental margin. The protoliths of amphibolites and orthogneiss samples are gabbro and granite, respectively, and their geochemical signatures are akin to those from continental arc igneous suites. The detrital zircon U-Pb data are characterized by predominantly Mesoproterozoic (84%) and lesser amount of Palaeoproterozoic and Neoproterozoic ages and suggest an early Tonian (youngest peak age of 915 +/- 18 Ma) maximum depositional age. Moreover, zircon U-Pb dating analyses from an orthogneiss sample yielded an age of 603 +/- 2 Ma, indicating the occurrence of mid-Ediacaran arc magmatism. This represents one of the oldest Ediacaran ages obtained from the metamagmatic basement rocks of the & Idot;stanbul-Zonguldak Terrane. Ar-Ar dating of muscovite from ortho- and paragneiss samples yielded ages of 533 +/- 25 Ma and 530 +/- 9 Ma, respectively, which are interpreted as early Cambrian cooling ages. Overall, the combined results reported here reveal that the Karadere Basement Unit is markedly different from NE African basement units but displays similarities to the basement rocks of both Avalonian and Amazonian terranes. It may have been formed and metamorphosed in an Avalonian-Cadomian active continental margin during the mid- to late Neoproterozoic and early Cambrian.

Long-term decline of Chernobyl-derived 137Cs in mosses and lichens from the Eastern Black Sea region of Türkiye

Çevik, Ugur; Çelik, Necati; Uzun, Kubra K.; Çelik, Ahmet; Koz, Bahadir; Özen, Songul A.; Yesilkanat, Cafer M.; Hacisalihoglu, Aylin; Özkalayci, Fatih; Kaya, Selim

This study presents the third consecutive assessment of 13 Cs activity concentrations in the same 50 moss (7 species) and lichen (2 species) sampling points in Ordu province, T & uuml;rkiye, one of the most heavily contaminated areas following the Chernobyl accident. Samples were collected from exactly the same locations used in the 1997 and 2007 studies and analyzed by high-resolution gamma spectrometry. In 2025, detectable 13 Cs was found in 14 of 30 moss sites (47%) and 3 of 20 lichen sites (15%). For samples below the minimum detectable activity (MDA), a value of MDA/2 (0.93 Bq kg-1) was substituted for lichens, while moss samples below the MDA were excluded from regression. The ecological half-life (Teco) for the 2007-2025 period was 4.38 +/- 1.05 years for mosses and 4.18 +/- 0.61 years for lichens. For mosses, this is nearly identical to the 1997-2007 mean (4.4 years), while for lichens it decreased from 5.6 to 4.18 years, suggesting a slight acceleration in environmental removal. All Teco values remain far shorter than the physical half-life of 13 Cs (30 years), confirming that environmental removal processes dominate over radioactive decay. Residual 13 Cs is concentrated in higher-altitude areas, reflecting the original deposition pattern and slower removal in cooler, more humid microenvironments. Nearly 39 years after the accident, the radiological risk posed by residual trace levels is negligible. This 28-year dataset provides one of the longest continuous records of 13 Cs in epiphytic bioindicators and serves as a valuable baseline for future contamination events.

Mono- and Bis-Ureido Sulfapyridines as Nanomolar Dual Cholinesterase Inhibitors: Enzyme Kinetics, Molecular Docking, and ADME/Tox Insights

Yapar, Gonul; Akocak, Suleyman; Lolak, Nebih; Topal, Meryem; Türkes, Cuneyt; Topal, Fevzi; Durgun, Mustafa; Koçyigit, Umit Muhammet; Isik, Mesut; Beydemir, Sukru

A series of mono- and bis-ureido sulfapyridine derivatives (SP-1-SP-10) were synthesized and evaluated as dual cholinesterase inhibitors targeting acetylcholinesterase (AChE) and butyrylcholinesterase (BChE). All compounds exhibited nanomolar inhibition. The most potent AChE inhibitors were SP-8 (K-i = 5.25 +/- 0.73 nM), SP-9 (5.09 +/- 0.44 nM), and SP-10 (5.74 +/- 0.55 nM), significantly outperforming tacrine (K-i = 105.03 +/- 8.85 nM) under identical conditions. For BChE, SP-9 showed the strongest inhibition (K-i = 26.38 +/- 2.07 nM) compared with tacrine (172.03 +/- 32.42 nM). Kinetic analyses revealed competitive inhibition toward AChE for all derivatives, whereas BChE inhibition displayed both competitive and noncompetitive patterns depending on structural features. Glide XP docking using AChE (PDB: 7XN1) and BChE (PDB: 4BDS), complemented by MM-GBSA binding free energy calculations, supported favorable ligand accommodation within the catalytic pockets and highlighted the importance of aromatic and hydrophobic interactions. In silico ADME/Tox analysis revealed a potency-drug-likeness trade-off: mono-ureido derivatives complied with Lipinski's rule of five and showed high predicted oral absorption, whereas bis-ureido derivatives, despite superior enzymatic potency, exhibited multiple drug-likeness violations. These findings identify the ureido-sulfapyridine scaffold as a potent enzymatic dual cholinesterase inhibitory framework, warranting further structural optimization to improve pharmacokinetic properties.

Offside calls are integral to maintaining fairness and competitive integrity in football, yet current officiating processes are constrained by limitations in accuracy despite technological advancements. This investigation developed and validated an automated offside detection system using YOLOv8 object detection architecture to address documented limitations of Video Assistant Referee (VAR) systems in offside decision accuracy. Supervised learning validation employed 400 annotated instances from professional match footage. The YOLOv8 medium variant was configured for four-class detection, with a focus on extreme player positions, reducing computational complexity by approximately 70% relative to comprehensive player tracking while accepting accuracy trade-offs in marginal scenarios. The system was optimized for computational efficiency through highly accurate tracking of player positions. Data were partitioned into 40% (n = 160) for training and 60% (n = 240) for testing, with 5-fold cross-validation. The automated system achieved robust performance metrics across 240 test instances: overall accuracy of 83.0%, precision of 85.0%, recall of 87.0%, and F1-score of 86.0%. Confusion matrix analysis revealed true positives (n = 130), true negatives (n = 70), false positives (n = 22), and false negatives (n = 18). Individual case analysis demonstrated 80% agreement with ground truth classifications. System performance significantly exceeded the random classification baseline (p < 0.001). The strategic extreme player tracking approach achieved substantial computational efficiency while maintaining decision accuracy across diverse match scenarios. A YOLOv8-based offside detection system demonstrates proof-of-concept feasibility for decision-support applications in football officiating. Performance (83% accuracy, 95% CI: 79-87%) on 240 test instances remains below professional benchmarks (> 93%), with single-camera constraints and limited dataset size restricting generalizability to proof-of-concept validation rather than operational deployment.

Konca, Erkan; Yilmaz, Coskun; Bayrakdaroglu, Serdar; Ceylan, Halil Ibrahim; Arslan, Ayla; Ocak, Hakan; Karakulak, Izzet; Sari, Rifat; Uzun, Recep Nur; Soylu, Hakan Huseyin

Background: Age-related declines in respiratory muscle strength and ventilatory efficiency can impair functional capacity and metabolic health in older adults. Inspiratory muscle training (IMT) has been proposed as a practical intervention to counteract these changes, yet its systemic effects remain unclear. This study aimed to examine the effects of short-term IMT on functional capacity, diaphragm thickness, and liver tissue characteristics in healthy elderly men. Methods: Thirty community-dwelling men aged 60-80 years were randomly assigned to an IMT or control group. The IMT group performed four weeks of breathing exercises using a POWERbreathe (R) device at 40% of maximal inspiratory pressure, with a weekly 10% increase in pressure. Pre- and post-intervention assessments included the six-minute walk test (6MWT), diaphragm thickness and liver density via computed tomography, and quality of life (QoL; SF-12). Results: Four weeks of inspiratory muscle training significantly improved diaphragm thickness (11.7%), fatty liver density (FLD) (+16.7%), and six-minute walk performance (+5.3%), with large time x group effects favoring the IMT group. While the physical quality of life showed modest, comparable improvements, mental health outcomes demonstrated a moderate, time-dependent improvement without a significant group-by-time interaction. Conclusions: Short-term IMT improved diaphragmatic function and functional capacity in older men and was associated with favorable changes in a liver-related biomarker; however, given that only a single liver-related metric was assessed, these findings should not be interpreted as evidence of overall improvements in liver health.

The mediating role of mental fatigue awareness in the relationship between perceived stress and psychological well-being among combat athletes

Yilmaz, Taner; Öz, Faik; Gül, Oguzhan; Dal, Suzan; Kara, Tayfun; Akkoç, Orkun; Küçük, Hamza; Fort, Patricia Roxana; Çitozi, Robert; Kadi, Mustafa Nurullah

Background Combat athletes face unique psychological demands, and stress may influence both their cognitive experiences and psychological well-being. This study aimed to examine the mediating role of mental fatigue awareness in the relationship between perceived stress and psychological well-being among combat athletes, thereby contributing to a clearer understanding of a potential cognitive mechanism linking stress to well-being in this population. Methods A total of 303 combat athletes voluntarily participated. Participants completed the Perceived Stress Scale, Mental Fatigue Awareness Scale, and Psychological Well-being Scale. Statistical analyses assessed the direct effects of perceived stress on psychological well-being, the effect of stress on mental fatigue awareness, and the potential mediating role of mental fatigue awareness. Results Perceived stress significantly increased mental fatigue awareness (b = 0.732, SE = 0.064, beta = 0.549, p = 0.001) and directly decreased psychological well-being (b = - 0.488, SE = 0.166, beta = - 0.191, p = 0.004). Mental fatigue awareness negatively affected psychological well-being (b = - 0.374, SE = 0.124, beta = - 0.195, p = 0.003) and partially mediated the relationship between stress and well-being (indirect effect: b = - 0.274, SE = 0.115, beta = - 0.107, p = 0.003; 95% CI = - 0.504 to - 0.049).Conclusion Perceived stress was associated with psychological well-being both directly and indirectly through increased mental fatigue awareness. These findings suggest that mental fatigue awareness may represent a relevant cognitive pathway linking stress perceptions to well-being. Accordingly, stress management and strategies aimed at regulating mental fatigue may be considered as potentially beneficial components in efforts to support psychological well-being among combat athletes.

The growing interest in natural products, particularly as food additives and pharmaceutical raw materials, is making research into discovering new sources especially important. Erodium cedrorum subsp. salmoneum (family Geraniaceae) is used traditionally to cure many ailments like diabetes and gastro-intestinal diseases. Within this background, the current study was designed to investigate, for the first time, the chemical composition, antioxidant and enzyme inhibitory poroperties of different extracts from the aerial parts and roots of E. cedrorum. Results revealed that the aqueous extract of the aerial parts accumulated the highest total phenolic (85.35 mg GAE/g) and flavonoids (28.65 mg RE/g) contents. A total of 61 and 45 compounds, belonging mainly to phenolic acids and flavonoids, were tentatively identified in the aerial parts and roots extracts respectively. The aqueous extract from the aerial parts exerted the highest antiradical, ions reducing and chelation properties. The EtOH and 70% EtOH extracts of the two organs recorded the best anti-acetylcholinesterase activity (2.18 -2.41 mg GALAE/g; p >= 0.05) while those of the roots recorded respectively the best anti-butyrylcholinesterase and antityrosinase activities. The most potent antibacterial activity was exhibited by the 70% EtOH extracts from aerial parts and the pure EtOH extracts from roots. In contrast, ethyl acetate extracts from both plant parts showed only marginal activity. Among the tested strains, the highest susceptibility was observed against C. sporogenes ATCC 19404, C. bifermentans ATCC 638, S. aureus ATCC 6538, S. aureus ATCC 43300 (MRSA), C. diphtheriae PCM 506, and H. pylori ATCC 43504. The extracts were also examined for their anti-cancer potential on the HeLa, AGS and SKOV-3 cell lines, as well as a non-cancerous cell line. The ethyl acetate extract of the roots exhibited the most potent anti-cancer activity, with the lowest IC50 values. In addition, selected top-ranked complexes against AChE, alpha-amylase, representative bacterial and oncogenic targets were further evaluated by molecular docking and 100 ns molecular dynamics (MD) simulations, indicating overall conformational stability and persistent binding. In conclusion, these results position E. cedrorum as a valuable natural source of antioxidant and enzyme-inhibitory agents, meriting further exploration for drug development purposes.
