- MakaleAçık Erişim2016Scopus SJRQ3Role of oxidative stress in the pathophysiology of Toxoplasma gondii infection
Dincel, Gungor Cagdas; Atmaca, Hasan Tarik
Oxidative stress (OS) plays an essential role in the pathogenesis of common neurodegenerative diseases. We have previously shown that Toxoplasma gondii (T. gondii) induces high nitric oxide (NO) production, glial activation, and apoptosis that altogether cause severe neuropathology in toxoplasma encephalitis (TE). The objective of this study was to investigate the cytotoxic effect of OS and to identify a correlation between the causes of T. gondii induced neuropathology. Expression levels of glutathione reductase (GR), Cu/Zn superoxide dismutase (SOD1), neuron specific enolase (NSE), and 8-hydroxy-2'-deoxyguanosine (8-OHdG) were investigated. Results of the study revealed that the levels of GR (P <0.005) and NSE (P <0.001) expression in the brain tissue markedly increased while SOD1 activity decreased (P <0.001) in the infected group compared to the non-infected group. In addition, intense staining for 8-OHdG (P <0.05) was observed both in the nucleus and the cytoplasm of neurons and glial cells that underwent OS. These results were reasonable to suggest that T. gondii-mediated OS might play a pivotal role and a different type of role in the mechanism of neurodegeneration/neuropathology in the process of TE. The results also clearly indicated that increased levels of NO and apoptosis might contribute to OS-related pathogenesis of TE. As a result, OS and expression of NSE might give an idea of the disease progress and may have a critical diagnostic significance for patients with T. gondii infection.

The aim of this study was to evaluate the oxidant-antioxidant balance in patients with abdominal aortic aneurysms (AAA). Forty-two consecutive patients with AAA and 46 control subjects were included. Total oxidant status (TOS) and total antioxidant status (TAS) levels were measured and the oxidative stress index (OSI) value determined. Serum TOS and OSI values in patients with AAA were higher than those in the controls (p<0.001, p<0.001, respectively). There was a positive correlation between abdominal aortic diameters, serum TOS levels (r=0.592, p<0.001) and OSI values (r=0.598, p<0.001). A cut-off value of 17.68 μmol H2O2 equivalent/L for TOS was associated with 86% sensitivity and 83% specificity and a cut-off value of 1.77 for OSI was associated with 86% sensitivity and 81% specificity for predicting AAA. Systemic oxidative imbalance develops in patients with AAA, particularly as a result of an increase in TOS.

- MakaleAçık Erişim2016Scopus SJRQ2Investigation of the inhibitory properties of some phenolic standards and bee products against human carbonic anhydrase I and II
Aygul, Imdat; Yaylaci Karahalil, Fatma; Supuran, Claudiu T.
Polyphenols are important secondary products of plants with the potential to inhibit carbonic anhydrases. The aim of this study was to investigate the inhibition effects of various phenolic standards, honey, propolis, and pollen species on human carbonic anhydrase I and II. The inhibition values (IC50) of the phenolics (gallic acid, protocatechuic acid, quercetin, catechin, tannic acid, and chrysin) ranged from 0.009 to 0.32 μg/mL, tannic acid emerging as the best inhibitor. The inhibition values of three different types of honey, heather, rhododendron, and chestnut ranged between 2.32 and 25.10 μg/mL, the chestnut honeys exhibiting the best inhibition. The ethanolic extracts of pollen and propolis exhibited good inhibitory properties, with IC50 values between 0.486 and 3.320 μg/mL. In order to evaluate the phenolic composition of bee products, phenolic profiles and total phenolic contents (TFC) were also measured. The inhibition ranking among the natural products studied was phenolic standards > propolis > pollen > honeys, and inhibition was related to TFC.

Budak, S.; Kaplan, M.
In this study, four different kinds of marble cutting matrices with a base composition of Fe-Cu-Sn and with variable amounts of Co powder were produced. For compaction, hot sintering process was carried out at 850°C under 35 MPa for 4 min. The mechanical and microstructural properties of the matrix materials were determined using SEM with EDS, three-point bending test, density and hardness measurement techniques. The addition of Co powders to the matrix and the subsequent hot sintering process affected the mechanical and microstructural properties of the cutting tool, such as hardness, total elongation, % porosity and phase distribution.

Fidan, M. Said; Yaşar, Ş. Şadiye; Yaşar, Mehmet; Atar, Musa; Alkan, Elif
Wood is an irreplaceable material used as decorative and structural element both indoors and outdoors. When used as a structural material, wood has several superior characteristics in comparison to other structural constituents, although it is adversely affected by fires as well as biotic pests and abiotic effects. In this study, cedar (Cedrus libani A. Rich.) wood samples were impregnated using either Tanalith-E or Wolmanit-CB as detailed in ASTM-D 1413-76 and surface-treated using water-based or synthetic varnish. The impregnated and varnished samples were left outdoors with the aim of investigating the effect of seasonal changes. The samples were later subjected to combustion analysis as detailed in the combustion test standard ASTM-E 160-50. The results of the study indicated that the weight loss was lower during winter (87.82%), for samples that were impregnated using Wolmanit-CB (88.90%) and those that were treated with synthetic varnish (88.45%). On the other hand the O2 and CO2 contents were determined to be the highest in fall during combustion without flame (18.16% and 17.96%, respectively); the CO content was the highest during combustion without flame for the winter samples (28,907 ppm) and the NO content was the highest during combustion without flame for the spring samples (76.15 ppm). In conclusion, the winter samples that were impregnated using Wolmanit-CB and treated with synthetic varnish were determined to be safer to employ in areas with high fire risk.

The amount and type, the inorganic element content, and the maturity of organic materials of Eocene coals, shaly coals, and coaly shales exposed in the Gümüşhane and Bayburt districts of the Eastern Black Sea Region of Turkey were investigated. The depositional environments and hydrocarbon potentials were also interpreted. The total organic carbon concentrations in the studied samples ranged from 0.50% to 63.08%. The samples from Özyurt,Kayadibi, and Tarhanas contained types II and III kerogen, and those from Sökmen and Manas contained type III kerogen. The samples contained Co,Cs,Ga,Hf,Th,U,Y,Mo,Be,Cd,Sb,and La,with average values similar to those of standard brown coals. The samples showed average contents of Co,Ga,Nb,Rb,V,Y,Cu,Pb,Zn,As,Be,and Se,similar to those of other Turkish coals. The sediment source of Eocene samples in the five areas was characterized by rocks with intermediate or mafic geochemical characteristics. The terrigenous/aquatic ratio of coal and shaly coal samples of the areas in question is >1. The sterane distribution was C29 > C28 > C27 and C29 > C27 > C28 for the Özyurt and Tarhanas areas, respectively. The average Tmax values for samples are between 424 °C and 460 °C. For samples Oz-1 and Ta-2,22S/(22S + 22R) homohopane (C32) ratios are 0.48 and 0.61,respectively; 20S/(20S + 20R) sterane (C29) ratios are 0.18 and 0.53,respectively; and Ts/(Ts + Tm) ratios are 0.015 and 0.64,respectively. The Pr/Ph ratios of the samples are >3. The studied samples have low sterane/hopane and high (C19+C20)/C23) ratios without anoxic biomarkers (17α(H)-28,30-bisnorhopane). Based on these data, the coals, shaly coals, and coaly shales were probably deposited under an oxic-suboxic mixture of marine and terrestrial environment conditions; these materials contain terrestrial organic matter and cannot generate hydrocarbon.

İncele - MakaleAçık Erişim2016Scopus SJRQ2Optimum Tuning of a Gyroscopic Vibration Absorber Using Coupled Gyroscopes for Vibration Control of a Vertical Cantilever Beam
Ünker, F.; Çuvalci, O.
This paper deals with the investigation of optimum values of the stiffness and damping which connect two gyroscopic systems formed by two rotors mounted in gimbal assuming negligible masses for the spring, damper, and gimbal support. These coupled gyroscopes use two gyroscopic flywheels, spinning in opposing directions to have reverse precessions to eliminate the forces due to the torque existing in the torsional spring and the damper between gyroscopes. The system is mounted on a vertical cantilever with the purpose of studying the horizontal and vertical vibrations. The equation of motion of the compound system (gyro-beam system) is introduced and solved to find the response measured on the primary system. This is fundamental to design, in some way, the dynamic absorber or neutralizer. On the other hand, the effect of the angular velocities of the gyroscopes are studied, and it is shown that the angular velocity (spin velocity) of a gyroscope has a significant effect on the behavior of the dynamic motion. Correctness of the analytical results is verified by numerical simulations. The comparison with the results from the derivation of the corresponding frequency equations shows that the optimized stiffness and damping values are very accurate.

Beker, Y.; Sayil, N.; Akin, O.; Coban, K.H.
Microtremor measurement obtained by ground vibrations is one of the useful methods for determination of ground characteristics. Also the method has been using to specify building resonance risk during earthquakes in the last decades. In this study, single station microtremor recordings have been taken on the ground and at the 6 buildings in the KTU campus area. The measurements taken on the ground and in the buildings have been evaluated by Nakamura (H/V ratio) method. The eastern of the campus area presents Class-IV and the middle and western part of the campus present Class-II- Class-III according to Kanai and Tanaka ground classification. The predominant frequencies of the buildings are not in a harmony with the values of the ground. It can be deduce that the resonance risk is low for these buildings during the earthquakes. The H/V ratios of the ground change between 1-3 and lower than the buildings ratios which change between 5.0-8.5.

Top, Yener; Adanur, Hakan; Öz, Mehmet
Risk factors in the workplace vary according to the sector and scale of the business. Small and medium-sized enterprises, especially those within the scope of the wood-products manufacturing industry, are considered to be risky, and have a relatively high accident rate. Here, we focus on the timber and furniture industries, two subsectors of the wood-products industry. A total of 47 enterprises was visited and asked to participate in a formal structured questionnaire. The findings show that the usage of personal protective equipment (PPE) was low, that lighting was often inadequate, and that these enterprises typically lacked routine organising and cleaning practices. The subsectors and the occupational health and safety practices were found to be independent, and a statistically significant relationship could only be established between the subsectors in terms of the use of dust masks and goggles. Despite the unfavourable working conditions, only one business out of 47 reported having an employee with symptoms of occupational diseases.

Çullu, Mustafa; Bolat, Hakan; Vural, Alaaddin; Tuncer, Ertan
Mineralogical and chemical properties, as well as the pozzolanic activities, of the volcanic rocks in the northeast of the Black Sea were examined. Physical and mechanical tests were conducted on samples collected from the study area and the feasibility of using volcanic rocks as pozzolanic admixture in the concrete industry was investigated. Mortar samples containing admixture in the ratios of 0%, 10%, 20%, 30%, 40%, and 50% were prepared by keeping fineness of pozzolanic admixtures fixed. By using these mortar samples, prismatic mortar samples of 4×4×16 cm were prepared and the samples were cured in water. Their 7-, 28-, and 90-day tensile strengths, compressive strengths, and ultrasonic pulse velocity were determined. As a result, it was determined that volcanic rocks can be used to produce blended cement by considering their specific gravity, pozzolanic activities, and compressive strengths. It was noted that increase in trass rate significantly reduced tensile and compressive strength of cement in the early stage as expected.

Cancer is a heterogeneous disease, two of whose characteristic features are uncontrollable cell proliferation and insufficient apoptosis. Various studies have investigated the antiproliferative effects of propolis, a natural bee product, from different countries, and its cytotoxic effects have been attributed to its polyphenol contents. The purpose of this study was to show the cytotoxic effects, and possible mechanisms involved, of ethanolic extract of Turkish propolis (EEP) on the human lung cancer (A549) cell line. Cytotoxic activity of EEP on A549 cells was revealed using the MTT assay. Mechanisms involved in the cytotoxic action of EEP on A549 cells were then investigated in terms of apoptosis, mitochondrial membrane potential and cell cycle using flow cytometry, endoplasmic reticulum stress using RT-PCR, and caspase activity using luminometric analysis. EEP exhibited selective toxicity against A549 cells compared to normal fibroblast cells. We determined that EEP arrested the cell cycle of A549 cells at the G1 phase, induced endoplasmic reticulum stress, caspase activity, and apoptosis and reduced mitochondrial membrane potential. These results indicate that Turkish propolis is capable of reducing cancer cell proliferation and may have a promising role to play in the development of new anticancer drugs in the future.

Demand for organic milk is partially driven by consumer perceptions that it is more nutritious. However, there is still considerable uncertainty over whether the use of organic production standards affects milk quality. Here we report results of meta-analyses based on 170 published studies comparing the nutrient content of organic and conventional bovine milk. There were no significant differences in total SFA and MUFA concentrations between organic and conventional milk. However, concentrations of total PUFA and n-3 PUFA were significantly higher in organic milk, by an estimated 7 (95% CI -1, 15)% and 56 (95% CI 38, 74) %, respectively. Concentrations of alpha-linolenic acid (ALA), very long-chain n-3 fatty acids (EPA + DPA + DHA) and conjugated linoleic acid were also significantly higher in organic milk, by an 69 (95% CI 53, 84) %, 57 (95% CI 27, 87)% and 41 ( 95% CI 14, 68) %, respectively. As there were no significant differences in total n-6 PUFA and linoleic acid (LA) concentrations, the n-6:n-3 and LA: ALA ratios were lower in organic milk, by an estimated 71 (95% CI -122, -20)% and 93 (95% CI -116, -70) %. It is concluded that organic bovine milk has a more desirable fatty acid composition than conventional milk. Meta-analyses also showed that organic milk has significantly higher alpha-tocopherol and Fe, but lower I and Se concentrations. Redundancy analysis of data from a large cross-European milk quality survey indicates that the higher grazing/conserved forage intakes in organic systems were the main reason for milk composition differences.

Demand for organic meat is partially driven by consumer perceptions that organic foods are more nutritious than non-organic foods. However, there have been no systematic reviews comparing specifically the nutrient content of organic and conventionally produced meat. In this study, we report results of a meta-analysis based on sixty-seven published studies comparing the composition of organic and non-organic meat products. For many nutritionally relevant compounds (e.g. minerals, antioxidants and most individual fatty acids (FA)), the evidence base was too weak for meaningful meta-analyses. However, significant differences in FA profiles were detected when data from all livestock species were pooled. Concentrations of SFA and MUFA were similar or slightly lower, respectively, in organic compared with conventional meat. Larger differences were detected for total PUFA and n-3 PUFA, which were an estimated 23 (95% CI 11, 35)% and 47 (95% CI 10, 84)% higher in organic meat, respectively. However, for these and many other composition parameters, for which meta-analyses found significant differences, heterogeneity was high, and this could be explained by differences between animal species/meat types. Evidence from controlled experimental studies indicates that the high grazing/forage-based diets prescribed under organic farming standards may be the main reason for differences in FA profiles. Further studies are required to enable meta-analyses for a wider range of parameters (e.g. antioxidant, vitamin and mineral concentrations) and to improve both precision and consistency of results for FA profiles for all species. Potential impacts of composition differences on human health are discussed.

Gocer, Hulya; Topal, Fevzi; Topal, Meryem; Küçük, Murat; Teke, Dilek; Gülçin, Ilhami; Alwasel, Saleh H.; Supuran, Claudiu T.
Taxifolin, also known as dihydroquercetin, is a flavonoid commonly found in plants. Carbonic anhydrase (CA, EC 4.2.1.1) plays an important role in many critical physiological events including carbon dioxide (CO2)/bicarbonate (HCO3-) respiration and pH regulation. There are 16 known CA isoforms in humans, of which human hCA isoenzymes I and II (hCA I and II) are ubiquitous cytosolic isoforms. In this study, the inhibition properties of taxifolin against the slow cytosolic isoenzyme hCA I, and the ubiquitous and dominant rapid cytosolic isoenzyme hCA II were studied. Taxifolin, as a naturally bioactive flavonoid, has a K-i of 29.2nM against hCA I, and 24.2nM against hCA II. For acetylcholinesterase enzyme (AChE) inhibition, K-i parameter of taxifolin was determined to be 16.7nM. These results clearly show that taxifolin inhibited both CA isoenzymes and AChE at the nM levels.

- MakaleAçık Erişim2016Scopus SJRQ1Complex subvolcanic magma plumbing system of an alkali basaltic maar-diatreme volcano (Elie Ness, Fife, Scotland)
Gernon, T. M.; Upton, B. G. J.; Ugra, R.; Yücel, C.; Taylor, R. N.; Elliott, H.
Alkali basaltic diatremes such as Elie Ness (Fife, Scotland) expose a range of volcanic lithofacies that points to a complex, multi -stage emplacement history. Here, basanites contain phenocrysts including pyrope garnet and sub-calcic augites from depths of similar to 60 km. Volcanic rocks from all units, pyroclastic and hypabyssal, are characterised by rare earth element (REE) patterns that show continuous enrichment from heavy REE (HREE) to light REE (LREE), and high Zr/Y that are consistent with retention of garnet in the mantle source during melting of peridotite in a garnet lherzolite fades. Erupted garnets are euhedral and unresorbed, signifying rapid ascent through the lithosphere. The magmas also transported abundant pyroxenitic clasts, cognate with the basanite host, from shallower depths (similar to 35-40 km). These clasts exhibit wide variation in texture, mode and mineralogy, consistent with growth from a range of compositionally diverse melts. Further, clinopyroxene phenocrysts from both the hypabyssal and pyroclastic units exhibit a very wide compositional range, indicative of polybaric fractionation and magma mixing. This is attributed to stalling of earlier magmas in the lower crust principally from similar to 22 to 28 km as indicated by pyroxene thermobarometry. Many clinopyroxenes display chemical zoning profiles, occasionally with mantles and rims of higher magnesium number (Mg#) suggesting the magmas were mobilised by juvenile basanite magma. The tuffs also contain alkali feldspar megacrysts together with Feclinopyroxene, zircon and related salic xenoliths, of the 'anorthoclasite suite' - inferred to have crystallised at upper mantle to lower crustal depths from salic magma in advance of the mafic host magmas. Despite evidence for entrainment of heterogeneous crystal mushes, the rapidly ascending melts experienced negligible crustal contamination. The complex association of phenocrysts, megacrysts and autoliths at Elie Ness indicates thorough mixing in a dynamic system immediately prior to explosive diatreme-forming eruptions. Gücer, Mehmet Ali; Arslan, Mehmet; Sherlock, Sarah; Heaman, Larry M.
In the northern part of the Central Pontides (N Turkey) there are different metamorphic rocks exposed, notably the Devrekani metamorphic rocks. Here, upper amphibolite-lower granulite facies metamorphic rocks contain predominantly paragneiss, orthogneiss and metacarbonate, and to a lesser extent, amphibolite and quartzite, with cross-cutting aplite, pegmatite and granite veins. This is the first report of these rocks and includes new data on the petrochemistry, geochronology and metamorphic evolution of the Devrekani orthogneisses from the Central Pontides. The orthogneisses show five different mineral parageneses with the characteristic mineral assemblage quartz + K-feldspar + plagioclase + biotite +/- hornblende +/- opaque (+/- ilmenite and +/- magnetite), and accessory minerals (zircon, sphene and apatite). These metamorphic rocks exhibit generally granoblastic, lepidogranoblastic and nematolepidogranoblastic with locally migmatitic and relic micrographic textures. They have well-developed centimeter-spaced gneissic banding and display gneissose structure with symmetric, asymmetric and irregular folds. The petrographic features, mineralogical assemblages and weak migmatization reflect high temperature conditions. Thermometric calculations in the orthogneisses indicate metamorphic temperatures reached 744 +/- 33 A degrees C. Field relations, petrography and petrochemistry suggest that the orthogneisses have predominantly granodioritic and some granitic protoliths, that show features of I-type, medium to high-potassic calc-alkaline volcanic arc granitoids. The orthogneisses have high contents of LILEs and low contents of HFSEs with negative Nb and Ti anomalies, which are typical of subduction-related magmas. The orthogneisses also show significant LREE enrichment relative to HREE with negative Eu anomalies (Eu-N/Eu* = 0.33-1.07) with La-N/Lu-N = 6.98-20.47 values. Based on U-Pb zircon dating data, the protoliths are related to Permo-Carboniferous (316-252 Ma) magmatism. It is likely that peak metamorphism took place during the Jurassic as reflected by the U-Pb zircon ages (199-158 Ma) and also 40Ar/39Ar from hornblende/biotite (163-152 Ma). The four biotite 40Ar/39Ar average ages from the rock samples are ca. 156 Ma, suggesting that the metamorphic rocks cooled to 350-400 A degrees C at ca. 156 Ma. Conclusively, the Devrekani metamorphic rocks can be ascribed as products of Permo-Carboniferous continental arc magmatism overprinted by Jurassic metamorphism in the northern Central Pontides.

Eyuboglu, Yener; Dudas, Francis O.; Santosh, M.; Zhu, Di-Cheng; Yi, Keewook; Chatterjee, Nilanjan; Jeong, Youn-Joong; Akaryali, Enver; Liu, Ze
The Eastern Pontides Orogenic Belt represents one of the best examples of fossil convergent margins in the eastern Mediterranean region. However, the origin and geodynamic setting of the late Mesozoic-Cenozoic magmatism in this belt remain controversial due to lack of systematic geological, geochemical and chronological data. The general consensus is that the late Mesozoic-Cenozoic igneous activity is related to northward subduction of oceanic lithosphere in the late Mesozoic and following collision between Tauride and Pontide blocks in the early Cenozoic Here we present a comprehensive study focusing on the origin and geodynamic setting of gabbro bodies exposed along a narrow zone, parallel to the southeastern coast of the eastern Black Sea basin, in the Northern Zone of the Eastern Pontides Orogenic Belt. The studied gabbro bodies are hosted within late Cretaceous basaltic, andesitic, and dacitic volcanics including pyroclastic rocks and interbedded sedimentary rocks. The gabbro bodies range in size from 0.1 km(2) to 1.5 km(2), and outcrop patterns vary from round or elliptical to markedly elongate with sharp and discordant contact with the host rocks. Their mineral assemblage includes mainly clinopyroxene, plagioclase, minor olivine, amphibole, magnetite and rarely orthopyroxene, biotite, zircon and titanite. The occurrence of sutured grain boundaries on clinopyroxene and plagioclase, and the presence of reverse compositional zoning in clinopyroxene and olivine suggest mixing between magmas of contrasting compositions during mineral growth. Thermobarometric computations indicate that the temperature at the beginning of crystallization was similar to 1250 degrees C and crystallization was polybaric Zircon and titanite U-Pb ages indicate that these small intrusions were emplaced into crustal rocks of the Eastern Pontides Orogenic Belt during Lutetian (45 +/- 2 Ma). The depletion of HFSE is consistent with the involvement of an arc-related source in the petrogenesis of these rocks, and low to moderate enrichment Ce, Rb, Ba, K, Pb, Sr and Th suggests that involvement of subducted oceanic sediment was modest. The low Th content and low Th/Yb indicate that the role of sediment addition was nevertheless limited. The Nd, Sr and Pb isotopic data are consistent with the interpretation that the dominant source component in these gabbros is a depleted, peridotitic mantle, and that crustal contamination is relatively unimportant. We suggest that mafic magmas that produced the gabbroic intrusions were derived from melting of a depleted mantle source under the forearc region of the Eastern Pontides Orogenic Belt during southward subduction of two oceanic plates separated by a mid-ocean ridge, leading to the formation of a slab window. We also infer fractional crystallization and assimilation during both magma storage in the crust-mantle transition zone and transfer into the overlying arc crust.
