- MakaleAçık Erişim2024Gluconic Acid Production by Using Recombinant Escherichia coli Waksman pqq+ Cells with a Novel Approach from Biomass Sources
Gül, Abdulkadir; Tanyıldızı, Muhammet Şaban
Gluconic acid (GA) is widely used in the pharmaceutical, food, detergent, textile, leather, and concrete industries. However, cost-effective and high-yield production of GA remains a challenge. Due to currently high raw material inputs of GA, various alternative carbohydrate sources are being investigated. Sucrose is one of the cost-effective biomass sources that can be used as feedstock. The most common industrial production of GA is based on wild-type bacteria and fungi, but there are many problems with this production. This study aimed to optimize the production of GA from glucose produced by hydrolysis of sucrose using recombinant E. coli Waksman (W) pqq+ strain. After sucrose was enzymatically hydrolyzed, significant medium components for GA production were determined as glucose, calcium carbonate (CaCO3), peptone, and ammonium phosphate ((NH4)3PO4) using Placket-Burman Design (PBD). Detailed optimization of the medium components that are significant in GA production was carried out using central composite design (CCD), and optimum values of the independent variables examined in maximum GA production (93.5 ± 2.95 g/L) were determined as glucose 95, CaCO3 25, peptone 2, and (NH4)3PO4 1.13 (g/L). Using results obtained in the Erlenmeyer experiments, GA production in the bioreactor was investigated by CCD. The maximum GA efficiency (3.20 ± 0.15 g/L. h) was obtained under conditions where the air supply rate was 10.82 L/min, stirring speed was 656.87 rpm, and CaCO3 concentration was 16.90 g/L. In conclusion, it has been shown that GA can be produced with a high yield with this novel approach using a recombinant strain for GA production from sucrose.
- MakaleYalnız künye2024Education and reproductive health: evidence from schooling expansion in Turkey
De, Prabal K.; Tümay, Muhammed
We investigate the role of additional years of schooling mandated by a compulsory schooling expansion law in affecting reproductive preferences and safe reproductive health behaviors in Turkey—a middle-to-high-income country with gender inequity in education but overall high levels of safe reproductive health practices at the time of passing the law. Using a fuzzy regression discontinuity design, we find that the additional schooling improved several health behaviors. However, the effects on some outcomes commonly analyzed in the existing literature, such as contraceptive use or fertility, were either weak or insignificant. Overall, our findings complement the current literature on the marginal health benefits of schooling expansion and suggest that policymakers consider the institutional and cultural factors while evaluating the scope and potential non-educational benefits of such expansions.
- MakaleAçık Erişim2024A different look at the environmental Kuznets curve from the perspective of environmental deterioration and economic policy uncertainty: evidence from fragile countries
Anwar, Ahsan; Barut, Abdulkadir; Pala, Fahrettin; Kilinc-Ata, Nurcan; Kaya, Emine; Lien, Duong Thi Quynh
Environmental degradation is one of the most significant issues that developing nations confront and needs to be resolved right away in order for them to achieve sustainable development. Government policies are crucial in this situation since emerging nations frequently struggle with the issue of policy ambiguity, which can result in environmental deterioration. In this context, this study investigates how policy uncertainty affects environmental degradation in the five fragile emerging economies known as the Fragile Five—Brazil, India, Indonesia, South Africa, and Turkey. Using data from 1996 to 2019, we estimate a Panel Quantile Regression analysis. The empirical findings indicate that economic policy uncertainty and technology innovation increases the environmental degradation whereas environmental degradation is slowed down by financial development and renewable energy consumption. Empirical evidence also confirms the presence of EKC hypothesis in fragile economies. Based on the findings, we suggest both a policy and an environmental framework for achieving sustainable development in fragile economies. Graphical Abstract: (Figure presented.)

Uzun, Sevda; Kulakaç, Nurşen; Boyraz, Mehmet Çağatay
The aim of this study is to examine the opinions and experiences of operating room nurses about ethical sensitivity phemenologically. This phenomenological study, which is in the qualitative research design, was conducted with 14 operating room nurses. Prior to the study, a pilot study was conducted with three nurse families. The interviews were conducted using a face-to-face in-depth semi-structured interview form and the data were analyzed with Colaizzi’s phenomenological method. Interview data were collected between 15 June 2022 and 15 October 2022 in the country. Three categories, seven main themes, and twenty-three sub-themes emerged as a result of the interviews. In the theme of ethical dilemmas experienced by operating room nurses, there are sub-themes of professional ethics, teamwork, and patient dignity and benefit. There are physical and psychological sub-themes in the theme of ethical sensitivity perception and practices of operating room nurses, and among the subthemes, ensuring patient privacy, providing professional competence, providing psychological support to the patient, being friendly and communicating effectively, and respecting the patient’s autonomy come to the fore. Views of operating room nurses on the importance of ethical principles were discussed under the headings of profession and patient. Our results are valuable in that they shed light on the perspectives and experiences of the operating room nurses while maintaining ethical sensitivity. The high ethical sensitivity perceptions of operating room nurses are associated with an increase in the quality of care. It is thought that the high perception of ethical sensitivity of operating room nurses is an important factor that increases the satisfaction of individuals receiving care and the job satisfaction and quality of care of nurses.

Aktaş, Eda; Ustuner Top, Fadime; Uzun, Sevda
The study aimed to explore mothers’ experiences of children with autism using a phenomenological approach. Semi-structured, in-depth interviews were conducted with 18 parents with children attending a special education centre in a northern province of Turkey. Criterion sampling, a form of purposive sampling, was employed to select participants. Interviews continued until data saturation was achieved. All interviews were audio-recorded and transcribed verbatim. Thematic analysis was utilized to analyze the data, and the study adhered to the COREQ checklist for reporting qualitative research. As a result of analyses, three main themes (the process of diagnosis, the effect of the child’s illness on parents and their challenges over time, and considerations regarding the child’s educational journey) and eight sub-themes (feelings and thoughts, reflections on the causes, reactions encountered, emotional effects, social effects, physical effects, perception of social support, and expectations from the educator and the education process) emerged. The study concluded that mothers of children with autism are emotionally, physically, and socially impacted by their child’s condition, facing considerable challenges in coping. In light of these findings, it is recommended that policymakers in our country develop plans that address the difficulties expressed by participants, review and potentially enhance financial and social support for families, and consider increasing special education hours.
- MakaleAçık Erişim2024Production and Enrichment of Erucic Acid Methyl Ester Analytical Standards from Natural Oils for Food Control Analysis
Karabaş, Yağmur; Gül, Abdulkadir; Karpuz, Ömer; Akdoğan, Arda; Baltacı, Cemalettin
Foods high in erucic acid (EA), a fatty acid considered a natural toxin, can pose various health risks. Studies have reported that people exposed to high levels of EA are more likely to experience cardiovascular disease, are susceptible to myocardial adiposity, and have increased rates of diabetes. Therefore, in 2019, the European Commission (EC) declared that the maximum EA content in vegetable oils should be no more than 2%. These regulations require EA analysis in import and export food samples. A methyl ester of EA (EAME) standard is required to analyze EA content. In many countries, this analytical standard is not available and is being imported. This study endeavored to economically produce the EAME standard, which is in high demand due to mandated analysis, using natural oils. Initially, the fatty acid amounts in the food samples were analyzed by the gas chromatography–mass spectrometry/flame ionization detection (GC-MS/FID) method. The analyses revealed that mustard seed oil had the lowest EA content, while yellow mustard seed oil had the maximum EA content. The esterification was conducted on the samples containing both high and low EA, resulting in an efficiency of approximately 80%. The crystallization process was subsequently repeated to increase the EA quantity until the highest EAME content was obtained. By this process, 14 different concentrations of EAME ranging from 2.33 to 55.66% were produced. These results demonstrate the first successful production of EAME standards from natural oils that can be used in validation and internal quality control studies for food control analysis.
- MakaleAçık Erişim2024Investigation of Antibiofilm and Antibacterial Properties of Green Synthesized Silver Nanoparticles from Aqueous Extract of Rumex sp.
Akay, Seref; Yüksel, Gamze; Özad Düzgün, Azer
The decrease in the effectiveness of conventional drugs as a result of the growth of resistance to antibiotics has increased the need for innovative tools to control the infections. At this point, metallic nanoparticles, in particular silver nanoparticles, have appeared as a promising method. In the current study, the extract of Rumex sp. (Labada, dock) leaves was used as a reducing agent for the formation of silver nanoparticles. Unlike similar studies, in this study the synthesis conditions were optimized by changing the extract ratio and silver nitrate concentration. Morphological investigations of synthesized silver nanoparticles showed that spherical homogeneous particles at size under 100 nm had been produced. SEM/EDS and FTIR analyses showed that plant components are involved in the synthesis of nanoparticles. It was also determined that higher extract ratio reduced nanoparticle size. The antimicrobial effects of the synthesized nanoparticles against Gram-positive and Gram-negative bacteria were tested, and it was determined that all nanoparticles exhibited activity against both groups. Rumex sp. silver nanoparticles (NPs) were revealed to exhibit antibiofilm activity against three different isolates with moderate and strong biofilm-forming ability. The NPs reduced the biofilm-forming capacity of Acinetobacter baumannii and Klebsiella pneumonaie by 2.66-fold and 3.25-fold, whereas they decreased the Escherichia coli biofilm-forming capacity by 1.25-fold. The investigation of microbial biofilm could play an important role in developing new strategies for treatment options. Our results suggest that Rumex sp. silver NPs may have a high potential for use in the treatment of pathogenic strains.
- MakaleYalnız künye2024A New Approach for Ground Filtering of Airborne Laser Scanning Data Using PointNet++
Akbulut, Zeynep; Karsli, Fevzi; Dihkan, Mustafa
Ground filtering is an essential process that separates ground and non-ground points for generating digital elevation models (DEMs) from airborne laser scanning (ALS) point cloud data. Traditionally, ground filtering was carried out using rule-based approaches, but with the advancements in deep learning networks, ground filtering can now be performed using these methods. Deep neural networks (DNNs) require large amounts of training samples to reveal key information within the dataset. OpenGF is a new ultra-large-scale dataset which has nearly half a billion points from nine different scenes ranging from highly dense urban areas to steep vegetation covered mountains for ground filtering task. In this study, PointNet++ architecture was employed for ground filtering on the OpenGF dataset. Firstly, we slightly modified PointNet++ architecture’s sampling strategy to decrease the number of duplicate points in the batching process. We proposed a new adaptive tiling strategy based on input point cloud density to cope with highly varying point sampling of the OpenGF dataset. Modified PointNet++ architecture was trained with both adaptive tiling strategy and original OpenGF tiles to investigate the advantages and drawbacks of the proposed strategy on the ground filtering task. To evaluate the performance of the proposed strategy, the results for different test areas were compared with those obtained using the original PointNet++ and rule-based cloth simulation filtering (CSF) algorithms. Despite using fewer epochs, the proposed data collection strategy achieved up to 13% higher accuracy on test scenes with the modified PointNet++ architecture compared to the network trained with original OpenGF tiles.
- MakaleAçık Erişim2024Determination of the Cytotoxicity and Antibiofilm Potential Effect of Equisetum arvense Silver Nanoparticles
Akar, Zeynep; Akay, Seref; Ejder, Nebahat; Özad Düzgün, Azer
This study aimed to synthesize and characterize silver nanoparticles (AgNPs) by green synthesis from Equisetum arvense (Ea) extracts and to investigate their cytotoxicity, antibiofilm activity, and α-glucosidase enzyme inhibition. Diverse characterization techniques were applied to verify the production of nanoparticles. SEM examination confirmed that the size of nanoparticles is in the range of 40–60 nm. Also, interactions between silver and natural compounds of plant extract were confirmed through FT-IR and EDX analyses. It was determined that Equisetum arvense silver nanoparticles had antibiofilm activity against three different clinical strains with high biofilm-forming ability. AgNPs reduced the biofilm-forming capacity of clinical A. baumannii isolate with strong biofilm-forming capacity by approximately twofold, while the capacity of clinical K.pneumonaie and E.coli isolates decreased by 1.5 and 1.2 fold, respectively. The α-glucosidase enzyme inhibition potential of the AgNPs, which is determined as 93.50%, was higher than the plant extract with, and the α- 30.37%. MTT was performed to assess whether incubation of nanoparticles with A549 and ARPE-19 cell lines affected their viability, and a dramatic reduction in cell growth inhibition of both A549 and ARPE-19 cells was observed. It has been shown that A549 cells treated with 200 and 150 µg/mL nanoparticles had less cell proliferation compared to control cells at 24-h and 48-h incubation time. According to these results, Ea-derived AgNPs appear to have potential anticancer activity against A549 cancer cells. Investigating the effects of green synthesis nanoparticles on microbial biofilm and various tumors may be important for developing new therapies. The outcomes of this study have showed that Ea-AgNPsmay have a high potential both in the treatment of pathogenic strains that form biofilms, as well as in anticancer therapy use.
- MakaleYalnız künye2024Selective Hydrogenation of Croton Aldehyde on Pt Nanoparticles Controlled by Tailoring Fraction of Well-Ordered Facets Under Different Pretreatment Conditions
Sarıbıyık, Oğuz Y.; Resasco, Daniel E.
This study focuses on the influence of pretreatment conditions and surface structures of well-tuned Pt nanoparticles on the selective hydrogenation of α-β unsaturated crotonaldehyde. The surface binding site preferences of the reactant during catalytic conversions are strongly influenced by the configuration of the nanoparticles’ surface fraction exposed. Since the orientation of the well-ordered surfaces are significantly affected by the pretreatment conditions, we have attempted to obtain relationships between morphology changes and catalytic activity by controlling the fraction of facets with ordered structures. Pt nanoparticles exhibit different types of exposed surfaces, namely Cubic (Cub) (100) and Cuboctahedra (CubOc) (111) + (100). These different surface structures play a crucial role in the preferential hydrogenation of the model molecule. Additionally, we investigated the impact of the pretreatment condition on the catalytic selectivity of the well-shaped nanoparticles. We employed various techniques such as high-angle annular dark-field scanning transmission electron microscopy (HAADF-STEM), transmission electron microscopy (TEM), and room temperature carbon monoxide adsorption Fourier transform infrared spectroscopy (RT-CO-FTIR) to determine the reconstructions of the particles under different pretreatment conditions. The results indicate that both the well-ordered surface fraction and pretreatment conditions of Pt nanoparticles had a moderate influence on both catalytic activity and selectivity. Overall, this study highlights the importance of analyzing the surface morphology and pretreatment conditions of nanoparticles in order to optimize catalytic activity and selectivity in reactions involving well-tuned surface structures. In this article, the 1st and 6th paragraphs in the “Discussion” section contain several errors. The corrected paragraphs are given below. This study aimed to assess the spiritual well-being levels and spiritual care needs of individuals diagnosed with lung cancer. The findings revealed that the patients exhibited high spiritual well-being levels, with the “harmony with nature” sub dimension scoring particularly high in relation to spiritual well-being. This aligns with several studies which reported high spiritual well-being of cancer patients (Martins et al., 2020; Rabow & Knish, 2015).” The study identified high spiritual needs among the patients, with the specific subdimension “informing them about their health” obtaining the highest scores. Notably, in the current study, it was observed that patients’ needs in terms of “informing them about their health” increased, deviating from the emphasis on love, support, and bonding seen in other studies (Kırca et al., 2023; Shi et al., 2023). This divergence is thought to be linked to differences in the disease stage. The original article has been corrected.

Aktaş, Haktan; Meral Aktaş, Hacer; Ürkek, Bayram; Şengül, Mustafa; Çetin, Bülent
Kefir, which has many beneficial effects on health, is one of the most consumed fermented milk products worldwide. It is important to increase consumption of the fermented product for public health. In this study, it was aimed to increase the beneficial effects of kefir on public health. Therefore, kefirs produced from different types of milk (cow, buffalo, sheep, and goat) were concentrated, and obtained spreadable kefir samples were investigated in terms of their microbiological characteristic (lactic bacilli, lactic cocci, yeasts and moulds, total bacteria, and coliform bacteria), benzoic acid content, physicochemical properties (fat, total solid, ash content, acidity, pH, syneresis, viscosity, colour, and rheological properties), and sensory characteristic. It was determined that APC, lactic bacilli, lactic cocci, and yeast counts of the concentrated kefir samples changed between 6.90 and 8.64, 6.89 and 8.61, 7.42 and 8.72, and 2.17 and 5.39 log CFU/g, respectively, during storage. Mould and coliform bacteria were not detected in the samples. The concentrated kefir samples contained benzoic acid in the range of 18.30–119.58 mg/L. Results from this study showed that type of milk caused differences on APC, lactic bacilli, lactic cocci and yeast count, total solids, ash, fat, acidity, pH, syneresis, colour, viscosity and rheological parameters, and benzoic acid content. In addition, milk type affected sensory properties of the kefirs. Concentrated kefirs produced from cow and buffalo milk were the most liked by panellists. Finally, it was determined that concentrated kefir was favoured as a new product by most of the panellists.

- MakaleAçık Erişim2024Exploring the enablers for building resilience in solar photovoltaic Energy supply chains
Erol, Ismail; Peker, Iskender; Benli, Tolga; Ar, Ilker Murat; Searcy, Cory
A solar photovoltaic energy supply chain (SPvESC) is a global network with several linkages, including mineral and metal mining, material processing, and module and panel manufacturing. Due to the wide range of uncertainties and the unfavorable environmental effects associated with current linear business models, this global network is vulnerable to disruptions. Strengthening the resilience of SPvESCs is crucial for addressing any disturbances. This requires identifying the key enablers of resilience in SPvESCs, an area that has been understudied in the existing literature. An enabler is an aspect that facilitates the achievement of a goal by another aspect. This research contributes to the existing literature by systematically investigating the enablers for SPvESCs to achieve resilience. Thus, the objective of this analysis is to identify enablers that have the potential to enhance the resilience of SPvESCs in Türkiye. This was done by applying the Nominal Group Technique (NGT) in conjunction with a review of the current literature. Neutrosophic (N)-DEMATEL was then utilized to determine the relationships between the identified enablers. Finally, the results were validated using N-DELPHI. The results revealed that sensing and seizing new business models, adaptability to changes in novel energy generation and information technologies, and business contingency plans for natural and man-made disasters were the most influential enablers. The findings provide implications for practitioners, policymakers, and researchers to help ensure improved resilience in SPvESCs.
