In this study, a series of new benzohydrazide derivatives (1a-i) were synthesized, and characterized as potential cholinesterase inhibitors for use in the treatment of Alzheimer’s disease (AD). The inhibitory activities of the synthesized compounds against acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) enzymes were evaluated using in vitro and in silico methods. IC50 values for AChE inhibition ranged from 29.50 to 46.21 nM, with compound 1c exhibiting the highest inhibitory potential at an IC50 of 29.50 nM. For the BChE enzyme, the strongest effect was observed with compound 1d at an IC50 of 55 nM. The synthesized derivatives exhibited higher inhibitory potential compared to the reference tacrine (IC50: 58.73 nM for AChE and 187.30 nM for BChE) against cholinesterase enzymes through both competitive and non-competitive inhibition mechanisms. Molecular docking (ΔG: -10.67 to -9.95 kcal/mol for AChE; -9.80 to -8.69 kcal/mol for BChE) and 100 ns molecular dynamics simulations supported the binding affinities and indicated the dynamic stability of the enzyme-inhibitor complexes, in agreement with the experimental inhibition data. In silico ADMET profiling indicated favourable drug-likeness and high predicted gastrointestinal absorption and blood-brain barrier permeability (TPSA 41.46-61.69 A2), and structural alert screening returned no PAINS matches. The findings suggest that these derivatives may serve as promising lead compounds for further investigation as potential cholinesterase inhibitors relevant to AD, pending experimental cytotoxicity evaluation.

A series of mono- and bis-ureido sulfapyridine derivatives (SP-1-SP-10) were synthesized and evaluated as dual cholinesterase inhibitors targeting acetylcholinesterase (AChE) and butyrylcholinesterase (BChE). All compounds exhibited nanomolar inhibition. The most potent AChE inhibitors were SP-8 (K-i = 5.25 +/- 0.73 nM), SP-9 (5.09 +/- 0.44 nM), and SP-10 (5.74 +/- 0.55 nM), significantly outperforming tacrine (K-i = 105.03 +/- 8.85 nM) under identical conditions. For BChE, SP-9 showed the strongest inhibition (K-i = 26.38 +/- 2.07 nM) compared with tacrine (172.03 +/- 32.42 nM). Kinetic analyses revealed competitive inhibition toward AChE for all derivatives, whereas BChE inhibition displayed both competitive and noncompetitive patterns depending on structural features. Glide XP docking using AChE (PDB: 7XN1) and BChE (PDB: 4BDS), complemented by MM-GBSA binding free energy calculations, supported favorable ligand accommodation within the catalytic pockets and highlighted the importance of aromatic and hydrophobic interactions. In silico ADME/Tox analysis revealed a potency-drug-likeness trade-off: mono-ureido derivatives complied with Lipinski's rule of five and showed high predicted oral absorption, whereas bis-ureido derivatives, despite superior enzymatic potency, exhibited multiple drug-likeness violations. These findings identify the ureido-sulfapyridine scaffold as a potent enzymatic dual cholinesterase inhibitory framework, warranting further structural optimization to improve pharmacokinetic properties.

The development of effective acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) inhibitors remains a crucial objective in the treatment of neurodegenerative diseases, including Alzheimer's disease (AD). In this study, a novel synthetic approach was established to prepare a series of diaryl-substituted tacrine derivatives. Initially, brominated aminobenzonitrile was subjected to Suzuki-Miyaura cross-coupling reactions with various aryl boronic acids to yielding diaryl-substituted aminobenzonitriles at yields of 60%-72%. These intermediates were then cyclized with cyclohexanone and cycloheptanone under Friedl & auml;nder conditions using BF3 & centerdot;OEt2 as a catalyst, affording six- and seven-membered diaryl tacrine analogues in yields of 52%-68%. All synthesized compounds were characterized by NMR, IR, and elemental analysis. The inhibitory activities of these compounds (7-16) against AChE and BChE were evaluated, and several analogs demonstrated potent dual-enzyme inhibition at nanomolar concentrations. Notably, compounds 8 and 13 exhibited half maximal inhibitory concentration (IC50) values of 6.848 and 8.545 nmol/L for BChE, respectively, indicating strong potential as selective cholinesterase inhibitors. The structure-activity relationship (SAR) analysis revealed the influence of aryl substituents on the potency and selectivity of enzyme inhibition. Compared to previously reported tacrine derivatives, the newly synthesized analogs exhibited superior inhibitory profiles, indicating their potential as lead compounds for further development in AD therapeutics. Comparison of Online and Hybrid Education Models for First Aid and Emergency Care Students

Karabela, Semsi Nur; Kahraman, Beliz Yekeler; Değirmenci Bingöl, Nurhan; Döğer, Berna Nur Berker

Background: The Coronavirus Disease 2019 pandemic disrupted education and training processes, necessitating a transition to remote learning that significantly altered teaching methods for both students and instructors. To highlight the challenges arising from this rapid shift in applied health sciences, the present study aimed to compare the perceptions of professional competence among First and Emergency Aid students who received online versus hybrid education during the pandemic. Materials and Methods: This descriptive cross-sectional study was conducted among second-year students at two universities: one providing fully online education (n = 59) and the other offering hybrid instruction (n = 61). Results: No statistically significant difference was observed between the groups in understanding theoretical courses (p = 0.088); however, students who received face-to-face instruction within the hybrid model demonstrated significantly better comprehension of practical courses (p < 0.001). Conclusion: In conclusion, while online education is adequate for acquiring theoretical knowledge, it is insufficient for developing clinical and technical skills. Accordingly, these findings suggest that practical courses in health education programs, such as First and Emergency Aid, should be conducted face-to-face whenever possible; when in-person attendance is required, appropriate safety measures should be implemented.

Montessori ve STEM Yaklaşımlarının Karşılaştırmalı İncelenmesi: Türkiye’deki Lisansüstü Araştırmaların Analizi

    Arslan, Adem

Çalışma, yöntemsel çeşitliliğin artırılması, farklı yaş gruplarının sürece dahil edilmesi, ebeveyn katılımının güçlendirilmesi ve her iki yaklaşımın öğretmen eğitimi programlarına entegre edilmesi gerektiğini önermektedir.

The effect of acupressure on symptom management in children: A meta-analysis of randomized controlled trials

Akay, Gamze; Yıldız, Muhammed; Oğuzhan, Hatice; Uzun, Sevda

Purpose Pediatric patients often experience distress, pain, and nausea during medical procedures. This meta-analysis aimed to synthesize current evidence on the potential efficacy of acupressure on symptom management among pediatric patients aged 0–19 years. Design and methods This study was designed as a meta-analysis of randomized controlled trials . The literature search was conducted in PubMed, Web of Science, Scopus, EBSCOhost, Google Scholar, and the YÖK Thesis Center in January 2025 and updated in August 2026, with no restriction on publication year. After the reviews, 17 studies were included in the study. The data were synthesized by meta-analysis and narrative methods. Results The pooled analysis indicated that acupressure was associated with statistically significant reductions in reported anxiety (SMD = −1.021, p < 0.001), pain (SMD = −1.397, p < 0.001), and nausea (SMD = −0.388, p = 0.006). However, substantial statistical heterogeneity was observed for anxiety (I2 = 78.8%) and pain (I2 = 94.4%), and the number of studies available for each outcome was limited (anxiety: n = 6; pain: n = 9; nausea: n = 4). Conclusions and practice implications Current evidence suggests that acupressure may be a promising complementary approach for managing anxiety, pain, and nausea in pediatric patients. However, given the small number of studies per outcome, small sample sizes, and high statistical heterogeneity, these findings should be interpreted with caution. Further rigorous, well-designed RCTs are required to confirm these observations before routine clinical implementation can be recommended.

Electric field–induced inflammatory and angiogenic responses in rat female reproductive tissues: evidence for early extracellular matrix remodeling

Ozmen, Ozlem; Asci, Halil; Kolay, Oznur; Selcuk, Elif Nur; Yuksel, Duygu; Comlekci, Selcuk; Karabacak, Pinar

High-intensity electric fields (EFs) are increasingly encountered in occupational and environmental settings, raising concerns regarding their potential biological effects on hormonally responsive organs. However, the cellular and molecular responses of female reproductive tissues to short-term exposure to high-voltage EFs remain insufficiently characterized. This study aims to investigate EF-associated histopathological and immunohistochemical alterations in major female reproductive organs. Forty adult female Wistar albino rats were randomly assigned to five experimental groups (n = 8 per group): a control group (0 min) and four EF-exposed groups subjected to a nominal electric field intensity of 10 kV/m EF for 1, 5, 15, or 30 min using a custom-designed parallel-plate exposure system. The field intensity was verified at predefined measurement points within the exposure setup. Ovaries, uterus, and uterine tubes were examined using hematoxylin–eosin staining and semi-quantitative histopathological scoring. Immunohistochemical analyses were performed to evaluate the expression of tumor necrosis factor-α (TNF-α), vascular endothelial growth factor (VEGF), and osteonectin as markers of inflammation, angiogenesis, and extracellular matrix remodeling. Exposure to the electric field was associated with time-dependent histopathological alterations, including tissue edema, hemorrhage, epithelial degeneration, and leukocyte infiltration. The most pronounced ovarian and uterine changes were observed in the 30-min exposure group, whereas the uterine tubes exhibited comparatively milder structural alterations. Immunohistochemical analysis demonstrated increased TNF-α and VEGF expression in higher-exposure groups, suggesting activation of inflammatory and angiogenic pathways. Osteonectin expression was elevated in all examined reproductive tissues and was also detected in regions without overt morphological damage, indicating its potential sensitivity to early tissue stress responses associated with EF exposure. Significant correlations were observed between histopathological injury scores and immunohistochemical marker expression levels. Overall, short-term exposure to high-intensity electric field was associated with inflammatory signaling, angiogenic responses, and extracellular matrix remodeling in female reproductive tissues in this experimental model. These findings provide histological and immunohistochemical evidence of tissue responses to EF exposure and underscore the need for further studies incorporating comprehensive exposure characterization and additional molecular endpoints to better define potential reproductive health implications to high-voltage electric fields.
