In sepsis-associated cardiotoxicity, remifentanil reduces oxidative stress, inflammation, and apoptosis to maintain mitochondrial biogenesis by inhibiting NFkB and cas-3 immunoexpressions and enhancing SIRT1 upregulation

Tepebaşi, Muhammet Yusuf; Aşci, Halil; Selçuk, Esma; Uysal, Dinçer; Bindal, Ahmet; Yüksel, Duygu; Özmen, Özlem

Objective: Sepsis-induced cardiotoxicity (SIC) is a critical complication characterized by inflammation, oxidative stress, and apoptosis, leading to myocardial dysfunction. The short-acting opioid analgesic remifentanil (REMI) possesses antioxidant and anti-inflammatory properties. This study aimed to evaluate the cardioprotective effects of REMI on lipopolysaccharide (LPS)-induced SIC by examining inflammation, oxidative stress, apoptosis, and mitochondrial function. Materials and methods: Thirty-two female Wistar albino rats were divided into four groups: control, lipopolysaccharide (LPS), LPS+REMI, and REMI. Myocardial and aortic tissues were analyzed for histopathology, immunoexpression of Caspase-3 (Cas-3), nuclear factor kappa beta (NF-κB), and tumor necrosis factor alpha (TNF-α). Oxidative stress markers, including total oxidant status (TOS), total antioxidant status (TAS), and oxidative stress index (OSI), were measured. Mitochondrial apoptosis-related gene expression of AMP-activated protein kinase (AMPK), BCL2 Associated X (BAX), B-cell lymphoma 2 (BCL-2), Sirtuin 1 (SIRT1), and Peroxisome proliferator-activated receptor gamma coactivator 1-alpha (PGC1α) was assessed. Results: LPS administration induced severe histopathological damage, increased oxidative stress (elevated TOS and OSI), and upregulated apoptotic (Cas-3, BAX/BCL-2 imbalance) and inflammatory (NF-κB, TNF-α) markers. REMI treatment significantly alleviated myocardial and aortic injury, reducing the histopathological score. It markedly decreased Cas-3, NF-κB, and TNF-α expression, lowered TOS and OSI levels, and modulated the BAX/BCL-2. Furthermore, REMI restored the expression of AMPK, SIRT1, and PGC-1α genes, indicating a protective effect on mitochondrial biogenesis and energy metabolism. Conclusions: REMI exhibits significant cardioprotective effects in LPS-induced SIC by attenuating inflammation, oxidative stress, and apoptosis while preserving mitochondrial homeostasis.

A Dose-Dependent Study Examining Dexmedetomidine's Possible Effects Against Oxidative, Fibrotic, and Apoptotic Damage Induced by Radiation Exposure in Spleen Tissue

Beyazal Polat, Hatice; Yilmaz, Hamit; Kilinc, Kagan; Gülhan, Belemir; Yilmaz Rakici, Sema; Tümkaya, Levent

Objective: This study aimed to investigate the potential splenic tissue damage induced by radiotherapy (RT) and the potential protective effect of different doses of dexmedetomidine on this damage at the histopathological, immunohistochemical, and biochemical levels. Materials and Methods: In our study, Sprague Dawley rats were randomly divided into four groups: Control, Radiotherapy (RT; 8 Gy), RT + Dexmedetomidine 100 mu g/kg (RT-D100), and RT + Dexmedetomidine 200 mu g/kg (RT-D200). A single dose of 8 Gy radiotherapy was administered to each RT group. Spleen tissues were examined histologically with hematoxylin-eosin and immunohistochemically with anti-Caspase-3, anti-TGF-beta 1, and anti-TGF-beta 3 using light microscopy. TBARS and total thiol levels were also analyzed to assess oxidative stress and antioxidant capacity. Results: Histopathological results showed a significant decrease in white pulp diameter, decreased cellular density, and increased congestion in the red pulp in the RT group. Significant fibrosis, sinusoidal dilatation, vacuolization, and amyloid deposition were detected in the white pulp in the RT group. Regarding anti-caspase-3 immunoreactivity, strong positivity increased in the red pulp in the RT group, while a significant increase was observed in the white pulp in both the RT-D100 and RT groups. While the proportion of TGF-beta 1 immunopositive cells did not change significantly in the RT group, they increased significantly in both dexmedetomidine groups (especially RT-D200). TGF-beta 3 expression increased significantly only in the RT-D100 group. In biochemical analyses, TBARS levels increased significantly in the RT-D100 group. Total thiol levels decreased in the RT group and increased in the dexmedetomidine-treated groups. Conclusions: While RT caused histopathological damage and increased oxidative stress in spleen tissue, dexmedetomidine reduced this damage in a dose-dependent manner. The different immunohistochemical profiles of TGF-beta 1 and TGF-beta 3 suggest that these cytokines may have different functions in the spleen. 100 mu g/kg dexmedetomidine stimulates a regenerative response through TGF-beta 3, while 200 mu g/kg dexmedetomidine may provide immune regulation and antioxidative defense through TGF-beta 1.

Turan, Ibrahim; Canbolat, Deniz; Demir, Selim; Kerimoglu, Gokcen; Colak, Fatih; Alemdar, Nihal Turkmen; Mentese, Ahmet; Aliyazicioglu, Yuksel

The nephrotoxicity mechanisms caused by cisplatin (CP), which is used in the treatment of different types of cancer, include several processes, such as DNA damage, apoptosis, inflammation, and oxidative stress. This study investigated the protective effects of Rhododendron luteum leaf extract (RLE) in a rat model of CP -induced nephrotoxicity. Dimethyl sulfoxide extract of RLE was prepared, and its contents were clarified using liquid chromatography-mass spectrometry (LC-MS -MS). Thirty female Wistar Albino rats were divided into five different groups and sacrificed on the seventh day. Biochemical and histopathological analyses were then performed. The presence of quercitrin, quinic acid, chlorogenic acid, protocatechic acid, isoquercitrin, and quercetin was detected in the RLE. While CP significantly increased the levels of oxidative stress, inflammation, and apoptosis markers in the rat kidney tissue and blood samples, treatment with RLE significantly reversed the levels of these parameters in a dose-dependent manner. Total antioxidant status (TAS) values and glutathione (GSH) levels increased significantly in the serum samples from the treatment group compared to the CP group, while blood urea nitrogen (BUN), creatinine, oxidative stress index (OSI), tumor necrosis factor-alpha (TNF-alpha), malondialdehyde (MDA) and total oxidant status (TOS) values decreased significantly. GSH levels increased significantly in the treatment group compared to the CP group, while TNF-alpha, caspase-3 (Casp-3), 8-hydroxy-2'-deoxyguanosine (8-OHdG), OSI, TOS, MDA levels, and damage scores decreased significantly at histopathological examination. In conclusion, RLE exhibited strong protective effects against nephrotoxicity and oxidative damage caused by CP in rats.

Aliyazicioglu, Yuksel; Demir, Selim; Yaman, Scrap Ozer; Sener, Sila Ozlem; Demir, Elif Ayazoglu; Aliyazicioglu, Rezzan; Turan, Ibrahim

The purpose of this study was to investigate the phytochemical analysis of D. pentaphyllum and its cytotoxic activity on human cervical cancer (HeLa) cells with possible mechanisms. Total phenolic contents (TPC) and phytochemical analysis of the extract were evaluated using spectrophotometric method and RP-HPLC, respectively. The cytotoxic effect of the extract was evaluated using the MTT assay. The mechanism of the cytotoxic effect of the extract was also evaluated in terms of apoptosis, cell cycle, and mitochondrial membrane potential (MMP) using the fluorometric methods. The TPC of the extract was calculated as 68.9 +/- 2.3 mg gallic acid equivalent per g sample and quercetin was determined as the major phenolic in the extract. The extract showed a selective cytotoxic effect (6.5-fold) on HeLa cells compared to normal fibroblast cells. The cytotoxic effect of the extract was found to be due to the arresting of the cell cycle in the S phase and increasing apoptosis through decreased MMP. Further studies are needed for the comprehensive effects of the extract on cell signaling.

Turan, Ibrahim; Demir, Selim; Kilinc, Kagan; Yaman, Serap Ozer; Misir, Sema; Kara, Hanife; Genc, Berna; Mentese, Ahmet; Aliyazicioglu, Yuksel; Deger, Orhan

Rosa canina is a member of the genus Rosa that has long been used for medical objectives. Several studies have reported cytotoxic effects of different Rosa species, but there has been only limited investigation of the cytotoxic effect of R. canina. The purpose of the current study was to examine the potential effect of R. canina extract on cell viability, the cell cycle, apoptosis, and the expression of telomerase in human colon cancer (WiDr) cells. The cytotoxic effect of the extract was determined using MTT assay. The mechanism involved in the cytotoxic effect of the extract was then evaluated in terms of apoptosis and the cell cycle using flow cytometry. Mitochondrial membrane potential (MMP) was investigated using the fluorometric method, and expression levels of telomerase were studied using RT-PCR. R. canina extract exhibited a selective cytotoxic effect on WiDr cells compared with normal colon cells. The extract induced cell cycle arrest at the S phase and apoptosis via reduced MMP in WiDr cells. R. canina extract significantly repressed telomerase expressions at treatment times of 48 and 72 h in WiDr cells. Our results suggest that R. canina may have considerable potential for development as a novel natural product-based anticancer agent.

The effects of 4-Vinylcyclohexene diepoxide on gene expression in dog testes

    Paksoy, Zahid

In this study, the effects of 4-Vinylcyclohexene diepoxide (VCD) on the testicular gene expression in dogs were investigated. Eighteen male dogs were used as material and were randomly allocated to 3 groups. The first group (n=6) was administered sesame oil by intraperitoneal (i.p.) route once a day for 8 days, and this group was served as a control. The second group (n=6) received 80 mg/kg VCD and the third (n=6) group was administered VCD (i.p.) at a dose of 320 mg/kg once daily for 8 days. The testes were removed on following day the last VCD administration. The testes were immediately placed in the RNA stabilization solution and stored at -20°C. Later mRNAs were obtained from tissue samples. After RNA isolation, the cDNAs were synthesized. Finally, MasterMix was prepared and gene expression levels were examined at the real-time PCR. The expression of B cell lymphoma 2 (Bcl-2), tumor suppressor protein (tp53), Bax, CASP2, CASP3, CASP8, CASP9 and β-actin were investigated in the all groups. Cross-group comparisons were made by using student t test. The tp53 gene expression in the second group was found to be higher compared to the control group. It was determined that the expression of Bcl-2 was reduced in the testes of animals treated with high-dose VCD. CASP-3 was found to be more expressed and CASP-9 was down-regulated in the third group. In addition, CASP-8 gene expression was significantly reduced in both treatment groups. As a result, it can be concluded that expression changes in the tp53, Bcl-2, CASP-3, CASP-8 and CASP-9 genes in the testes of male dogs may cause apoptotic changes with the VCD application.

Demir, Selim; Turan, Ibrahim; Aliyazicioglu, Rezzan; Yaman, Serap Ozer; Aliyazicioglu, Yuksel

Primula vulgaris belongs to the genus Primula, members of which are frequently used in folk medicine. Various studies have investigated the cytotoxic effect of different Primula species, but there have been limited studies on the cytotoxic effect of P. vulgaris. The aim of this study was to investigate the cytotoxic effects, and possible mechanisms involved, of P. vulgaris flower extract on human cervical cancer (HeLa) cells. The cytotoxic effect of the extract on HeLa cells was revealed using the MTT assay. Mechanisms involved in the extract's cytotoxic effect were then investigated in terms of apoptosis, mitochondrial membrane potential, and the cell cycle, using fluorometric methods. P. vulgaris flower extract exhibited selective cytotoxic effects against HeLa cells by arresting their cell cycle at the S phase, and inducing the number of apoptotic cells compared to normal fibroblast cells by reducing mitochondrial membrane potential in a concentration-dependent manner. This is the first study to reveal the antiproliferative effect of P. vulgaris flower extract. Further studies are now needed to identify the cytotoxic molecules in the extract and their mechanisms.

Turan, Ibrahim; Demir, Selim; Kilinc, Kagan; Burnaz, Nesibe Arslan; Yaman, Serap Ozer; Akbulut, Kubra; Mentese, Ahmet; Aliyazicioglu, Yuksel; Deger, Orhan

Background: Morus nigra L. belongs to the family Moraceae and is frequently used in traditional medicine. Numerous studies have investigated the antiproliferative effects of various extracts of different Morus species, but studies involving the in vitro cytotoxic effect of M. nigra extract are very limited. The purpose of this study was to evaluate the phenolic composition and antioxidant activity of dimethyl sulfoxide extract of M. nigra (DEM) and to investigate, for the first time, the probable cytotoxic effect in human prostate adenocarcinoma (PC-3) cells together with the mechanism involved. Methods: Total polyphenolic contents (TPC), ferric reducing antioxidant power (FRAP) and phenolic compounds of DEM were evaluated using spectrophotometric procedures and HPLC. The cytotoxic effect of DEM on PC-3 cells was revealed using the MTT assay. Mechanisms involved in the cytotoxic effect of DEM on PC-3 cells were then investigated in terms of apoptosis, mitochondrial membrane potential and cell cycle using flow cytometry, while caspase activity was investigated using luminometric analysis. Results: TPC and FRAP values were 20.7 ± 0.3 mg gallic acid equivalents and 48.8 ± 1.6 mg trolox equivalents per g sample, respectively. Ascorbic acid and chlorogenic acid were the major phenolic compounds detected at HPLC analysis. DEM arrested the cell cycle of PC-3 cells at the G1 phase, induced apoptosis via increased caspase activity and reduced mitochondrial membrane potential. Conclusions: Our results indicate that M. nigra may be a novel candidate for the development of new natural product based therapeutic agents against prostate cancer.

Morus rubra extract induces G1 cell cycle arrest and apoptosis in human lung and prostate cancer cells

Turan, Ibrahim; Demir, Selim; Kilinc, Kagan; Aliyazicioglu, Yuksel; Alver, Ahmet; Misir, Sema; Yaman, Serap Ozer; Akbulut, Kubra; Mentese, Ahmet; Deger, Orhan

Objective: Cancer is one of the most deadly types of disease and evasion from apoptosis and unstoppable cell proliferation are accepted as its distinctive features. Many studies have evaluated the cytotoxic effect of different Morus species but, there is no study about cytotoxic effect of Morus rubra. In this study we aimed to evaluate phenolic composition, antioxidant properties and cytotoxic effect of acidified dimethyl sulfoxide extract of M. rubra (AMRE). Method: Antioxidant properties, phenolic composition and cytotoxic effect of AMRE were determined using spectrophotometric methods, HPLC, and MTT assay, respectively. Then, mechanisms of cytotoxic effect of AMRE on human prostate (PC-3) and lung (A549) cancer cells were examined in regard to cell cycle, apoptosis and mitochondrial membrane potential using flow cytometric methods. Results: Total phenolic content and reducing power values were 11.9 mg gallic acid equivalents and 42.9 mg trolox equivalents per g sample, respectively. Ascorbic and gallic acid were detected in AMRE as major antioxidant compounds. We determined that AMRE increased cell cycle arrest at G1 phase and exhibited apoptotic features via decreasing mitochondrial membrane potential in both prostate and lung cancer cells. Conclusion: These findings demonstrate that M. rubra extract can affect the behavior of human prostate and lung cancer cells in vitro conditions, and this effect now needs to be investigated in vivo.

Increased expressions of ADAMTS-13 and apoptosis contribute to neuropathology during Toxoplasma gondii encephalitis in mice

Dincel, Gungor Cagdas; Atmaca, Hasan Tarik

Toxoplasma gondii (T. gondii) is a protozoan parasite with the potential of causing severe encephalitis among immunocompromised humans and animals. Our previous study showed that T. gondii induces high nitric oxide (NO) production, high glial activation (GFAP) and neurofilament expressions, leading to severe neurodegeneration in toxoplasma encephalitis (TE) in the central nervous system (CNS). The aim of this experimental study was to investigate ADAMTS-13 expression and apoptosis in CNS and to identify whether they have any correlation with toxoplasmosis neuropathology and neurodegeneration. Mice were infected with ME49 strain T. gondii and the levels of ADAMTS-13, caspase 3, caspase 8, caspase 9, TNFR1 and Bcl-xL expressions were examined in brain tissues by immunohistochemistry, during the development and establishment of chronic infections at 10, 30 and 60days post-infection. Results of the study revealed that the levels of ADAMTS-13 (P < 0.005), caspase 3 (P < 0.05), caspase 8 (P < 0.05), caspase 9 (P < 0.005) and TNFR1 (P < 0.05) expressions in the brain markedly increased while Bcl-xL expression decreased (P < 0.005). The most prominent finding from our study was that 10, 30 and 60days post-infection ADAMTS-13 increased significantly and this may play an important role in the regulation and protection of the blood-brain barrier integrity and CNS microenvironment in TE. These results also suggest that T. gondii-mediated apoptosis might play a pivotal role and a different type of role in the mechanism of neurodegeneration and neuropathology in the process of TE. Furthermore, expression of ADAMTS-13 might give an idea of the progress and is critical for diagnosis of this disease. To the best of the authors' knowledge, this is the first report on ADAMTS-13 expression in the CNS of T. gondii-infected mice.

Uçar, Meltem; Değer, Orhan; Gerigelmez, Asuman Yiğit; Cengiz, Sevil; Barlak, Yaşam; Ovalı, Ercüment

Purpose: To investigate the apoptosis-inducing capacity of dimethyl sulfoxide (DMSO) extracts of bee pollen and propolis in HL-60 Myeloid Cancer Cell Lines. Methods: DMSO extracts of pollen and propolis were incubated separately with HL-60 cells, and caspase-3 activity evaluated. In order to determine the cell cycle characteristics of HL-60 cells with and without extracts of pollen and propolis, the cells were analysed using flow cytometry. Results: The DMSO extract of propolis (0.5 mg/mL) increased apoptosis from undetectable levels to 60.1%, while maintaining cell viability. The DMSO extract of pollen (2 mg/ml) increased apoptosis from undetectable levels to 52.2% while decreasing cell viability by 62%. Caspase-3 activity in HL-60 cells incubated with DMSO extracts of pollen and propolis were 3.6-to 12-fold higher than in controls. Conclusion: Turkish pollen and propolis individually increase apoptosis and the activity of caspase-3 in HL-60 cells. This finding indicates that bee products may have beneficial effects in the treatment of cancer.

Increased expressions of eNOS and iNOS correlate with apoptosis of diabetic nephropathy in streptozotocin-induced type 1 diabetic rats; [Streptozotosin kaynaklı tip 1 diyabetik sıçanlarda meydana gelen nefropatilerde artan eNOS ve iNOS sunumlarının apoptozisle ilişkilendirilmesi]

    Dinçel, Güngör Çağdaş; Yildirim, Serkan

The present study was designed to evaluate the effects of high level of nitric oxide (NO) production and oxidative stress (OS) on nephropathology and to identify whether NO and OS have any correlation with apoptosis seen in diabetic kidney, elucidating the underlying mechanism(s) involved in the development of nephropathology in streptozotocin (STZ)-induced diabetic rats. Expression levels of caspase 3, caspase 9, endothelial nitric oxide synthase (eNOS), inducible nitric oxide synthase (iNOS), 8-hydroxy-2′-deoxyguanosine (8-OHdG), for the detection of oxidative damage to DNA, were examined in diabetic kidney tissues. Results of the study revealed that the levels of 8-OHdG (P<0.005), eNOS (P<0.005), iNOS (P<0.005), caspase 3 (P<0.005) and caspase 9 (P<0.005) were remarkably higher in diabetic kidney tissues than in controls. In addition, STZ-treated animals showed significant loss of body weight and renal enlargement. It was suggested that apoptosis, OS and increased NO levels are involved in the pathogenesis of diabetic nephropathy. The results also strongly suggested that STZ-induced apoptosis through activation of the intrinsic pathway and that might be most likely related to increased NO levels. Moreover, high NO production was not only mediated by eNOS but also by iNOS. Increased NO production may contribute to hyperfiltration and microalbuminuria in early diabetic nephropathy. Furthermore, expression of 8-OHdG might give an idea of the progress and may be essential as it has a diagnostic significance for this disease. In conclusion, we believe that eNOS and iNOS overexpressions induce diabetic nephropathy by mediating apoptosis in STZ-induced rats.

eNOS and iNOS trigger apoptosis in the brains of sheep and goats naturally infected with the border disease virus

    Dincel, Gungor Cagdas; Kul, Oguz

In this study, apoptotic and anti-apoptotic mechanisms and if present, which pathway to trigger the apoptosis in the brains of Border Disease Virus (BDV) infected lambs (n=10) and goat kids (n=5) were investigated. Briefly, apoptotic (caspase 3, caspase 9) and anti-apoptotic markers (Bcl-2), cytokine response (TNF-α, INF-γ), reactive gliosis and myelin loss were examined. eNOS, iNOS, caspase 9, caspase 3 and GFAP expressions were higher in BDV infected tissues compared to control animals (6 kids and 6 lambs) (p<0.05). Double immunoperoxidase test revelaed that TUNEL positive apoptotic cells showed significant association with increased eNOS-iNOS and iNOS-BDV expressions. However, no significant differences were found for TNFR1, TNF-α and INF-γ expressions in BD (p>0.05). There was a positive correlation between the intensity of myelin loss, GFAP activity and severity of infection. Inconclusion, as a novel finding, it is established that eNOS and iNOS overexpressions are coassociated with apoptosis in BDV infected neurons and neuroglia. The results also strongly suggested that BDV infected apoptotic cells mainly prefer the intrinsic pathway that might be most likely related to increased nitric oxide levels.

The effect of prenatal exposure to 900-MHz electromagnetic field on the 21-old-day rat testicle

Hanci, Hatice; Odaci, Ersan; Kaya, Haydar; Aliyazicioǧlu, Yüksel; Turan, Ibrahim; Demir, Selim; Çolakoǧlu, Serdar

The aim of this study was to investigate the effect of exposure to a 900-MHz electromagnetic field (EMF) in the prenatal term on the 21-old-day rat testicle. Pregnant rats were divided into control (CG) and EMF (EMFG) groups. EMFG was exposed to 900-MHz EMF during days 13-21 of pregnancy. Newborn CG rats were obtained from the CG and newborn EMFG (NEMFG) rats from the EMFG. Testicles were extracted at postnatal day 21. Lipid peroxidation and DNA oxidation levels, apoptotic index and histopathological damage scores were compared. NEMFG rats exhibited irregularities in seminiferous tubule basal membrane and epithelium, immature germ cells in the lumen, and a decreased diameter in seminiferous tubules and thickness of epithelium. Apoptotic index, lipid peroxidation and DNA oxidation were higher in NEMFG rats than in NCG. 21-day-old rat testicles exposed to 900-MHz EMF in the prenatal term may be adversely affected, and this effect persists after birth.
