This study analyzed the physicochemical characteristics, mineral composition, and antioxidant potential of fifteen floral honey samples collected from five districts of Gümüşhane, Türkiye. The average values for moisture (17.44 g/100 g), insoluble solids (0.04 g/100 g), conductivity (0.35 mS/cm), diastase activity (54.15 DN), free acidity (38.18 meq/kg), and fructose + glucose content (68.95 g/100 g) were mostly within the acceptable limits defined by national honey standards. Proline content ranged from 562.66 to 1466.15 mg/kg, while hydroxymethylfurfural (0.37–18.89 mg/kg) confirmed the freshness of all samples. Potassium, calcium, sodium, and magnesium were identified as the major mineral elements in the analyzed honey samples, with considerable variation observed among samples collected from different districts. Antioxidant activity (Total phenolic content (mg GAE/100 g): 9.21–98.25; SC₅₀ (mg/mL): 5.23–41.29) revealed a strong antioxidant potential of Gümüşhane honeys. This work provides the first detailed characterization of Gümüşhane honeys, confirming their high physicochemical quality, rich mineral profile, and notable antioxidant potential, and underscores their value as a distinctive regional product with functional and commercial significance.

Technological and nutritional aspects of fresh purslane (Portulaca oleracea L.) in ice cream production

    Hezer, Fatma; Gündoğdu, Engin

Purslane (Portulaca oleracea L.) is rich in ω-3 fatty acids, antioxidants, and minerals, and has notable neuroprotective, anti-inflammatory, antimicrobial, antidiabetic, antioxidant, anticancer, and antihypertensive properties. This research evaluated the effect of fresh purslane (FP) on the physicochemical and nutritional properties of ice cream, including α-linolenic acid (ALA; 18:3; ω-3), mineral content, and antioxidant properties, along with sensory characteristics. FP was added at 0, 5, 10, and 15% (w/w) levels. The addition of FP significantly increased the iron (Fe), calcium (Ca), copper (Cu), sodium (Na), oleic acid (OA), and ALA contents (P < 0.05). FP also increased the first dripping and complete melting times, while decreasing overrun and viscosity. Total antioxidant capacity (TAC) remained unchanged (P > 0.05), but total phenolic content (TPC) and 2,2-diphenyl-1-picryl-hydrazyl (DPPH) radical scavenging activity significantly increased (P < 0.01). Thirty-one volatile compounds were identified, with increases in octadecane, dodecane, and 2-hexenal concentrations due to FP addition. Although the addition of FP improved the ALA content and antioxidant properties, sensory results showed that FP over 5% (w/w) lowered taste and general acceptability scores. Thus, using 5% of FP in ice cream is optimal for enhancing nutritional properties.

This study investigates the antioxidant and enzyme inhibitory properties of Phlomis armeniaca, a perennial plant native to the eastern and southeastern regions of Turkey. Ethanol extracts of the plant were analyzed using various bioanalytical methods, including Fe3⁺-Fe2⁺ reducing power, Cupric Reducing Antioxidant Capacity, 1,1-diphenyl-2-picryl-hydrazyl (DPPH), and 2,2-azinobis 3-ethylbenzthiazoline-6-sulfonic acid (ABTS) radical scavenging activities, as well as total phenolic and flavonoid content assessments. The results showed that P. armeniaca is rich in phenolic (38.75 ± 0.59 µg gallic acid equivalents/mg extract) and flavonoid compounds (39.34 ± 4.08 µg quercetin equivalents/mg extract). The plant demonstrated significant inhibitory effects on acetylcholinesterase (half-maximal inhibitory concentration [IC50]: 45.00 ± 0.48 mg/mL) and butyrylcholinesterase (IC50: 84.51 ± 0.84 mg/mL) enzymes, indicating its potential use in treating neurodegenerative diseases such as Alzheimer's. The study also highlighted P. armeniaca’s high chlorogenic acid, salicylic acid, 4-dihydroxybenzoic acid, and quinic acid contents, supporting its antioxidant capacity. These findings suggest that P. armeniaca could be a valuable natural resource for developing therapeutic agents to manage oxidative stress and related conditions, warranting further pharmacological exploration.

Physicochemical Properties, Antioxidant Capacity and Sensory Acceptability of Instant Rosehip Teas Prepared by Spray-Drying and Freeze-Drying Methods

Baltaci, Cemalettin; Bostanci, Dilara Erkmen; Altintaş, Rahime; Dalkiran, Yusuf; Akdoğan, Arda; Okan, Onur T.

In this study, the physicochemical properties, antioxidant capacity, sensory properties, and phenolic and mineral profiles of instant rosehip teas prepared using spray-drying (SD) and freeze-drying (FD) were compared. The yield of instant tea produced using FD and SD was 23.75 and 21.25 g/100 g, respectively. The total color difference between the FD and SD tea was 50.74, with the SD sample exhibiting higher redness and yellowness. The FD tea was richer in ascorbic acid than SD tea (67.2 and 59.4 mg/100 g dry weight, DW, respectively). The mineral content ranged from 0.20 mg/kg DW (copper) to 2,837 mg/kg DW (potassium) in SD tea, and from 0.31 mg/kg DW (copper) to 3,083 mg/kg DW (potassium) in FD samples. The total phenolic content was 1,315 and 1,495 mg GAE/100 g DW of SD and FD samples, respectively. The antioxidant capacity of instant rosehip teas was determined as total antioxidant capacity, ferric reducing antioxidant power and DPPH radical scavenging activity. In all these assays, FD tea was found to have a higher antioxidant capacity. In the phenolic profile determined by liquid chromatography-tandem mass spectrometry (LC-MS/MS) technique, protocatechuic acid dominated in instant rosehip teas prepared using both methods. The main flavonoids identified were quercetin, quercitrin, hyperoside and luteolin 7-glucoside. Following the sensory analysis, the panelists have generally preferred the FD tea in terms of aroma and flavor, while opting for the SD tea in terms of appearance and color. In conclusion, although freeze-drying allowed obtaining a product with a higher antioxidant capacity and antioxidant content than spray-drying, both drying methods may be considered suitable for the production of instant rosehip teas and ensure obtaining a functional food product.

Phenolic Constituents, Antioxidant and Antimicrobial Activity and Clustering Analysis of Propolis Samples Based on PCA from Different Regions of Anatolia

Altuntaş, Ümit; Güzel, İsmail; Özçelik, Beraat

This study aimed to evaluate the biochemical composition and biological activity of propolis samples from different regions of Türkiye to characterize and classify 24 Anatolian propolis samples according to their geographical origin. Chemometric techniques, namely, principal component analysis (PCA) and a hierarchical clustering algorithm (HCA), were applied for the first time to all data, including antioxidant capacity, individual phenolic constituents, and the antimicrobial activity of propolis to reveal the possible clustering of Anatolian propolis samples according to their geographical origin. As a result, the total phenolic content (TPC) of the propolis samples varied from 16.73 to 125.83 mg gallic acid equivalent per gram (GAE/g) sample, while the number of total flavonoids varied from 57.98 to 327.38 mg quercetin equivalent per gram (QE/g) sample. The identified constituents of propolis were phenolic/aromatic acids (chlorogenic acid, caffeic acid, p-coumaric acid, ferulic acid, and trans-cinnamic acid), phenolic aldehyde (vanillin), and flavonoids (pinocembrin, kaempferol, pinobanksin, and apigenin). This study has shown that the application of the PCA chemometric method to the biochemical composition and biological activity of propolis allows for the successful clustering of Anatolian propolis samples from different regions of Türkiye, except for samples from the Black Sea region.

Acet, Tuba; Çorbacı, Cengiz; Özcan, Kadriye

In the paper, the phenolic compounds and biological activity of Corydalis oppositifolia DC. and Senecio cilicius Boiss., the endemic plants growing in Turkey, were investigated for the first time. The ethanol extract of C. oppositifolia exhibited more potent antimicrobial activity while the ethanol extract of S. cilicius exhibited the highest radical scavenging activity on ABTS (81.4±1.1a mg TE g−1 extract) and DPPH (23.4±0.2a mg TE g−1 extract). In C. oppositifolia, the highest total flavonoid and phenolic acid contents were found to be in the ethanol (94.2±3.5a mg QE g−1 extract) and methanol (320.5±3.3c mg GAE g−1 extract) extracts, respectively. In S. cilicius, the highest total flavonoid and phenolic acid contents were found to be in the methanol (56.9±0.9c mg QE g−1 extract) and ethanol (568.0±3.3a mg GAE g−1 extract) extracts, respectively. The ethanol extract of C. oppositifolia has the most prominent activity against α-amylase, AChE and BChE whereas the methanol extract of this plant displayed the highest inhibitory activity against α-glucosidase. HPLC analysis of the ethanol extracts revealed that the major flavonoids were kaempferol and quercetin whereas the major phenolic acids were protocatechuic acid, gallic acid, gentistic acid and 2,4-dihydroxybenzoic acid. The findings suggested that the plants and their extracts have the potential to be used in the treatment of diseases such as diabetes and Alzheimer's diseases.

An assessment of phenolic profiles, fatty acid compositions, and biological activities of two Helichrysum species: H. plicatum and H. chionophilum

Acet, Tuba; Ozcan, Kadriye; Zengin, Gokhan

In the present study, we aimed to search and compare the biological activities of the ethanol (EtOH), methanol (MeOH), and ethylacetate (EtOAc) solvent extracts of the flower, stem, and root parts of two Helichrysum plants (H. chionophilum (Hc) and H. plicatum subsp. plicatum (Hp)). The antioxidant properties were determined by using (2,2-diphenyl-1-picrylhydrazyl) (DPPH) and ABTS (2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) assays. The enzyme inhibitory effects of the extracts were investigated on butyrylcholinesterase (BChE), acetylcholinesterase (AChE), α-glucosidase, and α-amylase. Palmitic acid (C 16:0) was also determined as major fatty acids in the tested oils (31.21%–67.68%). In both plants, it was found that the EtOAc extracts of the flowers had a strong antioxidant and enzyme inhibitory effect. In conclusion, the results obtained in the present study showed that H. chionophilum and H. plicatum can be seen as a promising source for the natural bioactive compounds that can be used in therapeutic applications. Practical applications: The members of the genus Helichrysum have been widely taken for therapeutic purposes in traditional medicine as well as food. In this context, we investigated the chemical characterization and biological activities of two Helichrysum species extracts (H. chionophilum and H. plicatum subsp. plicatum). Antioxidant capacity, enzyme inhibition and anti-microbial effects were tested for biological activities. Chemical characterization was identified by high performance liquid chromatography (HPLC) (for phenolic) and gas chromatography-flame ioanization detector (GC-FID) (for fatty acids). Based on our findings, the species may be valuable for designing novel food products.

Demir, Selim; Turan, Ibrahim; Aliyazicioglu, Yuksel

Rhododendron luteum belongs to the genus Rhododendron, members of which are frequently used in folk medicine. Various studies have investigated the cytotoxic effect of different Rhododendron species, but there have been limited studies of the cytotoxic effect of R. luteum. The aim of this study was to investigate the antioxidant properties and cytotoxic effect of R. luteum leaf extract on the human cancer cells. The total phenolic content (TPC), total flavonoid content (TFC), and reducing power of the extract were evaluated using spectrophotometric procedures. The cytotoxic effect of the extract on five cancers (human breast, colon, lung, prostate, and liver carcinoma) and human fibroblast cells was determined using the MTT assay. TPC and reducing power values of extract were found 173.2 +/- 3.97 mg gallic acid equivalents, and 629.5 +/- 3.49 mg trolox equivalents per g sample, respectively. R. luteum leaf extract exhibited selective cytotoxicity especially against colon (1.9 fold) and liver (2.2 fold) cancer cells compared to normal fibroblast cells. This is the first study to reveal the cytotoxic effect of R. luteum leaf extract. Further studies are now needed to identify the cytotoxic molecules in the extract and their mechanisms.

Accumulation of phenolics in natural and micropropagated plantlets of Thymus pseudopulegioides Klokov & Des.-Shost. with their antioxidant potentials

Günaydin, Mustafa; Laghari, Abdul Hafeez; Bektas, Ersan; Sökmen, Munevver; Sökmen, Atalay

Thymus pseudopulegioides plantlets were propagated in vitro via direct organogenesis by using Murashige and Skoog (MS) media containing kinetin, thidiazuron, and 6-benzyladenine (BA) individually. Methanol extracts obtained both from plantlets and wild plants were analyzed for their total phenolics and flavonoid contents, then quantified by HPLC. The highest total phenolic (8.83 mg/g as gallic acid equivalent) and total flavonoid (0.92 mg/mL as rutin equivalent) values were from the MS media supplemented with 1.0 mg/L kinetin and 0.5 mg/L BA, respectively. The plantlets grown in those media also showed remarkable antioxidant activities with an IC50 value of 4.77 mu g/mL in DPPH and 100% inhibition in beta-carotene assays, respectively. HPLC analysis proved the production of protocatechuic, caffeic, vanillic, rosmarinic, ferulic, and o-coumaric acids and rutin. Rosmarinic acid production was predominant in natural samples (115.2 mg/100 g dry weight), while the aforesaid phenolic acids were prevalent in plantlets grown on MS media supplemented with KIN or BA at various concentrations. Rutin production was the highest (50.74 mg/100 g dry weight) in the plantlets grown on MS medium containing kinetin (1.0 mg/L). As an economically important chemical, rosmarinic acid was selected as the target chemical and a novel method was introduced to achieve its selective isolation.
