This study reports the green synthesis of silver (PFAgNPs) and copper (PFCuNPs) nanoparticles using an aqueous extract of pointed cherry laurel (Prunus laurocerasus L.) fruit (PF-E) and explores their applicability in food safety. The NPs were characterized via UV–Vis spectroscopy, FT-IR, XRD, SEM, EDX, and TEM confirming their successful formation and morphology. Total phenolic content (TPC), flavonoid content (TFC), and antioxidant capacity (TAC) were measured for both PF-E and the synthesized NPs. While PFCuNPs exhibited higher TPC, PFAgNPs showed superior TFC and TAC. Notably, antioxidant assays (DPPH, ABTS, FRAP) indicated stronger radical scavenging activity for PFCuNPs. Antibacterial tests against Escherichia coli, Enterococcus faecalis, Staphylococcus aureus, and Pseudomonas aeruginosa revealed broad-spectrum activity of PFAgNPs, with the highest inhibition against E. coli (19.4 ± 0.3 mm), whereas PFCuNPs showed dose-dependent efficacy, strongest against E. faecalis (9.6 ± 0.2 mm). LC–MS/MS analysis of the PF-E extract revealed quinic acid (63.42 mg/g), chlorogenic acid (2.38 mg/g), protocatechuic acid (0.07 mg/g), protocatechuic aldehyde (0.04 mg/g), hesperidin (0.03 mg/g), and naringenin (0.02 mg/g) as major components. The adsorption efficiencies of the synthesized PFAgNPs and PFCuNPs were evaluated against the toxic food contaminants patulin and HMF using both standard aqueous solutions and real food systems (apple juice). In standard aqueous solutions, PFAgNPs achieved removal efficiencies of 70.96% for patulin and 52.22% for HMF, while PFCuNPs showed 61.08% and 36.38%, respectively, at 5 g/L after 240 min. In real food systems, the adsorption efficiencies decreased, with PFAgNPs and PFCuNPs removing 34.24% and 32.90% of patulin, and 39.09% and 31.87% of HMF, respectively. The adsorption kinetics of HMF and patulin onto PFCuNP and PFAgNP were found to conform to both the pseudo-first-order and pseudo-second-order kinetic models for aqueous solutions as well as real samples. These findings confirm that the biosynthesized NPs offer dual functionality as antibacterial agents and toxin adsorbents, presenting a green, sustainable approach to food safety enhancement.

This study presents a combined evaluation of patulin contamination by integrating fungal identification, assessment of toxin production, and the feasibility of obtaining high-purity patulin from biological sources. Fungal isolates recovered from naturally decayed apples collected from different agricultural environments in Türkiye were identified using MALDI-TOF MS and morphological characteristics, and were assigned to Penicillium expansum at a probable species level. The patulin production potential of the isolates was evaluated by inoculating them onto red, yellow, and green apple matrices under controlled laboratory conditions over an extended incubation period. Quantitative analysis using HPLC–UV, supported by LC–MS/MS confirmation, revealed substantial variability in patulin levels (7.58–49.80 mg/kg) depending on both isolate and apple matrix. Higher patulin levels were generally observed in yellow apple tissues, whereas lower levels were detected in red apples, suggesting a possible influence of matrix-related compositional factors. In addition, patulin was isolated and enriched using column chromatography with TLC-guided fractionation, yielding a high-purity compound (98.8%) with low moisture content (0.21%). Structural identity was confirmed by LC–MS/MS analysis. Overall, the findings indicate that patulin production is influenced by strain-level variability and matrix-related factors. Furthermore, the study demonstrates the feasibility of producing high-purity patulin suitable for in-house analytical applications. However, the produced material should not be considered a certified reference material, and further validation would be required for broader analytical use.
- MakaleAçık Erişim2026Green-synthesis of iron nanoparticles using wild cherry laurel (Prunus laurocerasus L.) fruit extract and their patulin and HMF adsorption capacities along with biological activities
Kobya, Havva Nur; Gül, Esra; Karpuz, Ömer; Baltacı, Cemalettin
In this study, iron nanoparticles (YBFeNPs) were green-synthesized using the aqueous extract (YB-E) of wild cherry laurel (Prunus laurocerasus L.) fruit to evaluate their potential applications in food safety. The synthesized YBFeNPs were investigated through Fourier-transform infrared spectroscopy, X-ray diffraction, scanning electron microscopy, transmission electron microscopy, and energy-dispersive X-ray spectroscopy analyses. YBFeNPs showed higher total phenolic content, total flavonoid content, and total antioxidant capacity compared to the original extract, suggesting that plant metabolites enhance NP bioactivity. Antioxidant capacity was further validated by DPPH, ABTS, and FRAP assays, which demonstrated strong radical scavenging and reducing power. The antibacterial activity of YBFeNPs was assessed against Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, and Enterococcus faecalis, revealing concentration-dependent effects with the highest inhibition zone of 14.5 ± 0.4 mm for E. coli. The chemical composition of the YB-E extract was analyzed using liquid chromatography-tandem mass spectrometry, identifying quinic acid, a polyol, and phenolic compounds such as chlorogenic acid (1.45 mg/g) and naringenin (0.02 mg/g). Additionally, the adsorption capacities of YBFeNPs were investigated against patulin and hydroxymethylfurfural (HMF), toxic compounds posing significant threats to food safety. The NPs demonstrated effective adsorption of both patulin and HMF, achieving maximum adsorption rates of 61.05% and 46.12%, respectively, at a concentration of 5.0 g/L after 240 min.

Background/Objectives: Radiotherapy (RT) induces oxidative stress and structural damage in solid tissues, including the liver. This study aimed to investigate the histological and immunohistochemical effects of dexmedetomidine (DEX) and amifostine on their potential protective and regenerative properties against liver injury induced by radiation therapy. Methods: This study consisted of five randomized groups: control, RT, RT-D100, RT-D200, and RT-A (Amifostine). A total of 100 mu g/kg DEX, 200 mu g/kg DEX, and 200 mu g/kg amifostine were administered before radiotherapy as per the experimental design. After RT, liver specimens were analyzed for histological alterations, including periportal and perisinusoidal fibrosis, vacuolization, and pyknotic nuclei. Furthermore, immunohistochemistry investigations were conducted to evaluate apoptosis, mitosis, oxidative stress, and neural regeneration in non-neuronal liver tissue following radiotherapy and subsequent treatment. Results: The control group's liver tissue exhibited standard histological architecture, whereas the RT group displayed severe cellular degeneration, periportal and perisinusoidal fibrosis, cytoplasmic vacuolization, and an increase in pyknotic nuclei. The apoptotic index was markedly reduced in the RT-D100 and RT-D200 groups relative to the RT group. Furthermore, caspase-3 immunoactivity was negligible in the control group, while a significant increase was observed in the RT group. The administration of amifostine significantly increased GAP-43 levels. Conclusions: DEX mitigates RT-induced hepatic injury chiefly through antioxidant and anti-apoptotic pathways, whereas amifostine promotes hepatic regeneration by modulating GAP-43.

Objective: This study aimed to investigate the potential splenic tissue damage induced by radiotherapy (RT) and the potential protective effect of different doses of dexmedetomidine on this damage at the histopathological, immunohistochemical, and biochemical levels. Materials and Methods: In our study, Sprague Dawley rats were randomly divided into four groups: Control, Radiotherapy (RT; 8 Gy), RT + Dexmedetomidine 100 mu g/kg (RT-D100), and RT + Dexmedetomidine 200 mu g/kg (RT-D200). A single dose of 8 Gy radiotherapy was administered to each RT group. Spleen tissues were examined histologically with hematoxylin-eosin and immunohistochemically with anti-Caspase-3, anti-TGF-beta 1, and anti-TGF-beta 3 using light microscopy. TBARS and total thiol levels were also analyzed to assess oxidative stress and antioxidant capacity. Results: Histopathological results showed a significant decrease in white pulp diameter, decreased cellular density, and increased congestion in the red pulp in the RT group. Significant fibrosis, sinusoidal dilatation, vacuolization, and amyloid deposition were detected in the white pulp in the RT group. Regarding anti-caspase-3 immunoreactivity, strong positivity increased in the red pulp in the RT group, while a significant increase was observed in the white pulp in both the RT-D100 and RT groups. While the proportion of TGF-beta 1 immunopositive cells did not change significantly in the RT group, they increased significantly in both dexmedetomidine groups (especially RT-D200). TGF-beta 3 expression increased significantly only in the RT-D100 group. In biochemical analyses, TBARS levels increased significantly in the RT-D100 group. Total thiol levels decreased in the RT group and increased in the dexmedetomidine-treated groups. Conclusions: While RT caused histopathological damage and increased oxidative stress in spleen tissue, dexmedetomidine reduced this damage in a dose-dependent manner. The different immunohistochemical profiles of TGF-beta 1 and TGF-beta 3 suggest that these cytokines may have different functions in the spleen. 100 mu g/kg dexmedetomidine stimulates a regenerative response through TGF-beta 3, while 200 mu g/kg dexmedetomidine may provide immune regulation and antioxidative defense through TGF-beta 1.

This study aimed to investigate the utilization of sesame, sunflower seed, almond, peanut, and coconut as condiments for the enrichment of mulberry leather, known locally as mulberry pestil. The investigation involved a comprehensive analysis encompassing proximate composition, mineral content, color, physicochemical, and sensory properties. The studied parameters of enriched samples were compared with those of the plain sample. Employing multivariate statistical techniques enabled the differentiation of the enriched leather samples. Consequently, the leather samples were categorized into three groups by the principal component analysis according to their physicochemical and chemical attributes. Strong positive correlations were observed among the studied parameters, as indicated by Spearman's rank correlation coefficients. The physicochemical and chemical properties of enriched samples exhibited substantial variability. The enriched products had abundant calcium, potassium, magnesium, iron, and zinc levels. These findings revealed that incorporating these condiments enhanced the nutritional values of leather samples, especially in terms of minerals. 100 g of the enriched mulberry leathers had 68.0-70.7 g of carbohydrate, 4.6-5.7 g of protein, 3.6-5.9 g of fat, and 336-359 kcal of energy value. The enriched products might be proposed to provide nutrients and energy in emergencies, especially for adolescents and people affected by disasters.

The efficient extraction of bioactive molecules from medicinal plants is strongly related to the solvents and methods used. This study evaluated the impact of solvent selection and extraction methods on the bioactive potential of Rosa canina seed extracts, focusing on antioxidant, antimicrobial, and α-amylase inhibitory activities. Four solvents —dimethyl sulfoxide (DMSO), ethanol-water (80:20), ethyl acetate, and acetonitrile— were paired with maceration and ultrasonic techniques to generate eight extracts (DMM, DMU, ESM, ESU, EAM, EAU, ACM, ACS). Antioxidant activity was assessed via total phenolic (TPC) and flavonoid (TFC) content, FRAP, DPPH, and ABTS assays. Antimicrobial efficacy was tested against seven microorganisms (Gram+, Gram−, and yeast) using well diffusion and MIC methods, while α-amylase inhibition was quantified via IC50. The ethyl acetate ultrasonic extract (EAU) exhibited the highest TPC (63.86 mg GAE/g) and TFC (210.23 mg QE/g), along with superior antioxidant activity across all assays (P < 0.05). Ethyl acetate maceration (EAM) ranked second, underscoring ethyl acetate’s efficacy. Acetonitrile maceration (ACM) demonstrated the strongest α-amylase inhibition (IC50 = 20.46 mg/mL). All extracts showed notable antimicrobial activity against Aeromonas hydrophila and Klebsiella pneumoniae, though efficacy varied for other strains. DMSO-based extracts (DMM/DMU) consistently underperformed. These findings highlight ethyl acetate (EAU/EAM) as optimal for antioxidant-rich extracts and ACM for enzyme inhibition, positioning Rosa canina seeds as a promising source of natural bioactive compounds for functional food and therapeutic applications.
- MakaleAçık Erişim2025A strategy for the one-pot direct production of 5-hydroxymethylfurfural (HMF) from sucrose using organic weak acid in situ as a catalyst
Gül, Abdulkadir; Tanyıldızı, Muhammet Şaban
5-Hydroxymethylfurfural (HMF), which can be produced in high yields by acid-catalyzed dehydration of carbohydrates, is used as a feedstock in biofuel and biopolymer production. However, these acidic catalysts are usually added externally, which increases HMF production costs. Furthermore, other important parameters for high yields of HMF production are raw material selection, reaction medium, and separation-purification processes. The aim of this study is to produce HMF in one-pot without the addition of any exogenous catalyst by evaluating the fructose remaining in the production medium in which the glucose from sucrose hydrolysis is microbially fermented into gluconic acid (GA). HMF production from fructose in a biphasic system consisting of a fermentation liquid containing GA and fructose and 2-Methyltetrahydrofuran (2-MeTHF) was optimized using Central Composite Design (CCD). The variables examined included reaction time, reaction temperature, solvent type, salt type, and organic/aqueous phase ratio. In the study, by chelating GA and calcium ions, the low degree of ionization of GA was increased, and HMF was produced in high yields without the addition of a catalyst. Under optimum conditions, using 2-MeTHF/ fermentation liquid (6:1) and 7.56 wt% CaCl2, the yield of HMF produced in pressurized (hydrothermal) vessels at about 147 °C for 158 min reached 77.6 % of the theoretical yield. The amount of GA in the production medium was 92.64 % preserved from degradation. GA and HMF in different phases of the biphasic system were purified from the production medium in high yields. Furthermore, 2-MeTHF, which constitutes the organic phase of the biphasic system, was recovered at approximately 95 % by vacuum evaporation. As a result, HMF production from sucrose, which is a more economical and abundant raw material compared to glucose and fructose, was realized in one-pot without the addition of any catalyst.

Backgrounds: Approximately 18 million individuals were diagnosed with cancer in 2018. The rate is predicted to exceed 22 million by 2030. Radiotherapy is an essential part of cancer therapy, with well documented local and systemic side effects, including oxidative stress and apoptosis. Kidney tissues are also exposed to the deleterious effects of radiotherapy, resulting in acute or chronic kidney function impairment. This study compared the effects of the potent selective α2-adrenoreceptor agonist dexmedetomidine and amifostine on oxidative stress and apoptosis in kidney damage induced by x-irradiation in rats. Methods: Forty Sprague Dawley rats were assigned into five groups: control, x-irradiation, x-irradiation + amifostine, x-irradiation + dexmedetomidine 100 µg/kg, and X-ray irradiation + dexmedetomidine 200 µg/kg. Results: Necrotic tubules and degenerative Bowman’s capsules were present in the x-irradiation group. An increase was determined in malondialdehyde (MDA), Cleaved Caspase-3, and 8-OHdG levels compared to the control group (p ≤ 0.05). In contrast, there was a decrease in necrotic tubules, degenerative Bowman’s capsules, and the levels of MDA, Cleaved Caspase-3, and 8-OHdG in the amifostine and dexmedetomidine 100 µg/kg and 200 µg/kg treatment groups (p ≤ 0.05). Conclusions: Alpha 2 adrenergic receptor agonists exhibit protective effects against kidney injury induced in association with x-irradiation by reducing oxidative stress and apoptosis.

Iron oxide (BHFeNP) and silver nanoparticles (BHAgNP) were produced using a green synthesis method through the extract of buckwheat husk (BH) waste to remove dye pollutants from water bodies in this study. The BHFeNP and BHAgNP were characterized by TEM, SEM, XRD, EDX, and FTIR techniques. Then, the methylene blue (MB) removal performance of the synthesized nanoparticles (NPs) was investigated. MB adsorption experiments showed that BHFeNPs were more dependent on temperature, adsorbent amount, and pH, while BHAgNPs notably depended on adsorbent amount, initial MB concentration, and pH. BHAgNPs had higher adsorption rates than BHFeNPs, and the Langmuir isotherm was more suitable for the adsorption process. BHAgNP and BHFeNP were shown to have the maximum adsorption capacities of 96.3 and 53.4 mg/g at pH 8 and 25 °C, respectively, in the presence of 0.25 g/L NP. Kinetic and thermodynamic calculations revealed that MB adsorption was spontaneously endothermic and supported the pseudo-second-order kinetic model. The antibacterial potentials of BHFeNPs and BHAgNPs were investigated against two gram-negative (Escherichia coli and Pseudomonas aeruginosa) and two gram-positive (Enterococcus faecalis and Staphylococcus aureus) bacterial strains. BHAgNPs exhibited a higher antibacterial activity against the strains used. In cytotoxicity studies with ARPE-19, A549, HeLa, and DU-145 cell lines, it was observed that BHAgNP had a greater cell growth inhibition effect than BHFeNP and BH extract. Examining the morphological changes in the cells exhibited that BHAgNP significantly inhibited A549 and ARPE-19. Reusability studies of the generated NPs revealed MB removal capabilities of 92.5% and above after the fifth stage. The present study demonstrates that BHAgNP and BHFeNPs are viable nanoadsorbents for the elimination of environmental pollution due to their excellent removal performances and stable adsorbent characteristics.
