Öz, Mehmet; Fidan, Muhammed Said; Baltacı, Cemalettin; Akmeşe, Osman; Sefalı, Abdurrahman; Aygül, İmdat
Enzyme inhibition activities, phenolic compounds, antioxidant activities, bioactive compounds, antimicrobial activities, and chemical components of essential oil and methanol extracts obtained from the aerial parts of S. cretica subsp. anatolica were investigated. The main phenolic compounds of aerial parts were catechin, oleuropein, and epicatechin. The determined enzyme inhibitor activities highlight the potential of S. cretica subsp. anatolica as a source of bioactive compounds, particularly for carbonic anhydrase and cholinesterase inhibition. The essential oil and methanol extract exhibited remarkable activities against CA-II, AChE, and BChE, although they were less potent than standard inhibitors. The essential oils generally showed stronger antimicrobial activity compared to the 30% methanol extracts across most bacterial and fungal strains, as evidenced by minimum lethal concentration (MLC) and lower minimum inhibitory concentration (MIC) values and larger inhibition zones. Chloramphenicol used alone exhibited the highest antimicrobial efficacy, with the lowest MIC and MLC values and the largest inhibition zones. The essential oils of S. cretica subsp. anatolica were determined as esters, oxygenated sesquiterpenes, and aldehydes in aerial parts. The main components were found to be hexahydrofarnesyl acetone in the aerial parts.

Gül, Abdulkadir
In this study, beeswax waste, a low-value by-product of apiculture, was converted into a liquid fraction with bioactive potential through pyrolysis-assisted thermochemical transformation. The LC–MS/MS-based chemical profile, total phenolic content (TPC), total flavonoid content (TFC), antioxidant responses, and antibacterial, antidiabetic, anti-inflammatory, and anticholinesterase activities of beeswax waste pyrolysis liquid (BWPL) were investigated. The TPC and TFC values were determined as 95.30 ± 1.13 mg GAE/L and 51.67 ± 1.12 mg QE/L, respectively. LC–MS/MS analysis revealed benzoic acid and vanillin as the predominant compounds, along with various phenolic acids and flavonoid derivatives. BWPL showed 51.83 ± 1.51% inhibition in the ABTS•⁺ assay and 9.71 ± 0.28% inhibition in the DPPH assay. It also exhibited measurable redox capacity in Fe3+ and Cu2+ reducing power assays. In antibacterial assays, BWPL produced inhibition zones against all tested strains at 1 mg/mL and against E. coli and E. faecalis at 500 µg/mL. The IC50 values of BWPL for 5-LOX and α-glucosidase were 38.17 µg/mL and 1503.0 µg/mL, respectively. In addition, the IC50 values for AChE and BChE were calculated as 112.38 ± 0.87 mg/mL and 13.01 ± 0.42 mg/mL, respectively. PCA and correlation analyses indicated the multidimensional relationships between the LC–MS/MS-based chemical profile and biological activities. Overall, the pyrolytic conversion of beeswax waste into a chemically complex liquid fraction provides a preliminary scientific basis for waste valorization and for further investigation of pyrolysis-derived bioactive fractions.

The growing interest in natural products, particularly as food additives and pharmaceutical raw materials, is making research into discovering new sources especially important. Erodium cedrorum subsp. salmoneum (family Geraniaceae) is used traditionally to cure many ailments like diabetes and gastro-intestinal diseases. Within this background, the current study was designed to investigate, for the first time, the chemical composition, antioxidant and enzyme inhibitory poroperties of different extracts from the aerial parts and roots of E. cedrorum. Results revealed that the aqueous extract of the aerial parts accumulated the highest total phenolic (85.35 mg GAE/g) and flavonoids (28.65 mg RE/g) contents. A total of 61 and 45 compounds, belonging mainly to phenolic acids and flavonoids, were tentatively identified in the aerial parts and roots extracts respectively. The aqueous extract from the aerial parts exerted the highest antiradical, ions reducing and chelation properties. The EtOH and 70% EtOH extracts of the two organs recorded the best anti-acetylcholinesterase activity (2.18 -2.41 mg GALAE/g; p >= 0.05) while those of the roots recorded respectively the best anti-butyrylcholinesterase and antityrosinase activities. The most potent antibacterial activity was exhibited by the 70% EtOH extracts from aerial parts and the pure EtOH extracts from roots. In contrast, ethyl acetate extracts from both plant parts showed only marginal activity. Among the tested strains, the highest susceptibility was observed against C. sporogenes ATCC 19404, C. bifermentans ATCC 638, S. aureus ATCC 6538, S. aureus ATCC 43300 (MRSA), C. diphtheriae PCM 506, and H. pylori ATCC 43504. The extracts were also examined for their anti-cancer potential on the HeLa, AGS and SKOV-3 cell lines, as well as a non-cancerous cell line. The ethyl acetate extract of the roots exhibited the most potent anti-cancer activity, with the lowest IC50 values. In addition, selected top-ranked complexes against AChE, alpha-amylase, representative bacterial and oncogenic targets were further evaluated by molecular docking and 100 ns molecular dynamics (MD) simulations, indicating overall conformational stability and persistent binding. In conclusion, these results position E. cedrorum as a valuable natural source of antioxidant and enzyme-inhibitory agents, meriting further exploration for drug development purposes.

This study aimed to investigate, for the first time, the phytochemical composition, antioxidant, antimicrobial, and enzyme inhibitory properties of the methanolic extract of Gelasia sericea (Aucher ex DC.) Zaika, Sukhor. & N.Kilian. The extract demonstrated a moderate yield (6.55 g/100 g dry plant material) and was found to be rich in phenolic acids, particularly ferulic acid (17182.97 μg g-1), followed by caffeic acid, protocatechuic acid, and p-coumaric acid. Antioxidant analyses revealed high total phenolic (388.86 mg GA eq./100 g), total flavonoid (74.03 mg QE eq./100 g), and total antioxidant capacity (503.33 mg AA eq./100 g). The extract exhibited remarkable ABTS radical scavenging activity (95.99%, IC50 = 37.59 mg ml-1) and considerable ferric-ion reducing power (974.78 mg FeSO4 eq./100 g). Antimicrobial assays indicated moderate inhibitory effects, with stronger activity against Gram-positive bacteria (Staphylococcus aureus, Enterococcus faecalis) and the yeast Candida albicans, whereas Gram-negative bacteria were less susceptible. The extract’s MIC values ranged between 0.20-3.25 mg ml-1, and MLC values between 0.41-6.50 mg ml-1, which were higher than those of standard antibiotics. Enzyme inhibition assays revealed moderate activity against carbonic anhydrase II (IC50 = 0.0136 µg ml-1) and weak inhibition of cholinesterases and α-glucosidase, while α-amylase inhibition was relatively more pronounced (IC50 = 13350.00 µg ml-1). Overall, these findings highlight G. sericea as a valuable natural source of phenolic acids and antioxidants, with promising potential for applications in functional foods, nutraceuticals, and phytopharmaceuticals. However, the relatively limited antimicrobial and enzyme inhibitory activities suggest the need for further studies on bioavailability, in vivo efficacy, and compound isolation to optimize its practical applications.

Cordyceps sinensis (CS), a traditional medicinal fungus, is a rich source of bioactive metabolites with pharmacological potential. This study investigated the inhibitory effects of CS mycelium on human carbonic anhydrase isoenzymes I and II (hCA I and hCA II) and acetylcholinesterase (AChE), as well as its antioxidant properties. Enzyme inhibition was assessed using spectrophotometric methods, whereas antioxidant capacity was evaluated by DPPH, ABTS, CUPRAC, FRAP, and Fe3⁺-reducing assays. LC-MS/MS analysis identified key secondary metabolites, including apigenin, apigenin-7-glucoside, (+)-taxifolin, dihydrokaempferol, p-coumaric acid, salicylic acid, and L-theanine, with apigenin as the major constituent. CS strongly inhibited hCA I (IC50 = 1.11 µg/mL) and hCA II (IC50 = 1.77 µg/mL), exceeding the potency of acetazolamide, and also showed relevant AChE inhibition (IC50 = 8.00 µg/mL), comparable to tacrine. Antioxidant assays revealed moderate DPPH activity but strong ABTS and CUPRAC reducing capacities, indicating the structural contributions of flavonoids and phenolic acids. These findings suggest that CS possess dual bioactivity through enzyme inhibition and antioxidant effects, highlighting its therapeutic potential against neurodegenerative and oxidative stress–related diseases.

Balci, Neslihan; Aslan, Kubra; Erturk, Adem; Gulcin, Ilhami
This study evaluates the antioxidant activity, enzyme inhibition potential, and phenolic composition of ethanolic Siran propolis extract. Antioxidant capacity was measured using reducing power (CUPRAC, FRAP, and Fe3+-reducing) and radical scavenging (DPPH·, ABTS·+, and DMPD·+) assays. The extract showed strong reducing ability, surpassing standard antioxidants in CUPRAC and FRAP assays, correlating with high ferulic acid (40.40 mg/g) and p-coumaric acid (88.60 mg/g) levels identified by LC–MS/MS. While it demonstrated notable DPPH· scavenging (IC50: 30.14 μg/mL), ABTS·+ activity was weaker (IC50: 272.44 μg/mL). Enzyme inhibition assays indicated effective inhibition of α-glycosidase, AChE (IC50: 2.15 μg/mL), hCA I, and hCA II. Notably, AChE inhibition was stronger than the standard tacrine inhibitor (IC50: 8.82 μg/mL), suggesting neuroprotective potential. The observed bioactivities are attributed to synergistic effects of phenolic acids and flavonoids, including quercetin, luteolin, taxifolin, and resveratrol. These findings highlight Siran propolis as a promising natural antioxidant and enzyme inhibitor source with potential therapeutic applications and food formulations.

Fidan, Muhammed Said; Baltacı, Cemalettin; Akmeşe, Osman; Sefalı, Abdurrahman; Aygül, İmdat; Öz, Mehmet
In this study, essential oil from the aerial parts of Gelasia sericea (Aucher ex DC.) Zaika, Sukhor. & N. Kilian was extracted via hydrodistillation using a Clevenger apparatus. GC-MS/FID analysis determined aldehydes (28.31%) as the dominant group, with eucalyptol (8.71%) as the main constituent. Antioxidant activities were exhibited using DPPH, ABTS, and FRAP assays, with the essential oil demonstrating relatively higher antioxidant properties. The essential oil also showed higher levels of bioactive components, including total antioxidant capacity (160.26 mg AA eq./100 mL), total phenolic content (21.57 mg GA eq./100 mL), and total flavonoid content (14.85 mg QE eq./100 mL). While G. sericea essential oil demonstrated some antioxidant activity, it was significantly lower compared to the reference standard ascorbic acid (AA). The antimicrobial results showed relatively higher antimicrobial activity compared to most bacterial and fungal strains, as evidenced by lower MIC and MBC values and larger inhibition zones. Nevertheless, chloramphenicol (CH) and nalidixic acid (NA) continually showed superior antimicrobial efficacy, with the lowest MIC and MBC values and the largest inhibition zones. The essential oil also demonstrated potent enzyme inhibitory activity, especially against CA-II, AChE, and BChE, with the lowest IC50 value observed for CA-II, highlighting its potential as a potent enzyme inhibitor. However, the essential oil exhibited comparatively greater inhibition against AChE, BChE and α-amylase but was less effective overall. These findings suggest that the G. sericea essential oil has important antioxidant potential and enzyme inhibitory activity, while the essential oil shows varying efficacy across different enzymes.
- MakaleAçık Erişim2025Inhibition Effect of Diosmin Flavanoid on Some Metabolic Enzymes and Investigation of Antioxidant Properties
Balci, Neslihan
Diosmin is a natural flavonoid obtained by dehydrating flavanone glycoside hesperidin. This study evaluated the inhibition effects of diosmin flavonoid on human carbonic anhydrases (hCA I and hCA II) and acetylcholinesterase (AChE) enzymes, which are linked to various health issues, and compared the results with standard drugs. The antioxidant potential was assessed using five assays, namely DPPH, ABTS, FRAP, CUPRAC and Fe3+ reducing assays. The results determined that diosmin didn’t exhibit strong antioxidant activity. Additionally, the inhibition effects of diosmin on hCA I, hCA II, and AChE enzymes were examined. It effectively inhibited hCA I and hCA II enzymes with Ki values of 3.84±0.63 nM and 15.64±1.90 nM, respectively (Table 1). For comparison, the Ki values of the standard drug AZA were 1.44 ± 0.32 nM for hCA I and 3.66 ± 0.25 nM for hCA II. On the other hand, diosmin effectively inhibited AChE enzymes with Ki values of 9.31±2.40 nM. The Ki values of the standard drug Tacrine was 6.22±1.95 nM for AChE. These findings suggest that diosmin may serve as a selective enzyme inhibitor, particularly for hCA I, but has limited antioxidant capacity. Diosmin exhibited relatively weak antioxidant activity compared to standard antioxidants.

Abstract: In this study, the inhibition potential of 3- and 4-arylcoumarin derivatives on Theileria annulata enolase (TaENO) was assessed for the first time in the literature. Firstly, protein stabilization analyses of TaENO were performed and it was found that the enzyme remains stable with the addition of 6 M ethylene glycol at + 4 °C. Inhibitor screening analyses were carried out using 25 coumarin derivatives on highly purified TaENO (> 95%), and four coumarin derivatives [4-(3,4-dimethoxyphenyl)-6,7-dihydroxy-2H-chromen-2-one (C8); 4-(3,4-dihydroxyphenyl)-7,8 dihydroxy-2H-chromen-2-one (C9); 4-(3,4-dihydroxyphenyl)-6,7-dihydroxy-2H-chromen-2 one (C21); and 3-(3,4-dihydroxyphenyl)-7,8-dihydroxy-2H-chromen-2-one (C23)] showed the highest inhibitory effects with the IC50 values of 10.450, 13.170, 8.871 and 10.863 µM, respectively. The kinetic results indicated that these compounds inhibited the enzyme by uncompetitive inhibition. In addition, the successful binding of the most potent inhibitor (C21) into TaENO was confirmed by using MALDI-TOF mass spectrophotometry. Molecular docking analyses have predicted that C8 and C21 coumarin derivatives which showed high inhibitory effects on TaENO were interacted with high affinity to the potential regions out of the active site. Taken together, these coumarin derivatives (C8, C9, C21 and C23) are first known potent, nonsubstrate, uncompetitive inhibitors of TaENO and these results will facilitate further in vitro and in vivo analysis toward structure-based drug design studies. Eugenol was used as starting material to obtain some phenolic compounds. The synthesis of these phenolic compounds was performed in a two-step procedure. The structures of the formed products (novel eugenol derivatives 1–6) have been determined on the basis of NMR spectroscopy and other spectroscopic methods. The compounds were tested in terms of carbonic anhydrase (CA) inhibition potency. Carbonic anhydrases (CAs, EC 4.2.1.1) are metalloenzymes, which catalyse the reaction between carbon dioxide (CO2) and water (H2O), to generate bicarbonate (HCO3−) and protons (H+). CO2, HCO3− and H+ are essential molecules and ions for many important physiologic processes occurring in all living organisms. Acetylcholinesterase (AChE, E.C.3.1.1.7) is found in high concentrations in the red blood cells and brain. Novel eugenol derivatives (1–6) were tested for the inhibition of two cytosolic CA isoforms I, and II (hCA I, and II) and AChE. These compounds demonstrated effective inhibitory profiles with Ki values in ranging of 113.48-738.69 nM against hCA I, 92.35-530.81 nM against hCA II, and 90.10-379.57 nM against AChE, respectively. On the other hand, acetazolamide clinically used as CA inhibitor, shoed Ki value of 594.11 nM against hCA I, and 120.68 nM against hCA II, respectively. Also, AChE was inhibited by tacrine as an AChE inhibitor at the 71.18 nM level.

N-substituted maleimides were synthesized from maleic anhydride and primary amines. 1,4-Dibromo-dibenzo[e,h]bicyclo-[2,2,2]octane-2,3-dicarboximide derivatives (4a-f) were prepared by the [4+2] cycloaddition reaction of dibromoanthracenes with the N-substituted maleimide derivatives. The carbonic anhydrase (CA, EC 4.2.1.1) inhibitory effects of the new derivatives were assayed against the human (h) isozymes hCA I, II, IX, and XII. All tested bicyclo dicarboximide derivatives exhibited excellent inhibitory effects in the nanomolar range, with Ki values in the range of 117.73-232.87 nM against hCA I and of 69.74-111.51 nM against hCA II, whereas they were low micromolar inhibitors against hCA IX and XII. A series of 9,10-dibromo-N-aryl-9,10-dihydro-9,10-[3,4]epipyrroloanthracene-12,14-diones (4a-f) were synthesized from N-substituted maleimide derivatives and 9,10-dibromoanthracene. Compounds 4a-f were assayed against human carbonic anhydrases (hCA) IX and XII, which are the two tumor-associated isozymes, and hCA I and II, which represent the most common off-targets for the development of selective anticancer CA inhibitors.

- MakaleAçık Erişim2016Scopus SJRQ2Acetylcholinesterase and carbonic anhydrase isoenzymes I and II inhibition profiles of taxifolin
Gocer, Hulya; Topal, Fevzi; Topal, Meryem; Küçük, Murat; Teke, Dilek; Gülçin, Ilhami; Alwasel, Saleh H.; Supuran, Claudiu T.
Taxifolin, also known as dihydroquercetin, is a flavonoid commonly found in plants. Carbonic anhydrase (CA, EC 4.2.1.1) plays an important role in many critical physiological events including carbon dioxide (CO2)/bicarbonate (HCO3-) respiration and pH regulation. There are 16 known CA isoforms in humans, of which human hCA isoenzymes I and II (hCA I and II) are ubiquitous cytosolic isoforms. In this study, the inhibition properties of taxifolin against the slow cytosolic isoenzyme hCA I, and the ubiquitous and dominant rapid cytosolic isoenzyme hCA II were studied. Taxifolin, as a naturally bioactive flavonoid, has a K-i of 29.2nM against hCA I, and 24.2nM against hCA II. For acetylcholinesterase enzyme (AChE) inhibition, K-i parameter of taxifolin was determined to be 16.7nM. These results clearly show that taxifolin inhibited both CA isoenzymes and AChE at the nM levels.
