In cell culture investigations, it is assumed that the number of cells is counted automatically without the need for forward planning, additional tools, or consumables. The automatic cell counting process is greatly facilitated and made functional by the segmentation of cell culture lines. Many studies with high-accuracy results have been developed for cell or tissue segmentation in images acquired from microscopic systems, but this success cannot be achieved for segmentation of cell culture line (especially in images acquired from inverted microscope). There are still restrictions in segmentation of cell culture line studies in inverted microscope systems. In this context, a new inverted microscope image data set is created and higher accuracy auto-encoder models are developed for segmentation of HaCat cell culture line. The analyzes of the developed auto-encoder models are evaluated using four different performance evaluation procedures which are Accuracy, Sensitivity, Specificity and Jaccard. In terms of the performance evaluation procedures used in this study, the achievements of the auto-encoder models are clearly satisfactory for the automated segmentation of the HaCat cell culture line.
AtıfScopus1İncele - MakaleAçık Erişim2021The Effect of Rosa pimpinellifolia Extract on the Proliferation of Human Tumor Cells
Demir, Elif Ayazoglu; Demir, Selim; Türkmen, Nihal; Turan, Ibrahim
The aim of this study was to evaluate the antioxidant properties of R. pimpinellifolia and its cytotoxic effect against human colon (WiDr), liver (HepG2) and lung (A549) cancer cells compared with a normal fibroblast (BJ) cell line. The antioxidant properties of extract was determined using spectrophotometric methods, while the cytotoxic effect of extract was evaluated using MTT assay. The total phenolic content, total flavonoid content and reducing power values of extract were calculated as 16.4 +/- 0.4 mg gallic acid equivalent, 5.2 +/- 0.2 mg quercetin equivalent and 34.3 +/- 2.4 mg trolox equivalent per g sample, respectively. Although the extract exhibits selective cytotoxic effect on three cancer cell lines in a dose-dependent manner compared to BJ cells, the highest selectivity index value (2.7-fold) was obtained for the WiDr cell line. In conclusion, this is the first study to demonstrate the cytotoxic effect of R. pimpinellifolia on cancer cells. To demonstrate the molecular mechanism of this cytotoxic effect and to carry out in vivo studies may be a guide for future clinical studies.

The aim of this study was to investigate the antioxidant properties of the root extract of Onosma armeniacum and its cytotoxic effect against human liver (HepG2), lung (A549) and colon (WiDr) cancer cell lines compared with a normal fibroblast cell line. Antioxidant properties of the extract was determined using the colorimetric methods. The cytotoxic effect of the extract were investigated using MTT assay. The total phenolic content and reducing power values of extract were 26.1 +/- 0.4 mg gallic acid equivalent and 52.6 +/- 1.1 mg trolox equivalent per g sample, respectively. The extract exhibited selective cytotoxic effect in all studied cancer cell lines, especially HepG2 (3.6-fold). The results of this study reveal that O. armeniacum, which has been found to have acceptable antioxidant and cytotoxic effects, should be examined with more comprehensive studies.

Ayazoglu Demir, Elif; Demir, Selim; Turan, Ibrahim
Ethyl pyruvate (EP) is a simple aliphatic ester derived from pyruvic acid which is an endogenous metabolite. Although various studies have investigated the antioxidant and anti-inflammatory properties of EP, there has been only limited research into the cytotoxic effect of EP on cancer cells. The aim of this study was to determine the cytotoxic effects of EP on cells representing common cancer types. EP was purchased commercially and intermediate stock solutions were prepared with phosphate buffer saline. The cytotoxic effect of EP on human melanoma (VMM917), cervix (HeLa), breast (MCF-7), lung (A549), liver (HepG2), colon (WiDr) cancer and normal fibroblast (BJ) cells was determined using the MTT assay. Cisplatin was used as a positive control in cytotoxicity experiments. The results showed that EP exhibits selective cytotoxic effect on VMM917 (10.1-fold) and HeLa (3.04-fold) cells compared to BJ cells. This study shows for the first time that EP has a highly selective cytotoxic effect, especially on melanoma and cervix cancer cells. The mechanism of this effect needs to be elucidated by more extensive studies.
