Objective: In this study we investigated in vitro antifungal activity of fluconazole, green tea (Camellia sinensis) and quince leaf (Cydonia oblonga) extracts in Candida strains isolated from blood cultures. Methods: Fifty Candida spp were included to study. Camellia sinensis and Cydonia oblonga leaves collected from Rize and Erzurum/Tortum regions were prepared as an extract. Then stock solutions of the extracts and fluconazole powder obtained from the manufacturer were prepared for use in the Broth Microdilution (BMD) method according to Clinical and Laboratory Standards Institute (CLSI) M27 A3 recommendation. Fluconazole minimum inhibitory concentration (MIC) values obtained from BMD method were evaluated according to CLSI M27 A4 break points. MIC values obtained for Camellia sinensis and Cydonia oblonga were interpreted according to fluconazole break points since there is no break points in the literature. Results: Of the 50 Candida spp. included in the study, 37 (74%) were C. albicans while 13 (26%) were non-albicans species. The most common isolated non-albicans species was C. parapsilosis (14%). For all tested Candida strains MIC 50 and MIC 90 values for fluconazole were found to be 0.125 mu g/mL- 0.25 mu g/mL while they were determined as 0.125 mu g/mL-64 mu g/mL and 0.125 mu g/mL-0.125 mu g/mL for green tea and quince leaves, respectively. Conclusion: According to the low MIC values, it is seen that green tea and quince leaf are in vitro effective against Candida species. However, further extensive in vitro and in vivo studies should be done on the possibility that green tea and quince leaf can be used alternatively in the treatment of invasive Candida infections.

Synthesis, antifungal activity and carbonic anhydrase inhibitory properties of Cu (II) Bis (3, 4 dimethoxybenzoate) bis (nicotinamide) dihydrate

Afsin Kaya, A.; Kaya, Elif Celenk; Sarikaya, S. Beyza Ozturk; Onaran, Abdurrahman

The new 3,4-dimethoxybenzoate complex of Cu(II) with nicotinamide was synthesized and characterized by elemental analysis, FT-IR spectroscopic study, UV-Vis spectrophotometry and mass spectrometry. The thermal behavior of the complex was studied by simultaneous TG, DTG and DTA methods in static air atmosphere. The infrared spectral characteristics of the complex is also discussed. In this complex, all ligands are coordinated to the metal ion as monodendate ligands. The final decomposition products were found to be the respective metal oxide. As it is well known, metal complexes of biologically important ligands are sometimes more effective than the free ligands. In this study the new compound was synthesized due to its functional groups such as veratric acid and nikotin amide, which have known biological characteristics and effective in terms of both enzyme and antifungal properties from recent studies. Therefore, antifulgal activity and enzyme inhibition properties of this complex have been examined.

In vitro antifungal evaluation of various plant extracts against walnut anthracnose (Gnomonia leptostyla (Fr.) Ces et de Not.)

    Kalkişim, Özgün

Antifungal activity of different parts (fruit and leaf) of Cornus mas L. (Cornecea), Morus nigra L. (Moraceae) and Prunus laurocerasus L. (Rosaceae) methanol extracts has been tested against walnut anthracnose. Agar dilution method has been used in antifungal activity studies. A standard fungicide (80% Maneb; Hektas Group) was used in order to compare the herbal extracts. The highest antifungal effect was observed in the leaf extracts of C. mas and M. nigra for which mycelium growth inhibition ratios observed were 67.95% and 60.04%, respectively. Different antifungal effect ratios have been determined for other plant extracts used. Also, the effect of Gnomonia leptostyla disease on walnut yield was examined for three different types of walnut trees. According to these results, it has been concluded that walnut anthracnose disease has an effect on the yield.

Antifungal activity of Trachystemon orientalis L. aqueous extracts against plant pathogens

    Onaran, Abdurrahman; Yilar, Melih

Trachystemon orientalis (L.) G. Don (Boraginaceae), recognized as Hodan, Kaldirik in Turkey, is a perennial medical plant. The plant materials of Kaldirik were collected from Saz village of Düzce province (Turkey) in 2010 to determine its antifungal effect. The aqueous extracts obtained from leaves, roots and flowers and were applied against five plant pathogen fungi which they are respectively Ascochyta rabiei (Pass) Labr., Fusarium oxysporum (Schlechtendahl:Fries) f. sp. melonis (Snyder & Hansen), Fusarium oxysporum (Schlechtendahl:Fries) f. sp. radicis-lycopersici (Jarvis & Shoemaker), Verticillium dahliae (Kleb.) and Rhizoctonia solani Kühn. The original concentration of extracts obtained for fungal studies was 10% as. Sterile potato dextrose agar (PDA) was cooled in a water bath at 40°C, and the extracts were mixed with the sterile 250 ml PDA to obtain final concentrations of 1%, 3%, 5%, 7% 10% and 20%. Then, the PDA was poured into 60-mm Petri plates (10 ml plate-1) and a agar disc (5 mm in diameter) of plant pathogens (from 7 day-old PDA cultures) were inoculated on medium. They were incubated at 25±2°C for 7 days in incubator. As a result of the study, the aqueous extract of flower showed maximum inhibitory effect against the mycelial growth of plant pathogens, it was respectively followed by leaf and root extracts. Flower extract completely inhibited mycelial growth of A. rabiei, V. dahliae and R. solani in all applied doses. According to the result of the study, the first time was identified of T. orientalis aqueous extracts, have high antifungal effect against plant pathogens.
