Background/aim: To determine the levels of adipokines (leptin, adiponectin, resistin, and visfatin) and the indices of insulin sensitivity/ resistance, and to examine the relationship among them in patients with metabolic syndrome (MetS).; Materials and methods: The study groups included 45 subjects with MetS (31 women/14 men), and 45 sex- and age-matched non-MetS healthy volunteers (31 women/14 men). The levels of adipokines were determined by enzyme-linked immunosorbent assay.; Results: The levels of leptin and visfatin were significantly higher in the MetS than in the non-MetS subjects (P < 0.01). There was no difference in adiponectin levels in subjects with and without MetS (P = 0.052). Similarly, resistin did not show any statistically significant difference. A statistically significant positive correlation of leptin with insulin levels was observed, while negative correlations of visfatin levels with age, and resistin levels with the ratio of adiponectin to leptin, were found in the MetS (P < 0.05). The combination of adipokines, insulin resistance-sensitivity parameters, and MetS criteria parameters gave more signifcant differences than a single parameter.; Conclusion: Since the parameters mentioned above might affect, interact with, and/or interfere with each other, the combinations of these parameters might give more reliable results to evaluate the insulin resistance/sensitivity in MetS patients.

Objective: Metabolic syndrome (MetS) is a complex disease characterized by insulin resistance, abdominal obesity, hyperglycemia, hypertension, hypertriglyceridemia and low HDL-cholesterol level. The aim of the study was to evaluate the sequence variations in the LPIN1 gene in MetS. This gene codes lipin-1 protein which functions as Mg-dependent phosphatidic phosphatase enzyme and transcriptional coactivator. Material and Methods: The study groups included 73 MetS (19 M/54 F) and 56 non-MetS (16 M/40 F). Sequence variation in exons 2, 4, 5 and 14 of the LPIN1 gene were investigated by DNA sequencing method. Results: c.696 G>C variant (p.S232S) in exon 5 was observed in only one women with MetS. But this variation is not important because of coding same amino acid. Conclusion: Any important sequence variant was not detected in exons 2, 4, 5 and 14 in the LPIN1 gene in MetS.

Of Brassicaceous plants, kale (Brassica oleraceae L. var. acephala DC) contains polyphenols, flavonoids, isoflavones and glucosinolates and so has antioxidant and anticarcinogenic properties. Antioxidants inhibit negative effects of free radicals and may, therefore, protect tissues against oxidative damage. Oxidation of lipoproteins is a key event in the development of atherosclerosis. In the current study, the levels of total phenolic and flavonoid contents and total antioxidant capacity of methanolic and aqueous extracts of kale leaves were determined. In addition, the susceptibility of isolated lipoproteins - very low density lipoprotein (VLDL) and low density lipoprotein (LDL) to the Cu 2+-induced oxidation with various concentrations of metanolic and aqueous extracts was evaluated as t-lag values. Although aqueous extract had higher total antioxidant capacity, methanolic extract had higher total phenolic and flavonoid content (P<0.05). On the other hand, both extracts inhibited lipid peroxidation in both isolated VLDL and LDL. Inhibitory effect of extracts or increasing t-lag values, mainly in methanolic extract was found to be related to increasing the concentration of extracts. It was concluded that because of high antioxidant capacity and phenolic content, kale showed a protective effect on the oxidation of lipoproteins. Therefore, it may be speculated that kale consumption may play an important protective role in the cardiovascular and other related diseases resulting from imbalance of oxidant and antioxidant status.

Background: Propolis is a natural, resinous hive product that has several pharmacological activities. Its composition varies depending on the vegetation, climate, season and environmental conditions of the area from where it was collected. Surface enhanced laser desorption ionization time of flight mass spectrometry (SELDI-TOF MS) is a proteomic approach which has been used in cancer proteomics studies. Prostate cancer is one of the most commonly diagnosed cancers in men. It has shown that nutritional supplements rich in polyphenolic compounds such as propolis play a significant role in prostate cancer chemoprevention. The aim of this study is to evaluate if protein expression profile in PC-3 prostate cancer cell lines could be differentiated when incubated with dimethyl sulfoxide and water extracts of Turkish propolis. Results: The antioxidant potentials of dimethyl sulfoxide and water extracts of propolis were found in correlation with the amount of total phenolic compounds of them. Dimethyl sulfoxide and water extracts of propolis of 20 μg/mL reduced the cell viability to 24.5% and 17.7%, respectively. Statistically significant discriminatory peaks between control PC-3 cells and dimethyl sulfoxide extract of propolis-treated PC-3 cells were found to be the proteomic features at m/z 5143, 8703, 12661, 20184 and 32794, detected by CM10 ProteinChip, and the peak at m/z 3772, detected by Q10 ProteinChip. Between control PC-3 cells and water extract of propolis-treated PC-3 cells, statistically significant discriminatory peaks were found to be the proteomic features at m/z 15846, 16052 and 24658, detected by CM10 ProteinChip and the peaks at m/z 10348, 10899 and 11603, detected by Q10 ProteinChip. Conclusions: It was concluded that dimethyl sulfoxide and water extracts of Turkish propolis may have anti-proliferative activity through differentiating protein expression profile in PC-3 prostate cancer cell lines along with their antioxidant capacity.

Objective: Peroxisome proliferator-activated receptor gamma (PPARγ), lipids, lipoproteins, and adipokines have recently been shown to be associated with psychiatric diseases. Our major aim is to investigate the contribution of the PPARγ gene polymorphism, adipokines, lipids, and lipoproteins to the development of major depression. Material and Methods: The frequency of Pro12Ala in exon 2 and C478T in exon 6 of the PPARγ gene, lipids and adipokines in major depression (n = 78) and control subjects (n= 64) were analyzed. Genotypes of PPARy gene polymorphisms were examined. Serum leptin, adiponectin, and resistin were studied by enzyme-linked immunosorbent assay (ELISA). Serum apo A1, apo B, and Lp(a) levels were determined by immunonephelometry. Serum total cholesterol, triglyceride, HDL-cholesterol and LDL-cholesterol levels were analyzed by enzymatic methods. Results: The genotypes of exon 2 and exon 6 distribution did not differ between the control subjects and patients with major depression. Frequencies of genotypes of Pro12Ala, and Ala12Ala in exon 2 in overweight and obese patients with major depression were higher than those of overweight and obese controls. C478T polymorphism was highest in overweight and obese patients with major depression. Pro12Ala and Ala12Ala genotypes in exon 2 of PPARγ gene in patients were found to be associated with triglyceride and HDL-cholesterol. There were significant differences regarding glucose, total cholesterol, LDL-cholesterol, apo B, Lp(a), adiponectin and resistin levels between patient and control subjects. Conclusion: PPARγ exon gene polymorphisms, alterations in lipid profile and adipokines may be associated with the development of major depression.
