Reliability and Validity of the Turkish Version of the Walsh Family Resilience Questionnaire

    Arslan, Kübra; Hatipoğlu, Ertuğrul

A multidimensional scale to measure family resilience, which focuses on facilitating positive adaptation and strengthening the family unit when affected by high-stressful events, does not exist in Turkish. The aim of this study was to adapt the Family Resilience Scale into Turkish and to conduct a validity and reliability study. The Family Resilience Scale was translated into Turkish and back-translated. Simple random sampling was used to create a representative sample of adults from 321 families selected in Turkey in 2022. The reliability of the scale as a whole and of the subscales was examined using Cronbach’s alpha. Test-retest reliability, the scale was administered to 30 individuals two weeks apart to determine reliability using the test-retest method. Cronbach alpha coefficients were greater than 0.70 for all scales. The test-retest reliability for the General Family Resilience Scale score was found to be.725. According to the results of the confirmatory factor analysis, chi-square χ2 = 2.598; RMSEA= 0.071; SRMR= 0.06; IFI= 0.90; CFI= 0.90; GFI= 0.89; TLI= 0.88. This study provides evidence that the Family Resilience Scale has good validity, reliability and test-retest reliability for Turkish families. Further testing, including construct validity and testing in different contexts, will strengthen the evidence. This scale will find application in resilience research, clinical settings, and facilitate the development and evaluation of intervention programmes to build resilience in Turkish families.

One-step kinetic analysis of Listeria innocua growth as a surrogate for Listeria monocytogenes on arugula leaves with background microbiota: model development and validation

    Ozturk, Samet

This study investigated the growth kinetics of L. monocytogenes and L. innocua on fresh arugula leaves under abusive conditions, considering the presence of background microbiota (BM) including (APC (Aerobic Plate Count), TPC (Total Psychrotrophic Count), and LAB (Lactic Acid Bacteria)). Additionally, the feasibility of using L. innocua as a surrogate for L. monocytogenes was evaluated for industrial practices. Predictive models were developed to assess the effect of temperature and time on the growth kinetics of both microorganisms. Experiments were conducted in triplicate to observe growth kinetics at temperatures from 5 to 35°C. The growth curves were analyzed using one-step analysis with the USDA IPMP-Global Fit software, employing the Huang full/no-lag phase growth as the primary models and the Huang sub-optimal (HSRM) and Ratkowsky sub-optimal square-root as the secondary models. An additional set of isothermal data, collected at 15°C and 20°C, was used to validate the models. Results showed that the minimum growth temperatures were 2.91±0.50°C for L. monocytogenes and 2.88±0.50°C for L. innocua, while 2.05±0.89, 1.93±0.98 and 3.55±1.97°C for APC, TPC and LAB, respectively. The specific growth rates of L. monocytogenes and L. innocua ranged from 0.01 to 0.93 h⁻¹. The root mean square error (RMSE) of model validation and development was less than 0.3 log CFU/g, indicating that the combination of the Huang growth model with HSRM could accurately predict the growth of L. monocytogenes under abusive conditions. Validated models can provide useful input to quantitative risk assessment tools to predict the growth of L. monocytogenes on arugula leaves in the presence of BM during distribution or storage. The findings of this study support the use of L. innocua with R2=0.961 as a suitable surrogate in industrial practices for fresh produce.

Single-Laboratory Validation for the Determination of Aflatoxin B1, B2, G1, and G2 in Foods Based on Immunoaffinity Column and Liquid Chromatography with Postcolumn Derivatization and Fluorescence Detection

Baltacı, Cemalettin; İlyasoğlu, Huri; Yüksel, Ferhat

This study presents a method validation procedure for the determination of aflatoxin B1, B2, G1, and G2 in hazelnut, hazelnut paste, walnut, peanut, pistachio, corn, and wheat. The method consisting of clean-up with immunoaffinity column, high performance liquid chromatography with postcolumn derivatization and fluorescence detection was validated in accordance with Commission Regulation 2004/882/EC. The selectivity, linearity, decision limit, detection capability, detection and quantification limits, precision, recovery, ruggedness, and measurement uncertainty of the method were determined. The limit of detection and limit of quantification values (μg/kg) were: aflatoxin B1, 0. 02, 0. 07; aflatoxin B2, 0. 01, 0. 02; aflatoxin G1, 0. 02, 0. 07; and aflatoxin G2, 0. 01, 0. 03. The relative standard deviation values for the repeatability and within-laboratory reproducibility were below 4 and 5 %, respectively. The recovery values of the spiked samples ranged from 80 to 105 %. These results complied with minimum performance criteria established by regulation 2006/401/EC. Therefore, the procedure can be implemented for the routine analysis of aflatoxins in the studied matrices.
