Covalent Immobilization of Glutathione-S-Transferase onto Magnetic Iron Nanoparticles via Epichlorohydrin Spacer Arm

Kemal Caglar, M.; Gulesci, Nuri; Bilgin, Ramazan; Yucebilgic, Guzide

This study examined the synthesis of Fe₃O₄ magnetic nanoparticles (MNPs) and the covalent immobilization of glutathione-S-transferase (GSTs) onto these nanoparticles using an epichlorohydrin (ECH) spacer arm, as well as the optimal reaction conditions for both free and immobilized enzymes and the reusability of the immobilized enzyme. The optimal pH values for free and immobilized enzymes were established as 7.0 and 6.0, respectively. It was found that the ideal temperature for both free and immobilized enzymes was 37 and 40°C, respectively. Under the optimum conditions, the values of V_max for glutathione (GSH) and 1-chloro-2,4-dinitrobenzene (CDNB) substrates of free GSTs enzyme were measured as 204.8 U/mg prot. and 194.3 U/mg prot., respectively. The immobilized GSTs enzyme's CDNB and GSH substrates had V_max values of 33.29 U/mg prot. and 33.08 U/mg prot., respectively. The free GSTs enzyme's CDNB and GSH substrates have K_m values of 0.201 mM and 0.1873 mM, respectively. GSH and CDNB have K_m values of 0.3042 mM and 0.2523 mM, respectively. At various temperatures, we contrasted the thermal stability of free and immobilized GSTs. The immobilized GSTs enzyme retained 50% of its activity after 20 reuses. After 30 days of storage at 25°C and 4°C, it was discovered that 18% and 32% of the free GSTs enzyme activity were preserved, and 30 days of storage at 25°C and 4°C resulted in 20% and 41% preserved enzyme activity for the immobilized enzyme, respectively.

Characterization of thermal, chemical, mechanical, and fatigue behavior of 3D printed ABS-based elastomeric blends: ABS/EVA and ABS/TPU

Kuleyin, Hamdi; Budak, Selahattin; Yasan, Ömer Barışkan; Gümrük, Recep

The polymer blending method is widely used to obtain balanced mechanical properties and structural performance in many engineering applications. This study investigates the influence of Ethylene-Vinyl Acetate (EVA), and Thermoplastic Polyurethane (TPU) composition ratios on the thermal, chemical, viscoelastic, mechanical, and fatigue life behavior of Acrylonitrile-Butadiene-Styrene (ABS) based elastomeric blends. For this purpose, structural and mechanical properties of the blend material depending on their composition ratio were carried out through a series of characterization analyses, and tensile and fatigue tests. The findings indicated that ABS/EVA and ABS/TPU blends formed an immiscible structure. In addition, it was determined that the storage modulus of the blend with 10 % TPU improved because of the presence of new absorption peaks in the structure. The mechanical properties such as elastic modulus, yield strength, and ultimate tensile strength decreased concerning neat ABS with an increase in the EVA and TPU composition ratio for blend materials. However, the elongation to break was achieved approximately 6 times increasing with an increase in the TPU ratio for ABS/TPU blends. It was determined that the fatigue strength value in ABS/EVA and ABS/TPU blends decreased with the decrease in ABS composition ratio in the blend system. However, it was found that fatigue life improved by 25 % compared to ABS at low-stress amplitude for 10 % TPU additive. Finally, it can be revealed that the ABS-based elastomeric blends can be used to obtain desired properties according to their composition ratios for different applications.

Preparation and characterization of bacterial cellulose produced from fruit and vegetable peels by Komagataeibacter hansenii GA2016

    Güzel, Melih; Akpınar, Özlem

This study focused on the investigation of bacterial cellulose production potency of some fruit and vegetable peels (cucumber, melon, kiwifruit, tomato, apple, quince and pomegranate) with Komagataeibacter hansenii GA2016. Fruit and vegetable peels were hydrolyzed, used for bacterial cellulose (BC) production and their chemical, physical, thermal and structural features were compared to BC from Hestrin–Schramm medium (HSBC) and plant cellulose (CP). Except for pomegranate peel hydrolysate, all the fruit and vegetable peel hydrolysates supplied to K. hansenii GA2016 supported the BC production. Among the fruit and vegetable peel hydrolysates, the highest BC production was observed in kiwifruit peel hydrolysate (11.53%), while the lowest production was observed in apple peel hydrolysate (1.54%). Water-holding capacities of the BCs were ranged from 627.50% to 928.79% and higher than HSBC (609.30%), average fiber diameters were ranged from 47.64 nm to 61.11 nm and thinner than HSBC (74.29) and CP (10,420 nm), crystallinities were ranged from 80.27% to 92.96%, thermal capacities BCs were higher than HSBC and CP. For the BC productions, utilization of the fruit and vegetable peels as the sole nutrient source could reduce the production costs and among the polysaccharides, increase the use of BC in industry.

Physicochemical characteristics of a novel activated carbon produced from tea industry waste

Gundogdu, Ali; Duran, Celal; Senturk, H. Basri; Soylak, Mustafa; Imamoglu, Mustafa; Onal, Yunus

Tonnes of waste are generated during tea production in Turkey, one of the black tea producing countries in the world. These wastes are not used for any purposes throughout industrial sector. The objective of this study is to bring in economic value to waste materials by producing activated carbons from wastes of a tea plant and to characterize their physicochemical properties. Three types of activated carbons were produced by chemical activation using zinc chloride from tea industry wastes (TIW). Mesoporous structure and surface area rise as the ratio of ZnCl2/TIW increases. When the ratios of ZnCl2/TIW were selected as 1/2:1, 1:1, and 2:1, mesoporous activated carbons are found in the percentages of 10.9%, 39.9% and 77.7% with BET surface areas of 706, 1066 and 1141 m(2) g(-1), respectively. The resultant activated carbons were characterized by analyses of proximate-ultimate, FT-IR, SEM, TGA, methylene blue-iodine numbers, and surface functional groups. (C) 2013 Elsevier B.V. Kalkişim, Özgün; Balik, Hüseyin Irfan

This study was carried out between Çarşi{dotless}başi{dotless}/Trabzon and Piraziz/Giresun region which is assumed to be the best ecology for Tombul hazelnut cultivation. Visual pre-selection was performed in the region and the clones that may exhibit different characteristics were detected and marked. Later fruit samples were taken from those clones at the time of harvest and the superior clones were determined as a result of the examinations performed on the fruits. The efficiency and fruit features were analyzed for selected Tombul hazelnut clones. The study showed that the differences between efficiency, yield, kernel weight, fruit weight, shell thickness and bundle fruit number were significant (P<0.05) among Tombul hazelnut clones. On the other hand, differences of navel cavity, splitting fruit ratio, whitening and healthy kernel ratio among clones were not significant. The highest and lowest values of efficiency were 13.45 and 168.47 g/cm2, respectively, for clones 61V11 and 28M16, yield was 45.11-54.07%, respectively, for clones 61V11 and 28M17, kernel weight 0.89-1.19 g, respectively, for clones 61V11 and 28G16, fruit weight 1.67-2.19 g, respectively, for clones 61B08 and 28ES04, shell thickness 0.89-1.10 mm, respectively, for clones 28G08 and 28ES04 and bundle fruit number 2.15-4.38, respectively, for clones 61B08 and 28EY12. The clones 28ES01 (53.86), 61Ç07 (53,88%), 61V25 (53.90%) and 28M17 (54.07%) had more superior characteristics in terms of yield compared to others.
