Primula vulgaris belongs to the genus Primula and this genus is frequently used in traditional medicine. Numerous studies reported the antioxidant and cytotoxic effects of various extracts of different Primula species but, there were limited studies of the antioxidant and cytotoxic effect of P. vulgaris. The aim of this study was to determine the antioxidant properties and cytotoxic effects of dimethyl sulfoxide extract of leaves of P. vulgaris. The total polyphenolic content (TPC), total flavonoid content (TFC) and reducing antioxidant power of extract were evaluated using spectrophotometric procedures. The cytotoxic effects of extract were revealed on human lung (A549), liver (HepG2), breast (MCF-7), prostate (PC-3), and colon (WiDr) cancer cell lines and normal fibroblast cells using thiazolyl blue tetrazolium bromide (MTT) assay. TPC and FRAP values were 1890.1 +/- 11.2 mg gallic acid equivalents and 3233.1 +/- 176.2 mg trolox equivalents per 100 g sample, respectively. The extract showed selective cytotoxic effects on all studied cancer cell lines compared to normal fibroblast cells and IC50 values ranged from 133.3-253.8 mu g/mL. This is the first study to reveal the antioxidant properties of P. vulgaris leaf extract and its cytotoxic effect on cancer cell lines. Further studies are now needed to identify the antioxidant and cytotoxic molecules in the extract and their mechanisms.

Background: Morus nigra L. belongs to the family Moraceae and is frequently used in traditional medicine. Numerous studies have investigated the antiproliferative effects of various extracts of different Morus species, but studies involving the in vitro cytotoxic effect of M. nigra extract are very limited. The purpose of this study was to evaluate the phenolic composition and antioxidant activity of dimethyl sulfoxide extract of M. nigra (DEM) and to investigate, for the first time, the probable cytotoxic effect in human prostate adenocarcinoma (PC-3) cells together with the mechanism involved. Methods: Total polyphenolic contents (TPC), ferric reducing antioxidant power (FRAP) and phenolic compounds of DEM were evaluated using spectrophotometric procedures and HPLC. The cytotoxic effect of DEM on PC-3 cells was revealed using the MTT assay. Mechanisms involved in the cytotoxic effect of DEM on PC-3 cells were then investigated in terms of apoptosis, mitochondrial membrane potential and cell cycle using flow cytometry, while caspase activity was investigated using luminometric analysis. Results: TPC and FRAP values were 20.7 ± 0.3 mg gallic acid equivalents and 48.8 ± 1.6 mg trolox equivalents per g sample, respectively. Ascorbic acid and chlorogenic acid were the major phenolic compounds detected at HPLC analysis. DEM arrested the cell cycle of PC-3 cells at the G1 phase, induced apoptosis via increased caspase activity and reduced mitochondrial membrane potential. Conclusions: Our results indicate that M. nigra may be a novel candidate for the development of new natural product based therapeutic agents against prostate cancer.

Objective: Cancer is one of the most deadly types of disease and evasion from apoptosis and unstoppable cell proliferation are accepted as its distinctive features. Many studies have evaluated the cytotoxic effect of different Morus species but, there is no study about cytotoxic effect of Morus rubra. In this study we aimed to evaluate phenolic composition, antioxidant properties and cytotoxic effect of acidified dimethyl sulfoxide extract of M. rubra (AMRE). Method: Antioxidant properties, phenolic composition and cytotoxic effect of AMRE were determined using spectrophotometric methods, HPLC, and MTT assay, respectively. Then, mechanisms of cytotoxic effect of AMRE on human prostate (PC-3) and lung (A549) cancer cells were examined in regard to cell cycle, apoptosis and mitochondrial membrane potential using flow cytometric methods. Results: Total phenolic content and reducing power values were 11.9 mg gallic acid equivalents and 42.9 mg trolox equivalents per g sample, respectively. Ascorbic and gallic acid were detected in AMRE as major antioxidant compounds. We determined that AMRE increased cell cycle arrest at G1 phase and exhibited apoptotic features via decreasing mitochondrial membrane potential in both prostate and lung cancer cells. Conclusion: These findings demonstrate that M. rubra extract can affect the behavior of human prostate and lung cancer cells in vitro conditions, and this effect now needs to be investigated in vivo.

Demir, Selim; Turan, Ibrahim; Aliyazicioglu, Yuksel; Kilinc, Kagan; Yaman, Serap Ozer; Ayazoglu Demir, Elif; Arslan, Ayse; Mentese, Ahmet; Deger, Orhan

Many studies have reported cytotoxic effects of different Morus species, but there have been only limited studies on the cytotoxic effect of Morus rubra. The aims of this study were to evaluate the cytotoxic effect of dimethyl sulfoxide extract of M. rubra and to investigate, for the first time, its probable cytotoxic activity in human colon cancer (WiDr) cells, together with the mechanism involved. The cytotoxic activity of extract was determined using MTT assay. The mechanism involved in the cytotoxic effect of extract was then evaluated in terms of apoptosis, and the cell cycle using flow cytometry, mitochondrial membrane potential (MMP) was investigated using the fluorometric method, and expression levels of telomerase and C/EBP homologous protein (CHOP) were investigated using reverse-transcription PCR (RT-PCR). M. rubra extract exhibited moderate selective cytotoxicity on colon cancer cells compared with fibroblast cells. Extract induced cell cycle arrest at the G1 phase and apoptosis via reduced MMP in WiDr cells. Additionally, M. rubra extract significantly repressed telomerase and induced CHOP expressions in WiDr cells. Our results demonstrate that targeting telomerase and endoplasmic reticulum stress represents a promising strategy in colon cancer therapy, and M. rubra may have considerable potential for development as a novel natural product-based anticancer agent.

Cancer is the second leading cause of death and gastric cancer is the fourth most common cancer type worldwide. Investigation of autoantibodies in cancer patients has been a popular research area in recent years. The aim of the current study was to investigate carbonic anhydrase I and II (CA I and II) autoantibodies in the plasma of subjects with gastric cancer based on the information and considerations of autoimmune relation of gastric cancer. Anti-CA I and II antibody levels were investigated by ELISA in plasma samples of fifty two patients with gastric cancer and thirty five healthy peers. Anti-CA I and II antibody titers of the gastric cancer group were significantly higher compared with the control group (p = 0.004, p = 0.0001, respectively). Plasma anti-CA I levels of the metastatic group were lower than the non-metastatic group and this difference was found statistically significant (p < 0.05), but there was no statistical difference between plasma anti-CA II levels of the groups. CA I and II autoantibody titers in patients with gastric cancer were found higher compared to healthy subjects and the results suggest that these autoantibodies may be involved in the pathogenesis of gastric cancer.

Mn(II), Co(II), Ni(II), Cu(II), and Zn(II) transition metal complexes of 2-hydroxy-5-[(E)-(4-phenyl) diazenyl] benzaldehyde oxime and 2-hydroxy-5-[(E)-(4-nitrophenyl) diazenyl] benzaldehyde oxime ligands were synthesized and characterized through NMR, IR, ESI mass, and UV analysis. DNA binding abilities of the complexes were revealed using a UV-Vis spectrophotometer with the absorption titration and competitive binding techniques. Hydrolytic and oxidative DNA cleavage activities under different conditions were also proved. Topoisomerase I inhibition efficiencies and in vitro free radical scavenging activities of all complexes were examined. Finally, the selective cytotoxic potentials of all complexes were evaluated in human colon cancer, normal colon, and fibroblast cell lines using the water-soluble tetrazolium salt (WST-1) assay. The complexes had the ability to intercalate into stacked base pairs of DNA and topoisomerase I activity was reasonably inhibited in their presence in 0.4 mM concentrations. The abilities for scavenging of DPPH and hydroxyl radicals were found to be higher than those of known standard antioxidants (ascorbic acid, butylated hydroxyanisole, and mannitol). The results obtained from the cytotoxicity experiments are especially promising for further research, which must be carried out for the evaluation of the studied complexes as anticancer drugs.

The objective of study was conducted on to compare immunoglobulin (Ig) concentrations in calves raised under organic and conventional conditions. 30 newborn Holstein Friesian calves (15 rose under organic and 15 under conventional conditions) in two dairy cattle farms. Blood samples were taken from 30 newborn calves at birth (before taking colostrum) and on 14th day to compare immunoglobulin (Ig) concentrations in calves raised under organic and conventional conditions. Furthermore, blood and colostrum samples were also taken from the mothers included into the study. IgG and IgM concentrations in colostrum and blood serums were determined using commercial ELISA kits. IgG concentrations at birth (0.79±0.34, 1.64±0.95 mg/mL) and on the 14th day (46.5±21.5, 70.5±27.8 mg/mL) in calves raised under organic conditions were lower than those raised under conventional conditions (p<0.01). Calves raised under organic conditions also had lower IgM concentrations than calves raised under conventional conditions at birth (0.23±0.03 and 0.28±0.11 mg/mL, respectively) and on the 14th day (2.76±0.56 and 4,30±1.03 mg/mL, respectively) (p<0.05, p<0.01). IgG concentrations in the blood serum of cows raised under conventional conditions were higher than those raised under organic conditions (p<0.01), while no difference was found with respect to IgM concentrations (p>0.05). No difference was found in the IgG and IgM concentrations of colostrum obtained from cows raised under organic and conventional conditions. In both the husbandry systems, positive and significant (p<0.01) correlations were found between the Ig concentrations of the mothers and offsprings. Consequently, it can be suggested that higher Ig concentrations in cattle raised under conventional conditions might be due to poorer environmental conditions.

The aim of this study was to evaluate the oxidant-antioxidant balance in patients with abdominal aortic aneurysms (AAA). Forty-two consecutive patients with AAA and 46 control subjects were included. Total oxidant status (TOS) and total antioxidant status (TAS) levels were measured and the oxidative stress index (OSI) value determined. Serum TOS and OSI values in patients with AAA were higher than those in the controls (p<0.001, p<0.001, respectively). There was a positive correlation between abdominal aortic diameters, serum TOS levels (r=0.592, p<0.001) and OSI values (r=0.598, p<0.001). A cut-off value of 17.68 μmol H2O2 equivalent/L for TOS was associated with 86% sensitivity and 83% specificity and a cut-off value of 1.77 for OSI was associated with 86% sensitivity and 81% specificity for predicting AAA. Systemic oxidative imbalance develops in patients with AAA, particularly as a result of an increase in TOS.

Cancer is a heterogeneous disease, two of whose characteristic features are uncontrollable cell proliferation and insufficient apoptosis. Various studies have investigated the antiproliferative effects of propolis, a natural bee product, from different countries, and its cytotoxic effects have been attributed to its polyphenol contents. The purpose of this study was to show the cytotoxic effects, and possible mechanisms involved, of ethanolic extract of Turkish propolis (EEP) on the human lung cancer (A549) cell line. Cytotoxic activity of EEP on A549 cells was revealed using the MTT assay. Mechanisms involved in the cytotoxic action of EEP on A549 cells were then investigated in terms of apoptosis, mitochondrial membrane potential and cell cycle using flow cytometry, endoplasmic reticulum stress using RT-PCR, and caspase activity using luminometric analysis. EEP exhibited selective toxicity against A549 cells compared to normal fibroblast cells. We determined that EEP arrested the cell cycle of A549 cells at the G1 phase, induced endoplasmic reticulum stress, caspase activity, and apoptosis and reduced mitochondrial membrane potential. These results indicate that Turkish propolis is capable of reducing cancer cell proliferation and may have a promising role to play in the development of new anticancer drugs in the future.

Turan, İbrahim; Değer, Orhan; Aliyazicioğlu, Yüksel; Demir, Selim; Kilinç, Kağan; Sümer, Ayşegül

Background/aim: Propolis is a bee product with antioxidative, antimutagenic, and other beneficial properties, and it is used as a natural drug. It is rich in polyphenolic compounds. Its composition varies depending on the particular geographical region. Oxidative stress is caused by an imbalanced free radical production and antioxidant system. The effects of flavonoids on the expression of DNA repair enzymes have been examined previously; however, no study has investigated the effects of propolis. This study investigated the effects of ethanolic extracts of Turkish propolis (EEP) on the expression of DNA repair enzymes. Materials and methods: The effects of EEP and tertiary-butyl-hydroperoxide (t-BHP) on cell viability were determined using MTT. DNA damage was determined using comet assay. mRNA expression of target enzymes was detected using RT-PCR. Results: According to the cytotoxicity analysis, after a recovery time of 4 h, appropriate damage agent t-BHP and optimum EEP concentrations were 300 µM and 200 µg/mL, respectively. 8-Oxoguanine-glycosylase (hOGG-1) and endonuclease-VIII-like-1 (NEIL-1) expressions increased in the positive control group (t-BHP alone) and the study group (t-BHP+EEP). Maximum increase in NEIL-1 expression was at hour 12 in the positive control group and at hour 8 in the study group. Conclusion: EEP can be considered as a potential source of functional food and pharmaceutical agents.

Results: The mean serum anti-CA I antibody levels were significantly higher in women with PCOS and anti-CA II antibody levels were not significantly different in women with PCOS compared with control subjects. For serum anti-CA I antibody, the absorbance higher than 0.484 (mean + 3SD of control subjects) was taken as positive, Anti-CA I antibody was detected in 13 of 50 patients with PCOS (26%). Considering serum anti-CA II antibody, the absorbance higher than 0.654 (mean + 3SD of control subjects) was taken as positive. Anti--CA II antibody was detected in 2 of 50 patients with PCOS (4%). All patients with positive anti-CA II antibody also had positive anti-CA I antibody. Autoantibodies specifically reactive to CA I were found to be present at a higher frequency than CA II in the serum of subjects with PCOS in the present study. Conclusion: The results may suggest that autoimmune responses against CA I may be involved in the pathogenesis of PCOS.
