The common bean (Phaseolus vulgaris L.) is the most consumed food legume in the world and is a major source of dietary protein, carbohydrates, and valuable micronutrients, especially in developing countries. Diversity Arrays Technology (DArTseq), based on genome reduction with restriction enzymes, provides a rapid, high-throughput, and cost-effective tool capable of generating thousands of genotyped single nucleotide polymorphisms (SNPs) for a genome-wide analysis of genetic diversity. In this study, we aimed to characterize common bean accessions using SNPs detected by a DArTseq approach. A total of 43,018 SNPs were identified from 173 common bean accessions, including Andean and Mesoamerican genotypes. After filtering raw and redundant data, a total of 16,366 SNPs were considered for further analyses. According to population structure analysis, the genotypes were roughly divided into 2 gene pools of Andean and Mesoamerican types. Pairwise fixation index (Fst) values were calculated to resolve the differentiation between populations. This study demonstrated that discovering SNPs from the whole genome is a potential resource for identifying naturally diverse accessions and also the information could be used in breeding programs to develop new common bean varieties.

Genetic diversity and population structure of common bean (Phaseolus vulgaris L.) accessions through retrotransposon-based interprimer binding sites (iPBSs) markers

Nemli, Seda; Kianoosh, Tala; Tanyolaç, Muhammed Bahattin

The common bean (Phaseolus vulgaris L.) is an important crop throughout the world and is highly recommended for many developing countries due to its nutritional components. A retrotransposon-based DNA marker system, interprimer binding sites (iPBSs), developed from reverse transcriptase primer binding sites, was used to determine genetic diversity in common bean accessions. The genetic polymorphism and effectiveness of the marker system were evaluated. A total of 180 polymorphic bands were detected using 47 iPBS primers among 67 common bean accessions, with an average of 4 polymorphic fragments per primer. The genetic similarity between accessions was calculated using the software NTSYS-pc and ranged from 0.09 to 0.99. The average polymorphism information content value for the iPBS markers was 0.73. A model-based clustering method classified the common bean accessions into 4 populations using STRUCTURE software. The results indicated that the iPBS marker method can successfully determine the genetic diversity level in common bean accessions.

Hybrid performance and heterosis in F1 offspring of triticale (× Triticosecale Wittm.)

Yildirim, Mustafa; Gezginç, Hasan; Paksoy, Ali Haydar

In this study, the combined ability effects and heterosis were determined for number of days to anthesis (AT), plant height (PH), fertile tillers plant-1 (FT), spike length (SL), spikelets spike-1 (SS) and grains spike-1 (GS), 1000-kernel weight (TW), and grain yield plant-1 (GY) of 4 triticale genotypes [Karma (G1), Presto (G2), Ayşehanım (G3), and Mehmetbey (G4)] and their 6 F1 offspring. The mean squares of general combining ability (GCA) and specific combining ability (SCA) were statistically significant for all traits. According to the GCA/SCA, additive gene effects were found for all traits. According to the desired values for traits on the basis of GCA and SCA, parents G1 were good general combiners for 3 traits, G3 for 6, and G4 for 6. Crosses G1 × G2 were good specific combiners for 3 traits, G1 × G3 for 3, G1 × G4 for 3, G2 × G3 for 5, G2 × G4 for 6, and G3 × G4 for 4. Heterosis and heterobeltiosis were -16.78% and -25.71% for AT, -1.66% and -11.02% for PH, 34.25% and 9.87% for FT, 14.62% and 11.44% for SL, 8.09% and 3.87% for SS, 4.57% and 0.91% for GS, 9.67% and 6.13% for TW, and 30.36% and 9.11% for GY, respectively. The results revealed that G2 × G3 and G2 × G4 combinations could be recommended for improved yield of triticale.

Many ecological regions of Turkey are suitable for the production of durum wheat, and therefore the yield and quality of this wheat should be improved. In this study, stability properties associated with the pasta quality of 25 durum wheat (Triticum durum Desf.) genotypes grown in 3 dif erent locations (Tokat-Kazova, Diyarbaki{dotless}r, and Sivas-Ulaş) for 2 years (2005-2006, 2006-2007) were investigated. Durum wheat genotypes were composed of 12 registered varieties and 13 advanced experimental lines. Field trials were conducted in a randomized complete block design with 3 replications. Protein content, gluten index, sedimentation and specif c sedimentation volumes, yellow pigment content, and lipoxygenase activity of the durum wheats were measured as major quality characteristics. T e regression coef cient (bi) and mean square of deviation from regression (S2 d) were employed as the stability parameters. Genotypes, growing environments, and their interactions were found to be statistically signif cant (P < 0.01) for all investigated quality characteristics. No cultivars were determined to be simultaneously stable for all of the quality characteristics, yet some of the genotypes prevailed for certain quality characteristics. T e genotypes with the same origin took part in the same group as judged by the cluster analysis. Of the advanced experimental durum wheat lines, Line - 1, Line - 7, Line - 20, and Gdem - 12, which displayed better quality characteristics than the overall means, can be used as breeding materials. Of the registered varieties, Aydin - 93, Çeşit - 1252, and Gidara were determined to be stable by both parameters in certain quality characteristics.
